The distribution of the refractive index (RI) of the protoplasm in sea urchin eggs was determined from the optical path differences at various regions of the cell measured by interference microscopy assuming that the cell structure is symmetrical about the line passing through the center of the cell and that of the nucleus in unfertilized eggs and about the spindle axis in fertilized eggs during mitosis and cleavage. The RI of the cytoplasm in the unfertilized egg was uniform except for the cortical region, which had the RI higher than that of the underlying endoplasm. The RI of the cortex was generally higher than that of the underlying endoplasm, which did not appreciably change during mitosis and cleavage. The RI of the nucleus was lower than that of the cytoplasm. The RI of the mitotic apparatus was lower than that of the surrounding cytoplasm. The fertilization membrane had a thickness of about 0.6 μm in hydrated state and about 25 nm in dried state (mean values). The RI of the perivitelline space was about 0.00015 higher than that of seawater, equivalent to 0.08 g/100 ml of contents.  相似文献

Evidence has been presented suggesting that females are significantly more susceptible to alcohol-induced liver damage (ALD) than males. In the current study, we examined sexual dimorphism in hepatic pathology, metabolism and cytokine profiles using two different rat models of ALD. Male and female Sprague-Dawley or Wistar rats were fed ethanol-containing low-carbohydrate liquid diets using oral or intragastric methods for 42 or 60 days. In both models, ethanol treatment produced similar significant liver hyperplasia accompanied by increases in plasma ALT, steatosis, inflammation and necrosis (p < 0.05). Greater pathology scores were observed in the intragastrically infused rats. Males did not differ significantly from females in serum ALT values or pathology despite greater elevations in TNFalpha and IL-1beta mRNAs in ethanol-treated female rat livers (p < 0.05). Furthermore, there was no sexual dimorphism in blood ethanol concentrations or CYP2E1-induction even though sexually dimorphic alterations in other hepatic cytochrome P450 enzymes were observed. These data do not support previous observations that female rats have a greater susceptibility to ethanol-induced hepatotoxicity than males.  相似文献

The intragastric administration of ethanol as part of a low-carbohydrate diet results in alcohol hepatotoxicity. We aimed to investigate whether comparable liver injury can be achieved by oral diet intake. Male Sprague-Dawley rats were fed ethanol as part of low-carbohydrate diets for 36-42 days either intragastrically or orally. Liver pathology, blood ethanol concentration, serum alanine amino transferase (ALT), endotoxin level, hepatic CYP2E1 induction, and cytokine profiles were assessed. Both oral and intragastric low-carbohydrate ethanol diets resulted in marked steatosis with additional inflammation and necrosis accompanied by significantly increased serum ALT, high levels of CYP2E1 expression, and production of auto-antibodies against malondialdehyde and hydroxyethyl free radical protein adducts. However, cytokine profiles differed substantially between the groups, with significantly lower mRNA expression of the anti-inflammatory cytokine interleukin 4 observed in rats fed low-carbohydrate diets orally. Inflammation and necrosis were significantly greater in rats receiving low-carbohydrate alcohol diets intragastrically than orally. This was associated with a significant increase in liver tumor necrosis factor alpha and interleukin 1beta gene expression in the intragastric model. Thus, oral low-carbohydrate diets produce more ethanol-induced liver pathology than oral high-carbohydrate diets, but hepatotoxicity is more severe when a low-carbohydrate diet plus ethanol is infused intragastrically and is accompanied by significant increases in levels of proinflammatory cytokines.  相似文献

An extract of cultured Agroxieinma githago L. cells was found to show potent inhibitory activity against plans virus infection. The effects of cultural conditions on the growth of the cell suspension and on the production of the inhibitor were examined. Since the production of substance was dependent on growth. experiments were made to improve growth. The optimum temperature was 26 to 30°C and optimum pH of the medium before autoclaving was between 5 and 7. In a medium of higher osmotic pressure, the water content of the cultured cells was lowered markedly. The growth rate in a small volume of the medium was higher than that in a larger volume at an early stage of the cultivation, but it was not changed by different inoculum sizes. The cells required thiamine and 2,4-D for growth but no other vitamins or growth regulators. The optimum level of 2,4-D was 0.1 mg/l. Higher sucrose concentration in the medium gave higher production of cell mass and of the inhibitor. However, 3% of sucrose was selected as the most economical concentration. For normal cell growth, the presence of both NH4NO3 and KNO3 as nitrogen sources was required. The use of a single nitrogen source caused a long lag period or inhibition of the cell growth. KH2PO4 stimulated the growth when in was used in the level of 2.5 to 5 mM. The cell adhesion on the surface of the fermentor sometimes causes trouble in a large-scale cultivation. It was found that reducing the Ca2+ level in the medium prevented the cell adhesion and foaming remarkably. Based on the results obtained, a modified medium was established which was excellent for shortening the culture period and for efficient production of the anti-plant virus inhibitor.  相似文献

Intact rat liver cells from the perivenous region were isolated by collagenase perfusion after first destroying the periportal region by a brief portal infusion of digitonin. Periportal cells were isolated after retrograde digitonin infusion. Significantly higher alanine aminotransferase, gamma-glutamyltransferase and lactate dehydrogenase activities and lower glutamate dehydrogenase and pyruvate kinase activities in periportal than in perivenous cells demonstrate marked separation. The high yield allows further characterization in vitro of the cell populations.  相似文献
