When assessing changes in populations of species, it is essential that the methods used to collect data have some level of precision and preferably also good accuracy. One commonly used method to collect pollinators is colour pan traps, but this method has been suggested to be biased by the abundance of surrounding flowers. The present study evaluated the relationship between pan trap catches and the frequency of flowers on small (25 m2) and large (2–6 ha) spatial scales. If pan traps work well, one should assume a positive relationship, that is, more insects caught when they have more food. However, in contrast, we found that catches in pan traps were often negatively affected by flower frequency. Among the six taxa evaluated, the negative bias was largest in Vespoidea and Lepturinae, while there was no bias in solitary Apoidea (Cetoniidae, Syrphidae and social Apoidea were intermediate). Furthermore, red flowers seemed to contribute most to the negative bias. There was also a tendency that the negative bias differed within the flight season and that it was higher when considering the large spatial scale compared to the small one. To conclude, pan trap catches may suffer from a negative bias due to surrounding flower frequency and color. The occurrence and magnitude of the negative bias were context and taxon dependent, and therefore difficult to adjust for. Thus, pan traps seem less suited to evaluate differences between sites and the effect of restoration, when gradients in flower density are large. Instead, it seems better suited to monitor population changes within sites, and when gradients are small.  相似文献

Propensity scoring (PS) is an established tool to account for measured confounding in non-randomized studies. These methods are sensitive to missing values, which are a common problem in observational data. The combination of multiple imputation of missing values and different propensity scoring techniques is addressed in this work. For a sample of lymph node-positive vulvar cancer patients, we re-analyze associations between the application of radiotherapy and disease-related and non-related survival. Inverse-probability-of-treatment-weighting (IPTW) and PS stratification are applied after multiple imputation by chained equation (MICE). Methodological issues are described in detail. Interpretation of the results and methodological limitations are discussed.  相似文献

Many horse owners tend to group horses according to gender, in an attempt to reduce aggressive interactions and the risk of injuries. The aim of our experiment was to test the effects of such gender separation on injuries, social interactions and individual distance in domestic horses. A total of 66 horses were recruited from 4 different farms in Norway and Denmark and divided into six batches. Within each batch, horses were allotted into one mare group, one gelding group and one mixed gender group, with most groups consisting of three or four animals. After 4–6 weeks of acclimatisation, a trained observer recorded all social interactions using direct, continuous observation 1 h in the morning and 1 h in the afternoon for three consecutive days. Recordings of the nearest neighbour of each horse were performed using instantaneous sampling every 10 min. The horses were inspected for injuries before grouping, day 1 after grouping and after 4–6 weeks. No significant effect of gender composition was found on social interactions (P > 0.05), spacing (P > 0.07) or injuries (P > 0.23). Eighty percent of all aggressive interactions recorded were threats, not involving physical contact. Horses with the smallest space allowance showed the highest mean number of aggressive interactions (28.6 ± 6.1 interactions per 6 h) compared to the mean of all the other batches (8.3 ± 1.0 interactions per 6 h). Very few injuries were found and most were superficial. In conclusion, gender composition does not seem to have any effect on aggression level, spacing or injuries. However, the early social experience of horses, management of feeding and space allowance probably represents more important factors for successful group housing of domestic horses.  相似文献

Site-directed spin labeling (SDSL), the site-specific incorporation of nitroxide spin-labels into a protein, has allowed us to investigate ligand-induced conformational changes in the ligand-binding domain of human estrogen receptor alpha (hERalpha-LBD). EPR (electron paramagnetic resonance) spectroscopy of the nitroxide probe attached to ER produces different spectra depending upon the identity of the bound ligand; these differences are indicative of changes in the type and degree of motional character of the spin-label induced by different ligand-induced conformations of labeled ER. Visual inspection of EPR spectra, construction of B versus C cross-correlation plots, and cross-comparison of spectral pairs using a relative squared difference (RSD) calculation allowed receptor-ligand complexes to be profiled according to their conformational character. Plotting B and C parameters allowed us to evaluate the liganded receptor according to the motional characteristics of the attached spin-label, and they were particularly illustrative for the receptor labeled at position 530, which had motion between the fast and intermediate regimes. RSD analysis allowed us to directly compare the similarity or difference between two different spectra, and these comparisons produced groupings that paralleled those seen in B versus C cross-correlation plots, again relating meaningfully with the pharmacological nature of the bound ligand. RSD analysis was also particularly useful for qualifying differences seen with the receptor labeled at position 417, which had motion between the intermediate and slow motional regimes. This work demonstrates that B and C formulas from EPR line shape theory are useful for qualitative analysis of spectra with differences subtler than those that are often analyzed by EPR spectroscopists. This work also provides evidence that the ER can exist in a range of conformations, with specific conformations resulting from preferential stabilization of ER by the bound ligand. Furthermore, it documents the complexity and uniqueness of the ligand-receptor structure, and highlights the fact that structural differences exist between the receptor bound with ligands of different pharmacological character that, nevertheless, produce similar crystal structures.  相似文献

