During summer 2001, blue mussels Mytilus edulis with abnormal shell growth were collected near Krager?, southern Norway. The mussels had green spots in their mantle tissues, mainly posteriorly and ventrally, and in the adductor muscle. Mussels from 4 sites had a prevalence of green spots varying from 2 to 71% that correlated well with shell deformities. Histological examination revealed the presence of round or ovoid algae, 0.9 to 1.5 x 1.2 to 2.4 microm, free within haemocytes and in the lesions, characterised by an inflammatory response and the presence of cellular debris. The alga contain a relatively large nucleus, 1 chloroplast and 1 mitochondrion. Size and morphology suggest that the alga might be a picoeucaryot green alga. Infection of mussel tissues appears to start in the posterior mantle edge, near the siphons, and spread anterior-ventrally in the mantle connective and storage tissues-occasionally spots were also found in the gonad follicles. Large infected areas were also observed in sinuses within the adductor muscle. Only mussels that were 3 yr old or more were infected. Deformations apparently resulted from years of continuous shell formation by a contracted, partly deformed mantle. Most deformed mussels had eroded shells, allowing some light penetration through the exposed, thin nacre. Young, thin-shelled mussels were not infected. The present work suggests that the alga has, at least partially, a parasitic relationship with the mussels, and is associated with pathological alterations in mussel tissues.  相似文献

The filamentous fungus Fusarium oxysporum is known for its ability to produce ethanol by simultaneous saccharification and fermentation (SSF) of cellulose. However, the conversion rate is low and significant amounts of acetic acid are produced as a by-product. In this study, the growth characteristics of F. oxysporum were evaluated in a minimal medium using glucose as the sole carbon source in aerobic, anaerobic and oxygen-limited batch cultivations. Under aerobic conditions the maximum specific growth rate was found to be 0.043 h(-1), and the highest ethanol yield (1.66 mol/mol) was found under anaerobic conditions. During the different phases of the cultivations, the intracellular profiles were determined under aerobic and anaerobic conditions. The profiles of the phosphorylated intermediates indicated that there was a high glycolytic flux at anaerobic growth conditions, characterized by high efflux of glyceraldehyde-3-phosphate (G3P) and fructose-6-phosphate (F6P) from the pentose phosphate pathway (PPP) to the Embden-Meyerhof-Parnas (EMP) pathway, resulting in the highest ethanol production under these conditions. The amino acid profile clearly suggests that the TCA cycle was primarily active under aerobic cultivation. On the other hand, the presence of high levels of gamma-amino-n-butyric acid (GABA) under anaerobic conditions suggests a functional GABA bypass and a possible block in the TCA cycle at these conditions.  相似文献

Two industrial effluents, a pre-fermentation effluent and a post-fermentation effluent from a wheat starch production plant, were used as substrates for fuel ethanol production in anaerobic batch cultures using minimal nutritional amendment. The performances of three metabolically engineered xylose-utilizing Saccharomyces cerevisiae strains: TMB 3001 expressing XYL1, XYL2 and XKS1, redox metabolism modulated CPB.CR1 and glucose de-repressed CPB.CR2, as well as a reference strain CEN.PK 113-7D not fermenting xylose, were evaluated. For the recombinant strains a glucose consumption phase preceded the xylose consumption phase. In both effluents, biomass and ethanol production occurred predominantly during the glucose consumption phase, whereas xylitol and glycerol formation were predominant in the xylose consumption phase. Total specific ethanol productivities on glucose were 6-fold higher than on xylose in the pre-fermentation effluent and 15-fold higher than on xylose in the post-fermentation effluent. CPB.CR1 showed impaired growth compared to the two other xylose-utilizing strains, but displayed 18% increased ethanol yield in the post-fermentation effluent.  相似文献

Increased numbers of swine producers will be removing sludge from their anaerobic waste treatment lagoons in the next few years, due to sludge exceeding designed storage capacity. Information on availability of nitrogen (N) in the sludge is needed to improve application recommendations for crops. The objective of this study was to investigate possible effects of different companies and types of swine operations on the availability of N in sludge from their associated lagoons. A laboratory incubation study was conducted to quantify the availability of N (i.e. initial inorganic N plus the potentially mineralizable organic N) in the sludge. Nine sludge sources from lagoons of sow, nursery and finishing operations of three different swine companies were mixed with a loamy sand soil (200 mg total Kjeldahl N kg(-1) soil) and incubated at a water content of 0.19 g. water g(-1) dry soil and 25+/-2 degrees C for 12 weeks. Samples were taken at eight times over the 12-week period and analyzed for inorganic N (i.e. NH(4)-N and NO(3)-N) to determine mineralization of organic N in the sludge. Company and type of swine operation had no significant effects (P < 0.05) on the pattern of inorganic N accumulation over time. Thus, inorganic N accumulation from all sludge sources was fit to a first order equation [Nt = Ni + No (1-e(-kt)]. This relationship indicated that of the 200 mg of total sludge N added per kg soil, 23.5% was in the form of potentially mineralizable organic N (No) and 17.5% was in the form of inorganic N (Ni). The sum of these two pools (41%) represents an estimate of the proportion of total N in the applied sludge in plant available form after the 12 week incubation. While plant N availability coefficients were not measured in this study, the lack of significant company or type of swine operation effects on sludge N mineralization suggests that use of the same plant N availability coefficient for sludge from different types of lagoons is justifiable. The validity of this interpretation depends on the assumption that variation in other components of different sludge sources such as Cu and Zn does not differentially alter N uptake by the receiver crops.  相似文献

