Leukotriene F4 (LTF4 and LTF4 sulfone have been synthesized and their biological activities determined in the guinea pig. LFT4 displayed comparable activity to LTD4 on guinea pig trachea and parenchyma but was less active on the ileum. When injected intravenously into the guinea pig, LTF4 induced a bronchoconstriction (ED50 16 μg Kg−1) which was blocked by indomethacin and FPL-55712 and was 50–100 X less potent than LTD4 in this assay. LTF4 sulfone was approximately 2–5 times less active than LTF4 and . When injected into guinea pig skin with PGE2 (100 ng); LTF4 and LTF4 sulfone (10–1000 ng) induced changes in vascular permeability. The order of potency in this assay was LTE4 sulfone = LTD4 = LTD4 sulfone > LTE4 > LTF4 = LTF4 sulfone.  相似文献

Nine dysmenorrheic women were run in EMG and thermal biofeedback procedures with concurrent autogenic relaxation practice. Significant reductions in subjective estimates of symptomology associated with dysmenorrhea were noted in all subjects. EMG levels correlated positively with the reductions in symptoms. Thermal levels did not correlate with EMG. In fact no consistent patterns in thermal measures were noted. However, thermal biofeedback cannot be ruled out as an effective treatment for dysmenorrhea since reductions in symptoms occurred during thermal biofeedback training. Another significant aspect of the present study is the effectiveness of long treatment procedures. A six month period was employed and significant reductions in symptoms were noted following two months of biofeedback treatment. Finally, the importance of beginning biofeedback treatment prior to onset of menstrual symptoms is indicated.  相似文献

Two radiometers measuring reflectance factors have been used at a height of 1.50 m above some grasslands in C. France. The results show that both spectral data are sensitive to the photosynthetic activity of the grassland. Measurements made in April, May, June and July show that grasslands have quite different spectral behaviour according to soil conditions or to grazing level. Grasslands on dry or wet soils may be separated from those of normal soils for which overgrazing and trampling affecting the growth of species, are shown by the different spectral values. In the rat, differentiation and cell proliferation both affect DNA methylation. We studied 5-methylcytosine at the inner cytosine of the sequence C-C-G-G, a common methylation site, using endonuclease MspI (which cleaves C-C-G-G- and C-mC-G-G), and its isoschizomer HpaII (which cleaves only C-C-G-G). DNA from all tissues and cell lines studied was methylated at C-C-G-G, at levels ranging from 45 to 80%, but the methylation sites were not distributed uniformly. Our analysis suggests a model in which cells contain variable amounts of three DNA methylation states, averaging 30–40, 70–80 and 95–100% methylation, respectively. One biological parameter that alters methylation is the prolferative state of the cell. We observed that NRK, a non-transformed cell line, increased its DNA methylation from 45 to 67% when monolayer cultures became confluent and non-dividing. We also observed that a class of repetitive DNA was completely methylated in DNA from all sources except a transformed cell line.  相似文献

We have analysed in detail the Na+ content and Na+ influx during fertilization and first divisions of the sea urchin egg (Paracentrotus lividus) using a filtration technique devised to eliminate rapidly contamination by the Na+ of external sea water. In the first 5 min following fertilization the egg fills up with Na+ (+ 30%). Thereafter Na+ is extruded and the Na+ content stabilizes at about 60% of the unfertilized egg level by the second cleavage (2 h). The initial increase in Na+ content is due to a large increase in Na+ influx already detected at 20 sec. The Na+ influx reaches its maximum at 1 min and its minimum at 5 min. H+ excretion follows the same kinetics. A second increase in Na+ influx is noted 5–10 min after fertilization; it reaches its maximum at prophase metaphase (30 min) and its minimum during cleavage (60 min). These oscillations in Na+ influx were observed for the first three divisions. Fertilization also immediately stimulates the Na+ efflux which remains elevated throughout the cell cycle and is responsible for the depletion of the Na+ content of the embryos. Activation of the eggs by weak amine bases (5 mM NH4Cl) which bypasses the early cortical reaction produces only a depletion in the Na+ content of the egg similar to that produced by fertilization. NH4Cl also increases the Na+ influx soon after fertilization, although no transient variations are noted.  相似文献