Experiments were conducted to determine whether the biogenic amines octopamine (OA) and 5-hydroxytryptamine (5-HT) exert modulatory effects on pheromone responsiveness and random locomotor activity in male gypsy moths. When injected into males, OA significantly enhanced sensitivity to pheromone, while 5-HT enhanced general locomotor activity, results that were very similar to those previously shown for the cabbage looper. Maximal effect of the amines, however, was observed when injection occurred just prior to the onset of scotophase, rather than photophase, as we had originally hypothesized for this diurnally active insect. Male gypsy moths also displayed a prominent scotophase response, with sensitivity to pheromone greater in the scotphase compared with photophase, but with the level of random locomotor activity lower in scotophase than in photophase. The upwind flight behavior of males to a pheromone source in a wind tunnel, as well as the time spent at the source, were also significantly different in the two light regimes. Furthermore, when exposed to a 1 h scotophase (instead of the normal 8), or to continuous dark conditions, while males exhibited response to pheromone and locomotor activity during the same scotophase and photophase periods as observed in a 16:8 light : dark cycle, the levels of response, as well as qualitative aspects of the upwind flight behaviors in both periods were a function of the light intensity. Our combined results suggest that male gypsy moths display a bimodal rhythm of locomotor and pheromone response over the diel cycle, with light intensity and scotophase onset providing critical cues for the expression of behaviors, as well as the modulatory action of the amines. © 1992 Wiley-Liss, Inc.  相似文献

Baboons (genus Papio) are an interesting phylogeographical primate model for the evolution of savanna species during the Pleistocene. Earlier studies, based on partial mitochondrial sequence information, revealed seven major haplogroups indicating multiple para‐ and polyphylies among the six baboon species. The most basal splits among baboon lineages remained unresolved and the credibility intervals for divergence time estimates were rather large. Assuming that genetic variation within the two studied mitochondrial loci so far was insufficient to infer the apparently rapid early radiation of baboons we used complete mitochondrial sequence information of ten specimens, representing all major baboon lineages, to reconstruct a baboon phylogeny and to re‐estimate divergence times. Our data confirmed the earlier tree topology including the para‐ and polyphyletic relationships of most baboon species; divergence time estimates are slightly younger and credibility intervals narrowed substantially, thus making the estimates more precise. However, the most basal relationships could not be resolved and it remains open whether (1) the most southern population of baboons diverged first or (2) a major split occurred between southern and northern clades. Our study shows that complete mitochondrial genome sequences are more effective to reconstruct robust phylogenies and to narrow down estimated divergence time intervals than only short portions of the mitochondrial genome, although there are also limitations in resolving phylogenetic relationships. The calcareous green algaHalimeda can be a substantial contributor to aragonite sediment in reef ecosystems. In contrast to coral aragonite, little is known about the trace and minor element composition ofHalimeda aragonite, so it is difficult to test oceanographic hypotheses about factors controlling its past growth. We investigated adapting trace element cleaning protocols for modern and HoloceneHalimeda aragonite, modern and HoloceneHalimeda trace and minor element compositions, and the potential utility ofHalimeda aragonite for paleoceanographic investigations. We successfully adapted and applied sample treatment protocols developed for measuring trace elements in coral aragonite (generally less than 500 y old) toHalimeda aragonite (modern to approximately 5000 y old in this study). ModernHalimeda aragonite from John Brewer Reef in the Central GBR had mean Cd/Ca ratios of 5.19 ± 1.68 nmol/mol forHalimeda micronesica and 2.35 ± 0.38 nmol/mol for three closely related species important in bioherm accumulationHalimeda copiosa, Halimeda hederacea, andHalimeda opuntia. Mn/Ca ratios, with means from 89–239 nmol/mol for these four species, showed both intra-and inter-specific variability. Sr/Ca ratios (10.9 ± O.1 mmol/mol) and Mg/Ca ratios (1.35 ± 0.26 mmol/mol) were similar for all samples. HoloceneHalimeda aragonite samples from cores of two bioherms in the northern GBR seemed well preserved on the basis of mineralogy and Sr/Ca and Mg/Ca ratios similar to those in modernHalimeda aragonite. Cd/Ca ratios (overall mean 0.96 ± 0.15 nmol/mol) were lower than those measured in the modernHalimeda from the central GBR location. However, Mn/Ca ratios in both cores were substantially higher than in modernHalimeda aragonite. While it may be possible to extract paleoceanographic information fromHalimeda aragonite, substantial care is needed to evaluate and avoid the effects of post-depositional alteration.  相似文献

Ro52 is one of the major autoantigens targeted in the autoimmune disease Sj?gren syndrome. By sequence similarity, Ro52 belongs to the RING-B-box-coiled-coil (RBCC) protein family. Disease-related antibodies bind Ro52 in a conformation-dependent way both in the coiled-coil region and in the Zn2+-binding Ring-B-box region. Primarily associated with Sj?gren syndrome, Ro52 autoantibodies directed to a specific, partially structured epitope in the coiled-coil region may also induce a congenital heart block in the fetus of pregnant Ro52-positive mothers. To improve our understanding of the pathogenic effects of autoantibody binding to the Zn2+-binding region, a multianalytical mapping of its structural, biophysical, and antigenic properties is presented. Structure content and ligand binding of subregions, dissected by peptide synthesis and subcloning, were analyzed by fluorescence and circular dichroism spectroscopy. A novel matrix-assisted laser desorption ionization time-of-flight mass spectrometry strategy for time-resolved proteolysis experiments of large protein domains was developed to facilitate analysis and to help resolve the tertiary arrangement of the entire RBCC subregion. The linker region between the RING and B-box motifs is crucial for full folding, and Zn2+ affinity of the RING-B-box region is further protected in the entire RBCC region and appears to interact with the coiled-coil region. Murine monoclonal antibodies raised toward the RING-B-box region were primarily directed toward the linker, further supporting a highly functional role for the linker in a cellular environment. Taken together with our previous analysis of autoantigenic epitopes in the coiled-coil region, localization of autoantigenic epitopes in Ro52 appears closely related to molecular functionalities.  相似文献
