Objective

To develop a high-sensitivity immunochromatographic test for fumonisin B1 in plant extracts.

Results

Unlike conventional immunochromatographic tests, this assay is performed in two stages: competitive reaction with free specific antibodies and identifying immune complexes by their interaction with the anti-species antibody-conjugated gold nanoparticles. The use of a new geometry for the test strip membranes and a novel reagent application method ensures the proper order of these stages without additional manipulations. The contact of the ready-to-use test strip with the liquid sample suffices in initiating all stages of the assay and obtaining test results. The developed test was used on corn extracts; its instrumental limit of fumonisin B1 detection was 0.6 ng ml?1 at 15 min of assay duration.

Conclusions

The proposed approach is flexible and can be used for a wide range of low molecular compounds. The use of anti-species antibody-conjugated gold nanoparticles in immunochromatography significantly facilitates the development of test systems by eliminating the need to synthesize and characterize the conjugates with specific antibodies for each new compound to be detected.

The stability of single‐crystalline/monocrystalline‐like perovskite film is expected to be better than its microcrystalline counterparts. In the present work, highly orientated perovskite thin films (CH3NH3PbI3–xClx) are prepared by means of aquointermediates assisted solution process. It displays super‐duper preferred‐orientation along <110> direction that is close to the single crystal, and its diffraction intensity ratio of (110)/(310) is nearly two orders of magnitude higher in contrast to the films that prepared by traditional way. Owing to its superior performances, e.g., highly crystallized quality, stress‐free inside films, longer electron lifetime, faster temporal response time, etc., the highly orientated perovskite‐based solar cells accordingly allow realizing high efficiency while improving its thermal stability.  相似文献

To advance polymer solar cells (PSCs) toward real‐world applications, it is crucial to develop materials that are compatible with a low‐cost large‐scale manufacturing technology. In this context, a practically useful polymer should fulfill several critical requirements: the capability to provide high power conversion efficiencies (PCEs) via low‐cost fabrication using environmentally friendly solvents under mild thermal conditions, resulting in an active layer that is thick enough to minimize defects in large‐area films. Here, the development of new photovoltaic polymers is reported through rational molecular design to meet these requirements. Benzodithiophene (BDT)‐difluorobenzoxadiazole (ffBX)‐2‐decyltetradecyl (DT), a wide‐bandgap polymer based on ffBX and BDT emerges as the first example that fulfills the qualifications. When blended with a low‐cost acceptor (C60‐fullerene derivative), BDT‐ffBX‐DT produces a PCE of 9.4% at active layer thickness over 250 nm. BDT‐ffBX‐DT devices can be fabricated from nonhalogenated solvents at low processing temperature. The success of BDT‐ffBX‐DT originates from its appropriate electronic structure and charge transport characteristics, in combination with a favorable face‐on orientation of the polymer backbone in blends, and the ability to form proper phase separation morphology with a fibrillar bicontinuous interpenetrating network in bulk‐heterojunction films. With these characteristics, BDT‐ffBX‐DT represents a meaningful step toward future everyday applications of polymer solar cells.  相似文献

Plant carbon (C) and nitrogen (N) stoichiometry play an important role in the maintenance of ecosystem structure and function. To decipher the influence of changing environment on plant C and N stoichiometry at the subcontinental scale, we studied the shoot and root C and N stoichiometry in two widely distributed and dominant genera along a 2,200‐km climatic gradient in China's grasslands. Relationships between C and N concentrations and soil climatic variables factors were studied. In contrast to previous theory, plant C concentration and C:N ratios in both shoots and roots increased with increasing soil fertility and decreased with increasing aridity. Relative N allocation shifted from soils to plants and from roots to shoots with increasing aridity. Changes in the C:N ratio were associated with changes in N concentration. Dynamics of plant C concentration and C:N ratios were mainly caused by biomass reallocation and a nutrient dilution effect in the plant‐soil system. Our results suggest that the shifted allocation of C and N to different ecosystem compartments under a changing environment may change the overall use of these elements by the plant‐soil system.  相似文献