The incidence of the dietary source of energy on lipid transport and accumulation was investigated over a full nycthemeral cycle in adapted rats fed ad libitum. Starch, sucrose and lard were compared. Lipoprotein composition of the plasma, liver and plasma lipids and insulinemia were analyzed every 3 hours over 24 hours. The pattern of VLDL concentration was dependent on the nature of the energetic substrate. Feeding starch resulted in a remarkable stability of lipoproteins, liver and plasma lipids, despite clearcut diurnal variations in plasma non esterified fatty acids, insulinemia and liver glycogen. In sucrose-fed rats VLDL rose to a sharp maximum in the post prandial period (9-12:00) and were totally cleared by 18:00. In fat-fed rats, HDL were elevated during the night, suggesting a possible stimulation of their synthesis by dietary fat in the intestine. LDL were constantly elevated with peak values at 21:00 while VLDL were very low, even at night, despite elevated levels of non-esterified fatty acids. It is concluded that, in animals adapted to a high fat-diet, a high level of circulating non esterified fatty acids is not sufficient to promote the synthesis of VLDL. The main regulating factor appears to be the intensity of hepatic lipogenesis which is stimulated by sucrose and inhibited by lard. No correlation was found between variations in plasma VLDL and insulinemia.  相似文献

The uptake and metabolism of linoleic acid by rumen holotrichs (mainly Isotricha prostoma and I. intestinalis) has been examined in in vitro infusion experiments. Maximum absorption and metabolism of [1-14C]linoleate by 2 . 10(6) Isotricha suspended in 100 ml buffer was obtained using an infusion rate of 1.6 mg linoleate/h. After 90 min, 84% of the added substrate was recovered within the cells, mainly as free fatty acid or phospholipid. There was a rapid incorporation of radioactivity into phospholipid, mainly phosphatidylcholine, at the commencement of linoleate infusion but no further incorporation after about 40 min. The presence of bacteria during incubations, in approximately the same Isotricha/bacteria ratio as found in the rumen, reduced the uptake of linoleate and the accumulation of free fatty acid by holotrichs but the incorporation into phospholipid remained similar to that obtained in the absence of bacteria. Very little biohydrogenation of linoleic acid occurred in incubations with holotrichs alone. Bacterial suspensions converted linoleic acid to mainly trans monoene and a small amount of stearic acid, but in incubations containing both bacteria and holotrichs, both stearic acid and trans monoene were major products. Using the latter mixed culture, about 20% of the added [1-14C]linoleic acid was present in holotrich phospholipid of which 62% remained as octadecadienoic acid. The Isotricha population was 3 . 10(3)--2 . 10(4)/ml rumen fluid and it contributed about 23% of the linoleic acid in the rumen of a cow on a hay diet.  相似文献

Glutamine synthetase (GS) level is positively influenced by exogenously supplied sucrose in isolated pea roots (similarly as nitrate reductase - NR), glutamate dehydrogenase (GDH) level negatively. Comparison with previous results shows that GS level decreases more slowly than NR level when sucrose is omitted from the medium; the rate of changes in GS level corresponds rather to that in GDH level. The increase in GDH level in the tips of isolated roots cultivated in the medium lacking sucrose stops after approx. 24 h, but continues for at least 72 h in more mature root parts. GS level decreases during the first 24 h in the tips of isolated roots (compared with roots of intact seedlings) cultivated both with sucrose and without it (without sucrose more), however it again rises in the course of further cultivation with sucrose. The differences in GS and GDH levels caused by omission of sucrose are small in isolated roots from which root tips were removed, the difference in NR level in decapitated roots is similar to that found in isolated roots with root tips left. Decapitated isolated roots cultivated without sucrose contain higher amounts of soluble sugars than corresponding roots with root tips left. These facts are dismissed with regard to sugar consumption, transport, and compartmentalisation, and with respect to production in root tips and other plant parts of unknown compounds involved in GS and GDH regulation. The results obtained suggest that GDH functions in pea roots in the deaminating direction.  相似文献

The biological activity and a possible modulatory role of the N-terminal tetrapeptide Ser-Tyr-Ser-Met from alpha-MSH/ACTH was tested in the Anolis melanophore assay, the Xenopus melanophore assay, tyrosinase stimulation in mouse melanoma cells and in excessive grooming in the rat. ACTH1-4 did not exhibit biological activity in any of these four assays nor did it have modulatory properties in the Xenopus and the melanoma cell assay. However, in the Anolis assay ACTH1-4 potentiated pigment dispersion induced by alpha-MSH, alpha-MSH5-13 and ACTH1-24 by a factor of about 2. In the grooming assay ACTH1-4 potentiated the effects of alpha-MSH, alpha-MSH5-13, ACTH1-16 and ACTH5-16, but not those of ACTH1-24. Oxidized ACTH1-4 was without biological activity and potentiating properties in all four assays. This study shows that small fragments of the pro-opiomelanocortin precursor, which are devoid of biological activity, can modulate peripheral and central actions of alpha-MSH/ACTH.  相似文献
