Behavioural observations of both captive and free-ranging sand and mountain gazelles revealed differences in group size and composition, territory size and agonistic behaviour. Sand gazelles are seasonal breeders and form large herds (maximum = 56) during rut and post-rut as compared to mountain gazelles, which breed throughout the year in smaller groups (maximum = 5). As a result, mountain gazelle males maintain their territories year round, whereas sand gazelle rams show marked morphological changes and defend a territory only during the rut. Mountain gazelle males exhibited threat displays rather than engaging in actual combat more often than did sand gazelles. Although subject to the same environmental conditions, sand gazelles are seasonal breeders and the young are born in March-April, whereas mountain gazelles give birth throughout the year.  相似文献

Studies were conducted to determine factors that influence the survival of bradyzoites of Neospora caninum within tissue cysts in the brains of experimentally inoculated mice. Viable tissue cysts were detected in the brains of mice inoculated 13 mo previously with either of 2 isolates (NC-1 or NC-2) of N. caninum. Bradyzoites within tissue cysts of the NC-2 isolate survived for at least 14 days at 4 C in refrigerated brain homogenates. Bradyzoites within tissue cysts of the NC-2 isolate also survived in the intact brain of a mouse carcass refrigerated at 4 C for 7 days. Bradyzoites within tissue cysts of the NC-3 isolate were killed by freezing at -20 C for 1 day.  相似文献

We have identified and cloned a gene, ES2, encoding a putative 476 amino acid protein with a predicted M r of 52,568. The gene is localized within the DiGeorge/Velocardiofacial syndrome locus on 22q11.2 and is deleted in all the patients in which a deletion within 22q11 could be demonstrated, with the exception of one patient. ES2 is expressed in all the tissues studied. Sequence comparison showed identity with five ESTs and at the amino acid level the sequence was highly similar to, and collinear with, a hypothetical C. elegans protein of unknown function. Mutation analysis was performed in 16 patients without deletion, but no mutation has been found. The cDNA sequence is conserved in mouse and is localized on MMU16B1-B3, known to contain a syntenic group in common with HSA 22q11.2. Received: 25 March 1996 / Accepted: 15 May 1996  相似文献

Abstract The effectiveness of DNA immunization has been demonstrated in several model systems, usually following intramuscular injection of DNA in saline, or topical administration to the skin. In this study we have compared DNA delivered by three routes (intramuscular, intravenous, and intraperitoneal) and, for each route, in two vehicles (cationic liposome complex and pH sensitive liposome). These two lipid vehicles were evaluated because they are frequently used in gene therapy studies, but their immunogenicity has not been extensively studied. Each of these six combinations has been evaluated not only by assay of marker gene expression in a variety of tissues, but also by measurement of biologically-relevant parameters of immunity induction of antibodies, cytotoxic T lymphocytes, and protection against viral challenge. By both criteria (marker gene expression and induced immunity), the outcomes vary markedly among the six combinations. The combination leading to maximal marker gene expression (DNA with cationic lipid, administered i.v.) also induces detectable antibodies and CTL, and is the only one of the six combinations to induce immune responses comparable to those seen following i.m. injection of DNA in saline. However, marker gene expression can be detected in other combinations in the absence of induced immunity thus the value of marker gene expression in predicting the protection induced by a microbial antigen is questionable suggesting that, when evaluating various promoter constructs, marker gene expression may not adequately replace the direct measurement of biological outcomes.  相似文献

The cytochromes P-450 (P-450s) constitute an extremely large family ('superfamily') of haemoproteins that catalyse the oxidation of a wide range of physiological and non-physiological compounds. A remarkable feature of the P-450s is the manipulation of the same basic structure and chemistry to achieve an enormous range of functions in organisms as diverse as bacteria and man. Indeed, the P-450s have been described as ‘the most versatile biological catalyst known’. Much research is focussed on mammalian P-450s, with their roles in such processes as steroid transformations and the metabolism of carcinogens and other xenobiotics. However, our knowledge of the structure and function of the P-450s has been advanced by analysis of a limited number of its bacterial members, primarily P-450cam from Pseudomonas putida. Four P-450 structures have been solved to date, all of which are from bacterial sources. The aim of this review is to assess current knowledge of the many bacterial P-450s, with emphasis on their diverse biological roles and on the advances in our knowledge of this extremely important enzyme class, which have been made feasible through their study.  相似文献

