Dorycnium hirsutum (L.) Ser. and Dorycnium rectum (L.) Ser. are Mediterranean perennial legumes that may have potential as alternative forage plants to Medicago sativa (lucerne, alfalfa) for low rainfall dryland agriculture. Strategies for surviving periods of water deficit are vital for perennial plants in water-limited environments. This experiment compared leaf physiological and morphological adaptations to increasing water deficit among D. hirsutum, D. rectum and M. sativa. Plants were grown in the glasshouse in large pots (7.8 L, 1 m deep, 10 cm diameter) containing a sandy clay loam (14% available water content) to limit differences between root foraging among the species. Watering was withheld for 21 days and predawn and midday leaf water and osmotic potential were determined. Mid-morning rates of gas exchange were measured at five times as soil water was depleted. After 35 days of withholding water, plant recovery was measured. D. hirsutum and M. sativa reduced stomatal conductance at leaf water potentials below −1.8 MPa and water-stressed D. hirsutum osmotically adjusted by up to 0.68 MPa. D. rectum differed from the other species; leaf water potential was maintained at high levels until soil water content had reached low levels, and reductions in stomatal conductance and photosynthesis were not associated with leaf water potential. D. hirsutum and M. sativa displayed leaf morphological adaptations that may contribute to greater resistance of water deficit. Only one of five D. rectum plants survived the water-stress treatment compared to five of five for D. hirsutum and four of five for M. sativa. The water relations and physiology of D. hirsutum observed in this study suggest that it possesses adaptations suitable for arid environments. On the other hand, the poor survival and water relations of D. rectum indicate that it is poorly adapted to situations where water deficit is common.  相似文献

Bovine abortions caused by the apicomplexan parasite Neospora caninum have been responsible for severe economic losses to the cattle industry. Infected cows either experience abortion or transmit the parasite transplacentally at a rate of up to 95%. Neospora caninum vaccines that can prevent vertical transmission and ensure disruption in the life cycle of the parasite greatly aid in the management of neosporosis in the cattle industry. Brucella abortus strain RB51, a commercially available vaccine for bovine brucellosis, can also be used as a vector to express plasmid-encoded proteins from other pathogens. Neospora caninum protective antigens MIC1, MIC3, GRA2, GRA6 and SRS2 were expressed in strain RB51. Female C57BL/6 mice were vaccinated with a recombinant strain RB51 expressing N. caninum antigen or irradiated tachyzoites, boosted 4 weeks later and then bred. Antigen-specific IgG, IFN-gamma and IL-10 were detected in vaccinated pregnant mice. Vaccinated mice were challenged with 5 x 10(6)N. caninum tachyzoites between days 11-13 of pregnancy. Brain tissue was collected from pups 3 weeks after birth and examined for the presence of N. caninum by real-time PCR. The RB51-MIC3, RB51-GRA6, irradiated tachyzoite vaccine, pooled strain RB51-Neospora vaccine, RB51-MIC1 and RB51-SRS2 vaccines elicited approximately 6-38% protection against vertical transmission. However, the differences in parasite burden in brain tissue of pups from the control and vaccinated groups were highly significant for all groups. Thus, B. abortus strain RB51 expressing the specific N. caninum antigens induced substantial protection against vertical transmission of N. caninum in mice.  相似文献

The MukBFE complex is essential for chromosome segregation and condensation in Escherichia coli. MukB is functionally related to the structural maintenance of chromosomes (SMC) proteins. Similar to SMCs, MukB requires accessory proteins (MukE and MukF) to form a functional complex for DNA segregation. MukF is a member of the kleisin family, which includes proteins that commonly mediate the interaction between SMCs and other accessory proteins, suggesting that the similarities between the MukBFE and the SMC complexes extend beyond MukB. Although SMCs have been carefully studied, little is known about the roles of their accessory components. In the present work, we characterize the oligomeric states of MukE and MukF using size exclusion chromatography and analytical ultracentrifugation. MukE self-associates to form dimers (K(D) 18 +/- 3 mum), which in turn interact with the MukF dimer to form two distinct high affinity complexes having 2:2 and 2:4 stoichiometries (F:E). Intermediate complexes are not found, and thus we propose that the equilibrium between these two complexes determines the formation of a functional MukBFE with stoichiometry 2:2:2.  相似文献

