Zinc tetraaminophthalocyanine-Fe3O4 nanoparticle composites were prepared by organic-inorganic complex technology and characterized. It has been proved that the ZnTAPc dispersed randomly onto the surface of Fe3O4 nanoparticles to form molecular dispersion layer and there was a relatively strong bond between central zinc cation and oxygen. The nanoparticle composite took the shape of roundish spheres with the mean diameter of about 15 nm. Active amino groups of magnetic carriers could be used to bind laccase via glutaraldehyde. The optimal pH for the activity of the immobilized laccases and free laccase were the same at pH 3.0 and the optimal temperature for laccase immobilization on ZnTAPc-Fe3O4 nanoparticle composite was 45 degrees. The immobilization yields and K(m) value of the laccase immobilized on ZnTAPc-Fe3O4 nanoparticle composite were 25% and 20.1 microM, respectively. This kind of immobilized laccase has good thermal, storage and operation stability, and could be used as the sensing biocomponent for the fiber optic biosensor based on enzyme catalysis.  相似文献

缺氧诱导因子1a (hypoxia inducible factor-1 a, HIF-1 a)是细胞在缺氧等条件下稳定表达的具有转录活性的蛋白,通过与多种靶基因调控区的缺氧反应元件(hypoxia response element, HRE)结合, 调控靶基因表达, 使机体对缺氧、缺血等病理生理过程产生适应性反应。为从整体动物水平研究HIF-1 a的作用, 需要建立HIF-1 a相关遗传修饰小鼠。分别针对HIF-1 a mRNA序列的两个靶位点合成两对互补的寡核苷酸链, 构建可诱导的RNA干扰真核表达载体HIF-AB和HIF-CD。分别将CRE重组酶真核表达载体CRE-ERT2与HIF-AB或HIF-CD转染入RAW264.7细胞, 筛选得到稳定表达CRE-ERT2与HIF-AB, 或CRE-ERT2与HIF-CD的稳定细胞系。在用4-HT诱导去除上述细胞系中HIF-AB或HIF-CD所含的Neo基因后, 用CoCl2诱导HIF-1 a表达, 采用半定量RT-PCR检测HIF-AB或HIF-CD对HIF-1 a 基因表达的影响。结果发现干扰载体(HIF-AB和HIF-CD)对HIF-1 a mRNA序列的沉默效果分别为85%和72%。选择干扰效率较高的表达载体HIF-AB经显微注射获得HIF-1 a基因敲低小鼠模型, 经PCR以及测序验证获得2个转基因阳性小鼠(Founders, G0代)。G0代雄鼠与FVB/N雌鼠交配后获得2只F1代(first filial generation)转基因阳性小鼠, 经与EIIA-Cre转基因小鼠交配, 得到EIIA-Cre; HIFRNAiflox/+小鼠, RT-PCR结果显示, EIIA-Cre; HIFRNAiflox/+小鼠肝、肺、肾等组织的HIF-1 a mRNA水平明显降低, 分别约为正常对照的44%、38.2%和23.5%。该小鼠模型的建立为进一步研究HIF-1 a的功能及作用机制提供了新的手段。  相似文献

The elevated concentration of atmospheric CO2 may result in a decline of leaf nutritional quality (especially N) and an increase in some kinds of defensive secondary components (such as phenolics). The changes in the phytochemistry of trees, combined with the effect of elevated CO2 per se, have a potential negative influence on insect herbivores. Here, we review the effect of elevated CO2 on the performance of leaffeeding forest insects at individual-level and community-level. The elevated CO2 per se have little influence on the metabolism of insects. Over half of the tree-insect experimental systems show that the performance of individual insect become poorer under high-CO2 grown trees; but the others show that the insects have just little or no response to the treatments. The direction and magnitude of the changes in the performance of insects could be mediated by various factors. The effects of treatment are strongly species-dependent. The magnitude of changes in the phytochemistry, the sensitivity and adaptive capacity of insects to the poorer leaf quality, the differences in plant growth conditions and experimental methods, and the mediated effects of other environmental factors (such as soil nutrient availability, light, temperature, O3) were all closely related to the final performance of insects. However, the larvae’s consumption usually increased under enriched CO2 treatment, which was widely thought to be a compensatory response to poorer plant quality. The experiments on forest community-level found identically a reduction in herbivory, which was contrary to the results from small-scale experiments. Farnesyl pyrophosphate (FPP) is a common substrate for a variety of prenyltransferases for synthesizing isoprenoid compounds. In this study, (2E,6E)-8-O-(N-methyl-2-aminobenzoyl)-3,7-dimethyl-2,6-octandien-1-pyrophosphate (MANT-O-GPP), a fluorescent analog of FPP, was synthesized and demonstrated as a satisfactory substrate for Escherichia coli undecaprenyl pyrophosphate synthase (UPPS) with a Km of 1.5 μM and a kcat of 1.2 s−1 based on [14C]IPP consumption. Interesting, we found that its emission fluorescence intensity at 420 nm increased remarkably during chain elongation, thereby useful for real-time monitoring kinetics of UPPS to yield a Km of 1.1 μM and a kcat of 1.0 s−1, consistent with those measured using radiolabeled substrate. Using this assay, the IC50 of a known UPPS inhibitor farnesyl thiopyrophosphate (FsPP) was confirmed. Our studies provide a convenient and environmentally friendly alternative for kinetics and inhibition studies on UPPS drug target.  相似文献

The effects of Astragalus polysaccharides (APS) on the immune response in pigs immunized with foot-and-mouth disease virus (FMDV) vaccine were investigated. Fifteen pigs were randomly divided into five groups. Four groups were vaccinated with a FMDV inactivated vaccine. Pigs in three experimental groups were administered varying doses of APS (APS1, 5 mg/kg; APS2, 10 mg/kg; APS3, 20 mg/kg). The influence of APS on the number of CD3+CD4CD8+ cytotoxic T cells, CD3+CD4+CD8+ T helper memory cells, and CD3CD4CD8+ natural killer cells among peripheral blood lymphocytes (PBL) in the three APS groups were significant compared to the vaccine group. In vitro stimulation of PBL by Con A and LPS in APS groups induced a stronger proliferative response at 2 and 6 weeks post-inoculation (PI). APS markedly increased the titer of FMDV-specific antibody in a dose-dependent manner, and up-regulated mRNA expression of IFN-γ and IL-6. APS could potentially be used as an immunomodulator for a FMDV vaccine and provide better protection against FMDV.  相似文献

The nonhuman primates most commonly used in medical research are from the genus Macaca. To better understand the genetic differences between these animal models, we present high-quality draft genome sequences from two macaque species, the cynomolgus/crab-eating macaque and the Chinese rhesus macaque. Comparison with the previously sequenced Indian rhesus macaque reveals that all three macaques maintain abundant genetic heterogeneity, including millions of single-nucleotide substitutions and many insertions, deletions and gross chromosomal rearrangements. By assessing genetic regions with reduced variability, we identify genes in each macaque species that may have experienced positive selection. Genetic divergence patterns suggest that the cynomolgus macaque genome has been shaped by introgression after hybridization with the Chinese rhesus macaque. Macaque genes display a high degree of sequence similarity with human disease gene orthologs and drug targets. However, we identify several putatively dysfunctional genetic differences between the three macaque species, which may explain functional differences between them previously observed in clinical studies.  相似文献
