Molecular map of Chromosome 19 including three genes affecting bleeding time: ep,ru, and bm

The mouse ruby eye (ru) and pale ear (ep) pigment dilution genes cause platelet storage pool deficiency (SPD) and prolonged bleeding times. The brachymorphic (bm) gene, in addition to causing skeletal abnormalities, is also associated with prolonged bleeding times. All three hemorrhagic genes are found within 10 cM on Chromosome (Chr) 19. In this study, 15 microsatellite markers and five cDNAs, spanning 21 cM of Chr 19, were mapped in relation to the bm, ep, and ru genes in 457 progeny of an interspecific backcross utilizing the highly inbred strain PWK derived from the Mus musculus musculus species. Several markers were found to be closely linked to the three genes and should be useful as entry points in their eventual molecular identification.

芦苇耐盐变异植株及其细胞学鉴定

用甲基磺酸乙酯（ＥＭＳ）处理芦苇（Ｐｈｒａｇｍ ｉｔｅｓｃｏｍ ｍ ｕｎｉｓ Ｔｒｉｎ．）胚性愈伤组织。从处理后的愈伤组织诱导获得芦苇耐盐变异植株Ｒ５００２－１２。变异植株能在含有１％ ＮａＣｌ的ＭＳ培养基上生长。细胞学检查变异植株是混倍体,染色体数目变异范围在１００至３３ 之间。分蘖植株具有相似的形态学及染色体变异特性     

Somatic hybridization between Citrus reticulata and Citropsis gabunensis through electrofusion

Protoplasts from embryogenic calli of Citrus reticulata Blanco cv. Ponkan and Citropsis gabunensis (Engl.) Swing. & M. Kell (Cabon Cherry Orange), were isolated and fused using electric current. Maximum fusion frequency was obtained with AC at 75 kV/cm (1.0 MHz) for 15 s, followed by DC square-wave pulses at 1.25 kV/cm for 40 s. Fusion-treated protoplasts were cultured on MT medium containing no growth regulators, solidified with 0.6% Bacto Difco agar. Protoplast-derived calli were proliferated on MT medium containing 1 mg/l zeatin and 0.9% agar. A total of 31 lines of somatic hybrid calli were obtained by screening on the basis of chromosome count and isozyme analysis. The somatic hybrids were tetraploid (2n=36). Plants were regenerated from the calli via somatic embryogenesis. 人重组IL6／IL2融合蛋白的变性、复性及纯化

经超声破碎,分离已表达CH925包涵体,较系统地研究变性剂浓度、融合蛋白浓度对蛋白折叠的影响.在还原型及氧化型谷胱甘肽复性条件下,成功地将融合蛋白CH925折叠成具有IL6及IL2双活性蛋白,IL6的比活为2.3×10⁸U/mg, IL2比活为2.2×10⁶U/mg.经阴离子交换、凝胶过滤层析,获得一定纯度的CH925,配合反相HPLC.洗脱收集蛋白峰,CH925纯度为98%.     

A strong protein unfolding activity is associated with the binding of precursor chloroplast proteins to chloroplast envelopes

Protein conformational changes related to transport into chloroplasts have been studied. Two chimaeric proteins carrying the transit peptide of either ferredoxin or plastocyanin linked to the mouse cytosolic enzyme dihydrofolate reductase (EC 1.5.1.3.) were employed. In contrast to observations in mitochondria, we found in chloroplasts that transport of a purified ferredoxin-dihydrofolate reductase fusion protein is not blocked by the presence of methotrexate, a folate analogue that stabilizes the structural conformation of dihydrofolate reductase. It is shown that transport competence of this protein in the presence of methotrexate is not a consequence of alteration of the folding characteristics or methotrexate binding properties of dihydrofolate reductase by fusion to the ferredoxin transit peptide. Binding of dihydrofolate reductase fusion proteins to chloroplast envelopes is not inhibited by low temperature and it is only partially diminished by methotrexate. It is demonstrated that the dihydrofolate reductase fusion proteins unfold, despite the presence of methotrexate, on binding to the chloroplast envelopes. We propose the existence of a strong protein unfolding activity associated to the chloroplast envelopes.     

籼,粳稻及其杂种粗线期的核型分析

研究了典型籼,粳稻及其杂种粗线期的核型,结果表明：（１）籼稻品种南特号具有两对随体染色体,分别为第十和第十二染色体,粳稻品种巴利拉具１对随体染色体,为第十染色体,杂种南特号／巴利拉的随体染色体与南特号相同;（２）在粗线期,两品种均可见单核仁和双核仁的细胞存在,说明细胞贩核仁数目与随体数目并不对应;（３）两品种的对应染色体之间,在染色体相对长度,臂比及染色粒分布上均无明显差异。     

RGDS肽对大鼠主动脉球囊内膜剥脱后血管壁增殖的影响

在大鼠主动脉球囊内膜剥脱术后血管壁细胞过度增殖模型上,用合成的血小板膜纤维蛋白原受体（ｇｌｙｃｏｐｒｏｔｅｉｎⅡｂ／Ⅲａｃｏｍｐｌｅｘ,ＧＰⅡｂ／Ⅲａ）拮抗剂ＲＧＤＳ（Ａｒｇ－Ｇｌｙ－Ａｓｐ－Ｓｅｒ,５０μｍｏｌ·ｋｇ－１·ｄ－１）治疗可有效地抑制损伤血管壁的细胞计数增加和内膜增厚以及血管平滑肌细胞增殖,显著降低其血管组织３Ｈ－ＴｄＲ和３Ｈ－Ｌｅｕ的参入增加程度。实验结果提示ＲＧＤＳ肽作为血管成型术的辅佐剂,对于防治血管再狭窄可能具有潜在的临床应用前景。     

茶叶中总硒含量及其影响因素的研究

茶叶中硒素总量测定结果表明：土壤含硒量的高低是直接影响茶叶中硒的总量。茶树根、茎、叶、果中均有硒元素,叶片是茶硒积累的主要器官,尤其是老叶,其含量是嫩叶的几倍．茶树品种间含量的差异显著,最大差异达10倍以上．毛茶加工的成品茶含硒量受加工技术措施影响较大,其不同等级的含硒量与级别没有线性关系．
