An isolated, yet virtually intact contour feather (FUM‐1980) from the lower Eocene Fur Formation of Denmark was analysed using multiple imaging and molecular techniques, including field emission gun scanning electron microscopy (FEG‐SEM), X‐ray absorption spectroscopy and time‐of‐flight secondary ion mass spectrometry (ToF‐SIMS). Additionally, synchrotron radiation X‐ray tomographic microscopy (SRXTM) was employed in order to produce a digital reconstruction of the fossil. Under FEG‐SEM, the proximal, plumulaceous part of the feather revealed masses of ovoid microstructures, about 1.7 μm long and 0.5 μm wide. Microbodies in the distal, pennaceous portion were substantially smaller (averaging 0.9 × 0.2 μm), highly elongate, and more densely packed. Generally, the microbodies in both the plumulaceous and pennaceous segments were aligned along the barbs and located within shallow depressions on the exposed surfaces. Biomarkers consistent with animal eumelanins were co‐localized with the microstructures, to suggest that they represent remnant eumelanosomes (i.e. eumelanin‐housing cellular organelles). Additionally, ToF‐SIMS analysis revealed the presence of sulfur‐containing organics – potentially indicative of pheomelanins – associated with eumelanin‐like compounds. However, since there was no correlation between melanosome morphology and sulfur content, we conclude these molecular structures derive from diagenetically incorporated sulfur rather than pheomelanin. Melanosomes corresponding roughly in both size and morphology with those in the proximal part of FUM‐1980 are known from contour feathers of extant parrots (Psittaciformes), an avian clade that has previously been reported from the Fur Formation. 相似文献

The tumour microenvironment and genetic alterations collectively influence drug efficacy in cancer, but current evidence is limited and systematic analyses are lacking. Using chronic lymphocytic leukaemia (CLL) as a model disease, we investigated the influence of 17 microenvironmental stimuli on 12 drugs in 192 genetically characterised patient samples. Based on microenvironmental response, we identified four subgroups with distinct clinical outcomes beyond known prognostic markers. Response to multiple microenvironmental stimuli was amplified in trisomy 12 samples. Trisomy 12 was associated with a distinct epigenetic signature. Bromodomain inhibition reversed this epigenetic profile and could be used to target microenvironmental signalling in trisomy 12 CLL. We quantified the impact of microenvironmental stimuli on drug response and their dependence on genetic alterations, identifying interleukin 4 (IL4) and Toll‐like receptor (TLR) stimulation as the strongest actuators of drug resistance. IL4 and TLR signalling activity was increased in CLL‐infiltrated lymph nodes compared with healthy samples. High IL4 activity correlated with faster disease progression. The publicly available dataset can facilitate the investigation of cell‐extrinsic mechanisms of drug resistance and disease progression. 相似文献

We are reporting a male patient who suffered from chronic granulomatous disease associated with cytochrome b−245 deficiency and McLeod red cell phenotype, Duchenne muscular dystrophy, and retinitis pigmentosa. On cytogenetic analysis, he seemed to have a very subtle interstitial deletion of part of band Xp21. Since it was impossible to know whether this material was truly deleted or inserted elsewhere in the genome, somatic cell and molecular studies were carried out. In somatic cell hybrids, the deleted X chromosome was isolated on a Chinese hamster background. Southern blot analysis with 20 single-copy probes, that had been mapped to the X short arm, led to the discovery of one (probe 754) that is missing from this patient's X chromosome and also from his total DNA. This proves that he, indeed, has a deletion rather than a balanced insertion. The results provide cytological mapping information for the X-linked phenotypes present in this patient. Furthermore, probe 754 recognizes a restriction fragment length polymorphism of high frequency that makes it the most powerful probe currently available for linkage studies with X-linked muscular dystrophy. 相似文献