We have constructed a comparative map in mouse of the critical region of human 22q11 deleted in DiGeorge (DGS) and Velocardiofacial (VCFS) syndromes. The map includes 11 genes potentially haploinsufficient in these deletion syndromes. We have localized all the conserved genes to mouse Chromosome (Chr) 16, bands B1-B3. The determination of gene order shows the presence of two regions (distal and proximal), containing two groups of conserved genes. The gene order in the two regions is not completely conserved; only in the proximal group is the gene order identical to human. In the distal group the gene order is inverted. These two regions are separated by a DNA segment containing at least one gene which, in the human DGS region, is the most proximal of the known deleted genes. In addition, the gene order within the distal group of genes is inverted relative to the human gene order. Furthermore, a clathrin heavy chain-like gene was not found in the mouse genome by DNA hybridization, indicating that there is an inconsistent level of gene conservation in the region. These and other independent data obtained in our laboratory clearly show a complex evolutionary history of the DGS-VCFS region. Summary The NPK concentrations in cowpea leaves (cv California Blackeye No. 5) with stage of plant development did not vary significantly with tillage treatment but several features of the concentration of these different ions with age, stage of plant development, incidence of flooding and soil water content are significant.In the major rainy season when soil water content was adequate and seldom above field capacity, there was an almost linear increase in the concentration of N and P in the leaves up to 56 days after planting which coincided with fruit development. This suggested that the accumulation of N and P do take place after flowering which occurred 39 days after planting. The pattern of accumulation of N and P in the minor rainy season suggested that flooding and the occurrence of prolonged soil moisture contents above field capacity suppressed the accumulation of these two nutrients. Potassium accumulation was high in the early stages of cowpea growth and declined with age of the crop. Flooding and high soil moisture contents seem to have had little or no effect on potassium accumulation in the leaves. The pattern of accumulation of N and P suggests that the supply of these nutrients to the cowpea plant must remain high up to fruit development and that additional N and P would need to be added to the soil after a flooding event or when the soil moisture content is above field capacity for prolonged periods.  相似文献

The responses of oxonol dyes to single and multiple single turnovers of the photosynthetic apparatus of photosynthetic bacteria have been studied, and compared with the responses of the endogenous carotenoid pigments. The absorbance changes of the oxonols can be conveniently measured at 587 nm, because this is an isosbestic point in the ‘light-minus-dark’ difference spectrum of the chromatophores.The oxonols appear to respond to the light-induced ‘energization’ by shifting their absorption maxima. In the presence of K+, valinomycin abolished and nigericin enhanced such shifts, suggesting that the dyes respond to the light-induced membrane potential. Since the dyes are anions at neutral pH values, they probably distribute across the membrane in accordance with the potential, which is positive inside the chromatophores. The accumulation of dye, which is indicated by a decrease in the carotenoid bandshift, poises the dye-membrane equilibrium in favor of increased dye binding and this might be the cause of the spectral shift.The dye response has an apparent second-order rate constant of approx. 2 · 106 M?1 · s?1 and so is always slower than the carotenoid bandshift. Thus the dyes cannot be used to monitor membrane potential on submillisecond timescales. Nevertheless, on a timescale of seconds the logarithm of the absorbance change at 587 nm is linear with respect to the membrane potential calibrated with the carotenoid bandshift. This suggests that under appropriate conditions the dyes can be used with confidence as indicators of membrane potential in energy-transducing membranes that do not posses intrinsic probes of potential.  相似文献

Three activated carbon filters for point-of-use water treatment were tested in laboratory and field studies for chemical removal and microbiological effects on water. All removed free available chlorine in municipally treated water to below the limit of detection, but removed only about 50 to 70% of the total available chlorine and 4 to 33% of the total organic carbon. Standard plate count bacteria in the effluent increased steadily with time for 3 weeks and remained elevated over the 8-week period of the study. Total coliform bacteria were found to persist and proliferate on the filters for several days after transient contamination of the influent water. Silver-containing activated carbon filters suppressed total coliform but not total bacterial growth. Pseudomonas aeruginosa was recovered from the effluents of all filters at some time during the tests.  相似文献

The tensile strength of the cell walls ofBacillus megaterium andBacillus stearothermophilus was found to be about 2.4×107 N/m2. The internal pressure and water activity of the cells were 14 atm, 0.99 aw forB. megaterium and 28 atm, 0.98 aw forB. stearothermophilus. The greater strength ofB. stearothermophilus cells, considered as pressure vessels, restricts absorption of water by the protoplasm so that the water content on a dry weight basis is 3.4 g/g forB. megaterium cells in water but only 1.8 g/g forB. stearothermophilus.  相似文献
