Background

The Continuous Performance Test-Identical Pairs version (CPT-IP) is a well-established measure of sustained attention, and its more challenging versions are particularly suited to detect subtle processing deficits in patients with schizophrenia. However, while there are few longitudinal samples for the CPT-IP, no study has addressed stability for more than two month in patients with schizophrenia. Assessing long-term test-retest reliability of the CPT-IP would facilitate the ability of clinicians to draw conclusions from studies involving interventions as long term cognitive or pharmacological treatments. The present study assessed 12 month test-retest reliability for the two most challenging versions of CPT-IP (4-digit and shapes) in a matched sample of clinically stable schizophrenia outpatients and healthy controls.

Methods

Fifty clinically stable schizophrenia outpatients and 50 healthy controls were assessed with the CPT-IP for the 4-digit and shape conditions. From these, 40 patients and 47 controls were reassessed with an average interval of 12.3 months between test sessions. Test-retest reliability was analyzed with Pearson correlations and results were compared with previous data involving healthy controls and short-term studies in patients with schizophrenia.

Results

Especially d’ and hit rate discriminated well between patients with schizophrenia and healthy controls for both CPT-IP conditions and at both test sessions. Healthy controls demonstrated sufficient long term test-retest correlations of d’, hit rate and reaction time for both the 4-digit and shape conditions. However, in schizophrenia patients, long-term reliability correlations were at best moderate for d’ and hit rate only.

Conclusions

The current study provides further evidence that d’ and hit rate yield consistent cross-sectional discrimination sensitivity. At best moderate long-term test-retest reliability of d’ in schizophrenia outpatients may be not sufficient for practical use of this measure in long term clinical trials.  相似文献

Solanapyrone A, a phytotoxin and enzyme inhibitor isolated from a fungus (SUT 01B1-2) selectively inhibits the activities of mammalian DNA polymerase beta and lambda (pol beta and lambda) in vitro. The IC50 values of the compound were 30 microm for pol beta and 37 microm for pol lambda. Because pol beta and lambda are in a family and their three-dimensional structures are thought to be highly similar to each other, we used pol beta to analyze the biochemical relationship with solanapyrone A. On pol beta, solanapyrone A antagonistically competed with both the DNA template and the nucleotide substrate. BIAcore analysis demonstrated that solanapyrone A bound selectively to the N-terminal 8-kDa domain of pol beta. This domain is known to bind single-stranded DNA, provide 5'-phosphate recognition of gapped DNA, and cleave the sugar-phosphate bond 3' to an intact apurinic/apyrimidinic (AP) site (i.e. AP lyase activity) including 5'-deoxyribose phosphate lyase activity. Solanapyrone A inhibited the single-stranded DNA-binding activity but did not influence the activities of the 5'-phosphate recognition in gapped DNA structures and the AP lyase. Based on these results, the inhibitory mechanism of solanapyrone A is discussed.  相似文献

Two genetic variants with increased organ size were independently derived from diploid (2n = 2x = 16) chickpea (Cicer arietinum L.) line ICC 640. Radiation-induced mutant PM1101 had greatly enlarged leaves, leaflets, and pods, and an elongated stem with longer internodes but fewer nodes than ICC 640. F1, F2, and F3 data from crosses with ICC 640 showed that the mutant characteristics of PM1101 were the pleiotropic effects of a single, recessive genetic factor. For purposes of comparison, tetraploid derivatives of ICC 640 were produced by colchicine treatment of seed. In the tetraploids, leaflets and pods were enlarged, but less dramatically than in PM1101. Enlarged pollen grains and stomatal guard cells, and increased guard cell chloroplast number were found in tetraploids but not in PM1101, while both variants produced fewer seeds than ICC 640. Mutant PM1101 and the tetraploids represent two very different manifestations of gigantism in chickpea.  相似文献

DNA of lambda and seven related phages was digested with restriction endonuclease, EcoRI. Seven different fragment patterns were observed, only two of the eight phages showing identical profiles. When assessing changes in populations of species, it is essential that the methods used to collect data have some level of precision and preferably also good accuracy. One commonly used method to collect pollinators is colour pan traps, but this method has been suggested to be biased by the abundance of surrounding flowers. The present study evaluated the relationship between pan trap catches and the frequency of flowers on small (25 m2) and large (2–6 ha) spatial scales. If pan traps work well, one should assume a positive relationship, that is, more insects caught when they have more food. However, in contrast, we found that catches in pan traps were often negatively affected by flower frequency. Among the six taxa evaluated, the negative bias was largest in Vespoidea and Lepturinae, while there was no bias in solitary Apoidea (Cetoniidae, Syrphidae and social Apoidea were intermediate). Furthermore, red flowers seemed to contribute most to the negative bias. There was also a tendency that the negative bias differed within the flight season and that it was higher when considering the large spatial scale compared to the small one. To conclude, pan trap catches may suffer from a negative bias due to surrounding flower frequency and color. The occurrence and magnitude of the negative bias were context and taxon dependent, and therefore difficult to adjust for. Thus, pan traps seem less suited to evaluate differences between sites and the effect of restoration, when gradients in flower density are large. Instead, it seems better suited to monitor population changes within sites, and when gradients are small.  相似文献