Resveratrol (3,5,4-trihydroxystilbene) is a natural polyphenolic compound found in grapes and red wine and has been shown to exert protective effects on the liver preventing lipid accumulation induced by a high-fat diet. However, no studies have shown that the nutritional resveratrol intake by the parental generation has modified lipogenesis in an adult offspring. The aim of this study was to investigate whether maternal resveratrol intake during lactation affects lipogenesis in adult male rat offspring, and if it does, what is the molecular mechanistic basis. Six male pups born from mothers given a control diets during lactation (CC group) and six male pups born from mothers given a control diet as well as resveratrol during lactation (CR group) were fed a standard diet until sacrifice at 36 weeks. Adult male offspring from mothers given resveratrol during lactation (CR group) had lower body weight from the fourth week of lactation until adulthood, but no significant change was observed in the relative food intake. Low levels of plasma triacylglycerol were found in the CR group compared to the CC group. Histopathological analysis of the livers of adult male rat offspring revealed lipid accumulation in hepatocytes in the CC group, whereas lipid droplets were rare in the CR group. Hepatic protein levels of AMPK-phosphorylated at ser403, Sirt1, and Nampt in the CR group were upregulated significantly compared to the CC group. These results indicated the maternal resveratrol intake during lactation-induced activation of AMPK through Sirt1 upregulation. In this study, significant upregulation of the levels of precursor of sterol regulatory element binding protein-1c (SREBP-1c) and downregulation of the ratio of active-SREBP-1c/precusor-SREBP-1c were observed in the CR group compared to the CC group. These results suggested that proteolytic processing of SREBP-1c was suppressed by AMPK in the livers of the CR group. It is well known that SREBP-1c regulates the lipogenic pathway by activating genes involved in triglyceride and fatty acid synthesis. The present study showed significant downregulation of hepatic fatty acid synthase (FAS) and acetyl-CoA carboxylase (ACC) levels in the CR group. These results indicated that maternal resveratrol intake during lactation suppressed SREBP-1c cleavage and nuclear translocation and repressed SREBP-1c target gene expression such as FAS and ACC in the livers of adult male offspring. These changes attenuate hepatic triacylglycerol and fatty acid synthesis in adult male offspring.  相似文献

Butanol is an ideal renewable biofuel which possesses superior fuel properties. Previously, butanol-producing symbiotic system TSH06 was isolated in our lab, with microoxygen tolerance ability. To boost butanol yield for large-scale industrial production, TSH06 was used as parental strain and subjected to atmospheric and room temperature plasma (ARTP) and four rounds of genome shuffling (GS). ARTP mutant and GS strain were co-cultured with facultative anaerobic Bacillus cereus TSH2 to form a symbiotic system with microoxygen tolerance, which was then subjected to fermentation. Relative messenger RNA (mRNA) level of key enzyme gene was measured by real-time PCR. The highest butanol titer of TS4-30 reached 15.63 g/L, which was 34% higher than TSH06 (12.19 g/L). Compared with parental strain, mRNA of acid-forming gene in TS4-30 decreased in acidogenesis phase, while solvent-forming gene increased in solventogenesis phase. This gene expression pattern was consistent with high butanol yield and low acid level in TS4-30. In summary, symbiotic system TS4-30 was obtained with butanol titer improvement and microoxygen tolerance.

Sleep is a profound regulator of cellular immunity, and the curtailment of sleep in present day lifestyle leads to disruption of neuro-immune–endocrine interactions. No therapeutic remedy is yet known for the amelioration of detrimental effects caused by sleep deprivation (SD). The current study was aimed to elucidate the effects of acute SD on immune function and its modulation by water extract from leaves of Withania somnifera (ASH-WEX). Three groups of animals, i.e. Vehicle-Undisturbed sleep (VUD), Vehicle-Sleep deprived (VSD) and ASH-WEX fed sleep deprived (WSD) rats were tested for their anxiety-like behaviour and further used for the study of inflammatory and apoptotic markers expression in piriform cortex and hippocampus regions of the brain. VSD animals showed high level of anxiety in elevated plus maze test, which was ameliorated in WSD group. The stress induced expression of inflammatory and immune response markers GFAP, TNFα, IL-6, OX-18 and OX-42 in VSD animals was found to be modulated by ASH-WEX. Further, the stress induced apoptosis was suppressed in WSD group as indicated by expression of NF-κB, AP-1, Bcl-xL and Cytochrome c. This study provides scientific validation to the anxiolytic, anti-inflammatory and anti-apoptotic properties of ASH-WEX, which may serve as an effective dietary supplement for management of SD induced stress and associated functional impairments.  相似文献
