Stripe rust, leaf rust, and Barley Yellow Dwarf Virus (BYDV) are important diseases of barley (Hordeum vulgare L). Using 94 doubled-haploid lines (DH) from the cross of Shyri x Galena, multiple disease phenotype datasets, and a 99-marker linkage map, we determined the number, genome location, and effects of genes conferring resistance to these diseases. We also mapped Resistance Gene Analog Polymorphism (RGAP) loci, based on degenerate motifs of cloned disease resistance genes, in the same population. Leaf rust resistance was determined by a single gene on chromosome 1 (7H). QTLs on chromosomes 2 (2H), 3 (3H), 5 (1H), and 6 (6H) were the principal determinants of resistance to stripe rust. Two- locus QTL interactions were significant determinants of resistance to this disease. Resistance to the MAV and PAV serotypes of BYDV was determined by coincident QTLs on chromosomes 1 (7H), 4 (4H), and 5 (1H). QTL interactions were not significant for BYDV resistance. The associations of molecular markers with qualitative and quantitative disease resistance loci will be a useful information for marker-assisted selection. Exposure to acute stressors has been shown to impair cellular immunity in human beings and other animal species. Comparatively little is known, however, about the effects of long-term stressors on immune function and how individual behavioral characteristics may mediate differences in immune function and clinical disease susceptibility. To determine the effects of social stress on cellular immunity and reactivation of a latent herpesvirus, 20 Herpes B virus-positive male cynomolgus monkeys were exposed to four periodic reorganizations of social group memberships over 5 months. Observations were made to categorize individuals as high or low in expression of aggressive, fearful, and affiliative behaviors. Complete blood counts, lymphocyte proliferation tests, and natural killer cell cytotoxicity assays were performed immediately before and 4 days after reorganizations. Herpesvirus-specific immunoglobulin G antibody levels were measured, and oral and conjunctival swabs were cultured for virus. Reorganization was associated with increased lymphocyte counts (P = 0.0009) and decreased lymphocyte proliferation in response to phytohemagglutinin (P < 0.005), particularly among monkeys showing high levels of fear (P = 0.0137). High-aggressive monkeys showed lower baseline natural killer cell activity (P = 0.0013) and higher lymphocyte counts (P = 0.013) than low-aggressive monkeys. Herpesvirus antibody titers decreased over time (P < 0.004) and no positive virus cultures were obtained. Measures of cellular immunity and behavior were unrelated to virus-specific antibody titers. These results suggest that repeated exposure to a social stressor alters several measures of cellular immunity, and that some of these changes may be predicted by individual differences in agonistic behavior. In contrast to human studies, the results suggest that some psychological stressors may not cause reactivation of a common herpesvirus in this species. © 1996 Wiley-Liss, Inc.  相似文献

Abstract. Bogs, economically valuable wetlands, are subjected to exploitation in southern Canada. We addressed plant conservation within bogs mined for peat, in which small undisturbed remnants are left, mostly at the margins of the mined areas. The main goal of the study was to test whether these remnants act as refuges for plants which could recolonize areas that are planned for restoration after mining is completed. Mosses, lichens and vascular plants were sampled in remnants of 24 mined bogs in southeastern Canada during the summer of 1997. The vegetation was also sampled at the margins and centres of 24 nearby natural bogs in plots similar in size to these remnants. Using similarity analysis and ordination techniques, we found that plant species assemblages in remnants of mined bogs differ from those near the margins of natural bogs, and that certain species are associated with the centre of natural bogs, due to the presence of pools. We also showed that water conditions of remnants are affected by drainage due to peat mining. Sphagnum moss showed itself to be a key indicator of mining effects on vegetation. Implications for peat resource management and bog conservation are discussed.  相似文献