Mnemiopsis leidyi: larvae depend on microplankton (<200 µm) prey duringthe first few days following hatching until larvae are >0.5mm in length and can successfully capture and consume mesozooplanktonprey. Feeding and growth rates of newly hatched M. leidyi larvaewere measured in controlled laboratory experiments. When fednatural microplankton assemblages, newly hatched larvae consumedsignificant quantities of both autotrophic and heterotrophicprey, including diatoms, phototrophic, heterotrophic and mixotrophicdinoflagellates, euglenoid flagellates, aloricate and tintinnidciliates, and rotifers. Average per capita clearance rates were1.99–7.59 mL individual–1 h–1 ( = 4.01 mL individual–1 h–1; SD = 1.95)and total per capita ingestion was 0.01–4.70 µgC individual–1 day–1 x 102 ( = 0.83 µg C individual–1 day–1 x 102; SD =1.89). Larval growth rates were –0.13 to 0.56 mm individual–1day–1 (equivalent to –1.72 to 4.33 µg C individual–1day–1) over a range of larval sizes from 0.5 (<0.5µg C) to 5 mm (85 µg C). A diet consisting entirelyof microplankton prey supported larval growth for >2 weeks,and growth rate decreased when larvae reached 4–5 mm inlength, corresponding to the beginning of their morphologicaltransition from tentaculate to lobate feeding mode. The grossgrowth efficiency of larvae fed natural microplankton assemblageswas 3%.  相似文献

Humans and chimpanzees share >99% identity in most proteins. One rare difference is a human-specific inactivating deletion in the CMAH gene, which determines biosynthesis of the sialic acid N-glycolylneuraminic acid (Neu5Gc). Since Neu5Gc is prominent on most chimpanzee cell surfaces, this mutation could have affected multiple systems. However, Neu5Gc is found in human cancers and fetuses and in trace amounts in normal human tissues, suggesting an alternate biosynthetic pathway. We inactivated the mouse Cmah gene and studied the in vivo consequences. There was no evidence for an alternate pathway in normal, fetal, or malignant tissue. Rather, null fetuses accumulated Neu5Gc from heterozygous mothers and dietary Neu5Gc was incorporated into oncogene-induced tumors. As with humans, there were accumulation of the precursor N-acetylneuraminic acid and increases in sialic acid O acetylation. Null mice showed other abnormalities reminiscent of the human condition. Adult mice showed a diminished acoustic startle response and required higher acoustic stimuli to increase responses above the baseline level. In this regard, histological abnormalities of the inner ear occurred in older mice, which had impaired hearing. Adult animals also showed delayed skin wound healing. Loss of Neu5Gc in hominid ancestors approximately 2 to 3 million years ago likely had immediate and long-term consequences for human biology.  相似文献

The adult mammalian cochlea receives dual afferent innervation: the inner sensory hair cells are innervated exclusively by type I spiral ganglion neurons (SGN), whereas the sensory outer hair cells are innervated by type II SGN. We have characterized the spatiotemporal reorganization of the dual afferent innervation pattern as it is established in the developing mouse cochlea. This reorganization occurs during the first postnatal week just before the onset of hearing. Our data reveal three distinct phases in the development of the afferent innervation of the organ of Corti: (1) neurite growth and extension of both classes of afferents to all hair cells (E18-P0); (2) neurite refinement, with formation of the outer spiral bundles innervating outer hair cells (P0-P3); (3) neurite retraction and synaptic pruning to eliminate type I SGN innervation of outer hair cells, while retaining their innervation of inner hair cells (P3-P6). The characterization of this developmental innervation pattern was made possible by the finding that tetramethylrhodamine-conjugated dextran (TMRD) specifically labeled type I SGN. Peripherin and choline-acetyltransferase immunofluorescence confirmed the type II and efferent innervation patterns, respectively, and verified the specificity of the type I SGN neurites labeled by TMRD. These findings define the precise spatiotemporal neurite reorganization of the two afferent nerve fiber populations in the cochlea, which is crucial for auditory neurotransmission. This reorganization also establishes the cochlea as a model system for studying CNS synapse development, plasticity and elimination.  相似文献