Groundwater plumes containing dissolved uranium at levels above natural background exist adjacent to uranium ore bodies, at uranium mines, milling locations, and at a number of explosive test facilities. Public health concerns require that some assessment of the potential for further plume movement in the future be made. Reaction-transport models, which might conceivably be used to predict plume movement, require extensive data inputs that are often uncertain. Many of the site-specific inputs are physical parameters that can vary spatially and with time. Limitations in data availability and accuracy means that reaction-transport predictions can rarely provide more than order-of-magnitude bounding estimates of contaminant movement in the subsurface. A more direct means for establishing the limits of contaminant transport is to examine actual plumes to determine if, collectively, they spread and attenuate in a reasonably consistent and characteristic fashion. Here a number of U plumes from ore bodies and contaminated sites were critically examined to identify characteristics of U plume movement. The magnitude of the original contaminant source, the geologic setting, and the hydrologic regime were rarely similar from site to site. Plumes also spanned a vast range of ages, and no complete set of time-series plume analyses based on the spatial extent of U contamination exist for a particular site. Despite the accumulated uncertainties and variabilities, the plume data set gave a clear and reasonably consistent picture of U plume behavior. Specifically, uranium plumes: ??Appear to reach steady-state, that is, they quit spreading, rapidly (within a few years). ??Exceed roughly 2 km in length only in special cases (e.g., where in situ leaching has been carried out). The majority are much smaller. ??Exhibit very similar U chemistry between sites. This implies analogous contaminant attenuation mechanisms despite their location. 相似文献

Mouse monoclonal antibodies to carcinoembryogenic antigen (CEA) obtained by the somatic cell hybridization technique of K?hler and Milstein were used in a modified enzyme bridge immunoperoxidase staining method. Both high and low affinity antibodies were tested and their staining properties compared with those of a commercial polyvalent rabbit antiserum. The staining pattern of neoplastic epithelial cells in all seven antibodies in samples of primary adenocarcinoma of the colon was similar, indicating that no gross differences were found in the exposure of the different antigenic determinants of CEA in formalin fixed tissue. The background staining of the monoclonal antibodies are negligible. It is concluded that monoclonal antibodies are superior to conventional antisera in immunoperoxidase staining of CEA. 相似文献

Bulk seedlots of two unpedigreed multiprovenance seed production areas (SPAs) each of Eucalyptus camaldulensis and Eucalyptus tereticornis and one pedigreed seedling seed orchard (SSO) of E. tereticornis were planted in genetic gain trials at three southern Indian trial sites. At the time of seed collection, fewer than 30%
trees flowered in these orchards, except in one E. camaldulensis SPA where 73% of the trees flowered, which had an estimated outcrossing rate of 86%. The E. tereticornis SSO was dominated by pollen from five highly fecund families of the Indian Mysore gum land race, which contributed 59% of
the fruits produced. The SPA and SSO seedlots were compared with a bulked natural-provenance seedlot of E. camaldulensis (Morehead, Laura, and Kennedy Rivers, Queensland), another natural-provenance seedlot (Petford, Queensland), commercial eucalypt
clones at two sites, and a Mysore gum seedlot at one site. At 3 years, progeny from all the four SPAs displayed good survival
(79–93%) and performance similar to that of the natural provenances and the commercial clones. Progeny from the E. tereticornis SSO had significantly lower growth (at two sites) and lower survival at all three test sites. The Mysore gum seedlot displayed
high fecundity and lower growth but better survival than the SSO progeny. Seed orchard genetic composition and flowering contributions
thus affected progeny performance and the extent to which orchard genetic diversity was captured in the progeny. SPA progeny

Ants (Hymenoptera: Formicidae) are known to be a thermophilic group with fewer species at higher latitudes. They are ubiquitous in the sub‐boreal forests of west‐central British Columbia (Canada) in the early seral ages of forest succession, nesting almost exclusively in woody debris. Individual species begin to disappear with advancing seral ages as the canopy closes, suggesting the hypothesis that heat gain from insolation is critical to many species. We experimentally manipulated shading of two wood‐nesting ant species –Leptothorax muscorum (Nylander) and Formica aserva Forel – to emulate advancing seral age, in an environment documented to support their normal habitation. Shaded down woody debris (DWD) was found to be ca. 2 °C cooler than unshaded DWD, whereas stumps were ca. 1.5 °C cooler when shaded than when unshaded. We found significantly fewer colonies of both L. muscorum and F. aserva in shaded wood, as compared to unshaded, in a pattern consistent with how each species has been observed to respond to advancing seral age. Temperature monitoring during the experiment indicated that soil was cooler than either stumps or DWD, possibly explaining the absence of soil‐nesting ant species in these forests. The species‐specific differences in ant colony abundance in response to small temperature differences, found in this study, support other research on the significance of temperature in structuring ant assemblages. 相似文献