Questions: How does Calluna vulgaris regenerate after burning wet and dry heaths of different age? Location: Central Norway. Methods: Patches of Calluna dominated old and young wet heath and old dry heath were burned. The mean temperatures reached in the categories of heath were measured. Cover, frequency, height and number of seedlings of Calluna were studied in plots. Results: The mean burning temperatures varied from 680 to 740°C, and were highest in wet heath that had not been burned for at least 50 years. Regeneration of Calluna was evident in the year of burning, but only from seeds. Even heath with a short burning interval lacked vegetative regeneration. Calluna cover increased yearly and exceeded 50 % three years after burning. Regeneration was evident in the year of burning, and was highest in heath with a short burning interval. The height increased regularly at all sites, but was most rapid in dry heath, probably because of better microclimatic conditions. There were more seedlings in the heath with a short burning interval. Conclusions: Calluna only regenerates from seeds afterburning in central Norway. The density of Calluna increased rapidly, which is positive for the conservation and management of coastal heaths in central Norway.  相似文献

We investigated the dose-response effect of positive end-expiratory pressure (PEEP) and increased lung volume on the pulmonary clearance rate of aerosolized technetium-99m-labeled diethylenetriaminepentaacetic acid (99mTc-DTPA). Clearance of lung radioactivity was expressed as percent decrease per minute. Base-line clearance was measured while anesthetized sheep (n = 20) were ventilated with 0 cmH2O end-expiratory pressure. Clearance was remeasured during ventilation at 2.5, 5, 10, 15, or 20 cmH2O PEEP. Further studies showed stepwise increases in functional residual capacity (FRC) (P less than 0.05) measured at 0, 2.5, 5, 10, 15, and 20 cmH2O PEEP. At 2.5 cmH2O PEEP, the clearance rate was not different from that at base line (P less than 0.05), although FRC was increased from base line. Clearance rate increased progressively with increasing PEEP at 5, 10, and 15 cmH2O (P less than 0.05). Between 15 and 20 cmH2O PEEP, clearance rate was again unchanged, despite an increase in FRC. The pulmonary clearance of aerosolized 99mTc-DTPA shows a sigmoidal response to increasing FRC and PEEP, having both threshold and maximal effects. This relationship is most consistent with the hypothesis that alveolar epithelial permeability is increased by lung inflation.  相似文献

Receptor-mediated recognition and adhesion to laminin, a specific glycoprotein from basement membranes, exert an important role in many biological phenomena. Studying cell surface proteins of B16-F10, a metastatic murine melanoma cell line, we identified a 120-140 kDa glycoprotein (gp120/140) that binds laminin. This glycoprotein was recognized by a polyclonal antibody raised against the human fibronectin receptor beta 1-integrin chain, as well as immunoprecipitated by an anti-alpha 6 chain (monoclonal antibody GoH3), characterizing it as an alpha 6/beta 1-integrin. Its binding to laminin was specific and displayed moderate affinity, as its apparent dissociation constant was 18 nM. To characterize the influence of carbohydrate moieties on the laminin-gp120/140 interaction, metaperiodate oxidation, metabolic inhibition of glycosylation, and enzymatic deglycosylation studies were performed. Our results indicate that gp120/140 Asn-linked oligosaccharides play a part in this interaction. Reciprocally, both metaperiodate and N-glycanase treatment of native laminin reduced its binding to gp120/140, characterizing the latter as a lectin-like molecule. These results point to glycosylation processes as a possible mechanism for variable binding specificity profiles among integrins.  相似文献

Physiol.81(4): 1677-1682, 1996.The purpose of this study was tocompare the short-term strength and neural adaptations to eccentric andconcentric training at equal force levels. Forty-two sedentary women(age = 21.5 yr) were ranked based on the initial quadriceps strengthscore, and trios of subjects were randomly assigned to either aneccentric (n = 14), a concentric (n = 14), or a nonexercising controlgroup (n = 14). Training involved atotal of 824 eccentric or concentric quadriceps actions at 1.05 rad · s1administered in four sets of 6-10 repetitions, four times per weekfor 6 wk. Before and after training, all subjects were tested forunilateral maximal isometric and eccentric and concentric actions at1.05 rad · s1and for a 40-repetition eccentric and concentric fatigue series of theleft and right quadriceps. Surface electromyographic activity of thevastus lateralis and medialis was monitored during testing. Concentrictraining increased concentric (36%, P < 0.05), isometric (18%, P < 0.05), and eccentric strength (13%), and eccentric training increasedeccentric (42%, P < 0.05),isometric (30%, P < 0.05), andconcentric (13%) strength. Eccentric training improved eccentric andisometric strength more (P < 0.05)than did concentric training. The electromyographic adaptations weregreater with eccentric training. Cross-education was 6%, and neithertraining mode modified fatigability. The data suggest that training ofthe quadriceps muscle with submaximal eccentric actions brings aboutgreater strength adaptations faster than does training withmaximal-level concentric actions in women. This greater adaptation islikely to be mediated by both mechanical and neural factors.

