通过13个地点蒙古栎群落物种相似性与两地之间距离的回归比较,发现蒙古栎群落物种相似性的大小一般决定于不同地点之间的地理位置,两地之间距离越近,其相似性越高,反之则越低,但是群落不同的演替阶段也影响群落的相似性。通过分析蒙古栎群落物种组成的差异及物种相似性系数的比较,蒙古栎群落可分为大兴安岭蒙古栎群落、长白山蒙古栎群落、辽西蒙古栎群落和辽冀蒙古栎群落等几种类型。 相似文献

Hepatocellular carcinoma (HCC) is the most common type of liver cancer, and treatment options for HCC are limited. In addition, the discovery of new natural compounds with anti-hepatocarcinoma activity is attracting increasing attention. For this reason, phytochemical investigation of Croton crassifolius led to the isolation of 17 diterpenoids, including three new clerodane diterpenoids, named crassifolius A-C (1–3), along with 14 known ones (4–17). Their structures were established by 1D, 2D NMR, HR-ESI-MS, detailed calculated electronic circular dichroism (ECD) spectra and the assistance of quantum chemical predictions (QCP) of 13C NMR chemical shifts. The cytotoxicities of all these compounds against human liver cancer lines (HepG2 and Hep3B) were determined. Among them, compound 1 exhibited good cytotoxicity with IC50 value of 17.91 μM against human liver tumor cells Hep3B. Following further studies of the anti-tumor mechanism of compound 1-induced cell growth inhibition, we found that compound 1 caused apoptotic cell death in Hep3B cells by detecting morphologic changes and Western blotting analysis. 相似文献

目的:制备抗人突触小体相关蛋白25(SNAP25)的鼠源单克隆抗体。方法:利用大肠杆菌表达SNAP25蛋白,纯化后免疫BALB/c小鼠制备杂交瘤细胞,筛选针对SNAP25的阳性杂交瘤细胞株,鉴定抗体亚型;用杂交瘤细胞株制备腹水单抗,纯化后利用SDS-PAGE检测抗体纯度。结果:表达并纯化得到纯度大于90%的SNAP25蛋白,免疫小鼠后经2轮筛选得到12株阳性杂交瘤细胞株,其中抗体重链包括IgG1、IgG2型,轻链大部分为κ链;选择具有相对较高抗原结合活性的14号杂交瘤细胞株制备腹水,纯化后得到纯度大于90%的抗体。结论:获得1株高纯度的针对SNAP25的鼠源单克隆抗体,为肉毒毒素的检测奠定了基础。 相似文献

Cucurbits are economically important crops worldwide. The genomic data of many cucurbits are now available. However, functional analyses of cucurbit genes and noncoding RNAs have been impeded because genetic transformation is difficult for many cucurbitaceous plants. Here, we developed a set of tobacco ringspot virus (TRSV)-based vectors for gene and microRNA (miRNA) function studies in cucurbits. A TRSV-based expression vector could simultaneously express GREEN FLUORESCENT PROTEIN (GFP) and heterologous viral suppressors of RNA silencing in TRSV-infected plants, while a TRSV-based gene silencing vector could knock down endogenous genes exemplified by PHYTOENE DESATURASE (PDS) in Cucumis melo, Citrullus lanatus, Cucumis sativus, and Nicotiana benthamiana plants. We also developed a TRSV-based miRNA silencing vector to dissect the functions of endogenous miRNAs. Four representative miRNAs, namely, miR159, miR166, miR172, and miR319, from different cucurbits were inserted into the TRSV vector using a short tandem target mimic strategy and induced characteristic phenotypes in TRSV-miRNA-infected plants. To improve the enzymatic hydrolytic efficiency and reduce production cost, a statistically designed experimental approach was used to optimize the composition of cellulase mixture so as to maximize the amount of glucose produced from steam-exploded corn stover (SECS). Using seven purified enzymes (cellobiohydrolases, Cel7A, Cel6A, Cel6B; endoglucanases, Cel7B, Cel12A, Cel61A; and beta-glucosidase) from Trichoderma viride T 100-14 mutant strain, a multi-enzyme mixture was constituted after screening and optimization. The final optimal composition (mol%) of the multi-enzyme mixture was Cel7A (19.8%), Cel6A (37.5%), Cel6B (4.7%), Cel7B (17.7%), Cel12A (15.2%), Cel61A (2.3%) and beta-glucosidase (2.8%). The subsequent verification experiments followed by glucose assay together with scanning electron microscopy (SEM) observation confirmed the validity of the models. The multi-enzyme mixture displayed a high performance in converting the cellulosic substrate (SECS). The amount of glucose produced (15.5mg/ml) was 2.1 times as that of the crude cellulase preparation. The results indicated that the optimized cellulase mixture is an available and efficient paradigm for the hydrolysis of lignocellulosic substrate. The enhanced cellulolytic activity displayed by the constructed cellulase mixture could be used as an effective tool for producing bioethanol efficiently from cellulose. 相似文献

矿区分离丛枝菌根真菌对万寿菊吸Cd潜力影响 总被引:2,自引:0,他引:2
盆栽试验研究土壤不同施Cd水平(0、5、20、50μg/g)下,接种矿区污染土壤中丛枝菌根真菌对万寿菊根系侵染率、植株生物量及Cd吸收与分配的影响。结果表明:接种丛枝菌根真菌显著提高Cd胁迫下万寿菊的根系侵染率和植株生物量;随着施Cd水平提高,各处理植株Cd浓度显著增加。各施Cd水平下万寿菊地上部Cd吸收量远远高于根系Cd吸收量,在土壤施Cd量达到50μg/g时,接种处理地上部Cd吸收量是根系的3.48倍,对照处理地上部Cd吸收量是根系的1.67倍;同一施Cd水平下接种处理植株Cd吸收量要显著高于对照。总体上,试验条件下污染土壤中分离的丛枝菌根真菌促进了万寿菊对土壤中Cd的吸收,并在一定程度上增加Cd向地上部分的运转,表现出植物提取的应用潜力。 相似文献

生物多样性保护与持续利用已为各国政府和大众所重视,现仅就国际和国内关注的生物多样性研究的热点加以介绍。对DIVERSITAS这一生物多样性科学的国际项目进行评价,对生物多样性起源、维持、编目、监测等方面提出补充意见。生物多样性恢复研究是以生态系统动态演替为理论基础的,此方面工作亟待加强。生物多样性信息系统是生物多样性科学的重要组成部分。农田生物多样性研究把生物多样性与农业生产紧密结合。生物安全是当前国际上极为关注的问题。文中对其主要研究内容提出作者的观点。 相似文献