The tetrameric red fluorescent protein, DsRed, undergoes a rapid red to green color change evoked by short wavelength (lambda < 760 nm) femtosecond irradiation--a phenomenon that underpins the application of DsRed as an "optical highlighter" probe for tracking live cells, organelles, and fusion proteins. This color change results from selective bleaching of the "mature" red-emitting species of DsRed and an enhancement of emission from the "immature" green species, likely caused by dequenching of fluorescence resonance energy transfer occurring within the protein tetramer. Here, we have examined the role of residues known to influence the rate and completeness of chromophore maturation on the cellular and biophysical properties of DsRed mutants. Surprisingly, a single amino acid mutation (N42Q) with increased basal green emission yet rapid chromophore maturation displayed a multiphoton-evoked color change that was brighter, more consistent, more vivid, and easier to evoke than DsRed, despite the larger proportion of green chromophores. Rapidly maturing mutants with more complete chromophore maturation, exhibited little color change and increased resistance to multiphoton bleaching. We describe improved optical and cell biological properties for two DsRed-derived variants which we showcase in photolabeling studies, and discuss these data in terms of implications for fluorescence resonance energy transfer-based probes.  相似文献

Over the last decade or so, a wealth of research has established that bacteria communicate with one another using small molecules. These signals enable the individuals in a population to coordinate their behaviour. In the case of pathogens, this behaviour may include decisions such as when to attack a host organism or form a biofilm. Consequently, such signalling systems are excellent targets for the development of new antibacterial therapies. In this review, we assess how Gram-negative bacteria use small molecules for cell-cell communication, and discuss the main approaches that have been developed to interfere with it.  相似文献

Light petroleum extracts from thalli of Fucus vesiculosus (L.) and Laminaria digitata (Huds.) Lamour. stimulate growth of axenically cultured Fucus spiralis L. and Enteromorpha compressa (L.) Grev. in amounts per litre culture medium corresponding to one to 10 mg wet weight of the brown algae. This indicates the presence of a lipophilic, very active growth regulator in the blades. Extracts from the meristematic zone of Laminaria are much less active. Vitamins D3, D2 and 1,25-(OH)2-D3 at 1·10?8– 1·10?7 M increase the fresh weight of Enteromorpha compressa and Fucus spiralis up to 100% or more. The dry weight of Nemalion helminthoides (Veil, in With.) Batt. is also increased under the same conditions. Ergosterol and cholesterol are also growth-stimulating but higher concentrations (e.g. 1·10?6 M) are necessary. The effects of the D vitamins manifest themselves without any auxins being added to the medium.  相似文献

BACKGROUND: Previous studies in Denmark have shown that bus drivers and tramway employees were at an increased risk for developing several types of cancer and that bus drives from central Copenhagen have high levels of biomarkers of DNA damage.AIMS: The present study evaluates 1-hydroxypyrene concentrations and mutagenic activity in urine as biomarkers of exposure in non-smoking bus drivers in city and rural areas on a work day and a day off and in non-smoking mail carriers working outdoors (in the streets) and indoors (in the office). METHODS: Twenty-four hour urine samples were collected on a working day and a day off from 60 non-smoking bus drivers in city and rural areas and from 88 non-smoking mail carriers working outdoors (in the streets) and indoors (in the office). The concentration of 1-hydroxypyrene was measured by means of HPLC and the mutagenic activity was assessed by the Ames assay with Salmonella tester strain YG1021 and S9 mix. The N-acetyltransferase (NAT2) phenotype was used as a biomarker for susceptibility to mutagenic/carcinogenic compounds. RESULTS: Bus drivers excreted more 1-hydroxypyrene in urine than did mail carriers. The differences were slightly smaller when NAT2 phenotype, cooking at home, exposure to vehicle exhaust, and performing physical exercise after work were included. The NAT2 slow acetylators had 29% (1.29 [CI: 1.15-1.98]) higher 1-hydroxypyrene concentrations in urine than the fast acetylators. Male bus drivers had 0.92 revertants/mol creatinine [CI: 0.37-1.47] and female bus drivers 1.90 revertants/mol creatinine [CI: 1.01-2.79] higher mutagenic activity in urine than mail carriers. CONCLUSION: The present study indicates that bus drivers are more exposed to polycyclic aromatic hydrocarbons (PAH) and mutagens than mail carriers. Mail carriers who worked outdoors had higher urinary concentration of 1-hydroxypyrene, a marker of exposure to PAH, than those working indoors. The individual levels of urinary mutagenic activity were not correlated to excretion of 1-hydroxypyrene. This might be due to the fact that the most potent mutagenic compounds in diesel exhaust are not PAH but dinitro-pyrenes. Among bus drivers, fast NAT2 acetylators had higher mutagenic activity in urine than slow NAT2 acetylators and female bus drivers had higher mutagenic activity than male bus drivers.  相似文献

In seasonal species, photoperiod exerts tight regulation of reproduction to ensure that birth occurs at the most favorable time of yr. A distinct photoneuroendocrine circuit composed of the retina, suprachiasmatic nucleus (SCN) of the hypothalamus, and pineal gland transduces daylength into a rhythmic secretion of melatonin. The duration of the night-time rise of this hormone conveys daylength information to the organism. Melatonin is known to mediate the control of seasonal reproduction, but how it modulates sexual activity is far from understood. Recent data indicate that the product of the KiSS-1 gene is a potent stimulator of the hypothalamic-pituitary-gonadal axis and may play, together with its receptor GPR54, a central role in the neuroendocrine regulation of gonadotropin secretion. This article briefly reviews these findings and presents arguments that KiSS-1 could take part in the seasonal control of reproduction.  相似文献