 Genes cloned from diverse plants for resistance to different pathogens have sequence similarities in domains presumably involved in pathogen recognition and signal transduction in triggering the defense response. Primers based on the conserved regions of resistance genes often amplify multiple fragments that may not be separable in an agarose gel. We used denaturing polyacrylamide-gel electrophoresis to detect PCR products of plant genomic DNA amplified with primers based on conserved regions of resistance genes. Depending upon the primer pairs used, 30–130 bands were detected in wheat, rice, and barley. As high as 47%, 40%, and 27% of the polymorphic bands were detected in rice, barley, and wheat, respectively, and as high as 12.5% of the polymorphic bands were detected by certain primers in progeny from a cross of the wheat cultivars ‘Stephens’ and ‘Michigan Amber’. Using F6 recombinant inbred lines from the ‘Stephens’בMichigan Amber’ cross, we demonstrated that polymorphic bands amplified with primers based on leucine-rich repeats, nucleotide-binding sites and protein kinase genes, were inherited as single loci. Linkages between molecular markers and stripe rust resistance genes were detected. This technique provides a new way to develop molecular markers for assessing the genetic diversity of germplasm based upon potential candidate resistance genes in diverse species. This study examined (i) the effect of artificially created microtopography and straw mulch on the soil moisture and (ii) energy balance and the establishment of a Sphagnum cover on a cutover peatland. Straw mulch caused rainfall interception approaching 2 mm per event. Although interception represented 44% of the total rainfall over the measurement period, water that evaporated from the mulch used energy that would otherwise have been used to evaporate soil water. Thus, the net effect of interception by mulch was negligible. The soil heat flux below the mulch was only 13% of the bare soil value and was decoupled from the daily net radiation. Net radiation over the bare soil was 15% greater than over the mulch. However, because of the greater heat flux into the bare peat, the energy available for sensible and latent heat fluxes was similar between the mulch covered and bare peat. Average evaporation from mulch and bare soil was estimated to be 2.6 and 3.1 mm d−1, respectively. Soil water tension 1 cm below the surface remained above −100 cm (mb) all season (100% of the time) when a mulch was used, compared to only 30% of the time in the bare soil. Correspondingly, the water table was sustained above the 40 cm depth, 60% of time in the mulch covered site, compared to only 40% of the time in the bare peat site. Negative relief elements of the microtopography were wetter and cooler than positive relief elements. However, when under a mulch, the negative relief elements provided no additional benefit, in terms of temperature or soil moisture amelioration. The control site with a mulch cover was equivalent or better than negative relief elements with a mulch cover. Taking into account the poorer performance of positive relief elements, even when mulch covered, the creation of surface microtopography reduced the overall moisture content of the site. Sphagnum established and spread only when the diaspores were protected with a straw mulch. All microtopography types tested had no effect on the establishment of Sphagnum mosses when the microtopography treatments, including positive and negative relief elements, were treated as a whole, although being in a depression helped Sphagnum establishment.  相似文献

Wild strawberry (Fragaria vesca L.) is an attactive model system for studying ripening in non-climacteric fruit, because of its small diploid genome, its short reproductive cycle, and its capacity for transformation. We have isolated eight ripening-induced cDNAs from this species after differential screening of a cDNA library. The predicted polypeptides of seven of the clones exhibit similarity to database protein sequences, including acyl carrier protein, caffeoyl- CoA 3-O-methyltransferase, sesquiterpene cyclase, major latex protein, cystathionine -synthase, dehydrin and an auxin- induced gene. A ninth cDNA clone that was constitutively expressed is predicted to encode a metallothionein-like protein. None of these proteins appear to be directly related to events generally associated with ripening such as cell wall metabolism or the accumulation of sugars and pigments, rather, their putative functions are indicative of the wide range of processes upregulated during fruit ripening.  相似文献
