Novel polymer substrates for SFM investigations of living cells, biological membranes, and proteins.
Extended studies were performed to prepare substrates for scanning force microscopy of biological samples with a surface roughness below 1 nm rms over an area of 500 x 500 nm2. The substrate smoothness and the lack of tip obscuring material are indispensable in order to visualize detailed structures of membrane surfaces, particularly when scanning the lamellipodia of mechanically sensitive goldfish glial cells where peripheral lamellipodia are only 20-30 nm in thickness. Appropriate substrates are poly(vinyl phenyl ketone) or furan polymers with corrugations of 0.3 and 0.15 nm rms, respectively, to which the growth-promoting protein laminin adsorbs directly with an acceptably increased roughness. Cells show normal growth behavior on these substrates and stick to the substrates in a stable fashion during several scans. Thus details of the membrane's surface may be resolved and are not obscured by the substrate texture. The uncoated furan polymer substrates are suitable for immobilization of membrane preparations such as purified membrane fragments containing bacteriorhodopsin or Na, K-ATPase and protein preparations such as antibodies. 相似文献

Numerous studies have addressed variation in the local occurrence and abundance of species, but only recently have investigators begun considering the influence of spatial context on community structure. We examined the influence of geographic position within a species’ range on its dominance (relative abundance) within the community. Our three primary questions were: 1) Does dominance within a community vary across a species’ range? 2) Do dominance‐location trends differ between widely and narrowly distributed species? and 3) Does dominance within a guild vary across a species range? We examined 40 passerine species (20 widely distributed, 20 narrowly distributed) and their communities using North American Breeding Bird Survey data, of which, a subset of 11 species belonging to a single foraging guild were used for addressing guild effects. Data were gathered from communities at the center (10 locations) and periphery (10 locations) of each species’ range. Focal species were significantly less dominant at locations on the periphery of their ranges compared to central locations, and did not differ between widely and narrowly distributed species. In examining factors influencing these patterns, it was found that narrowly distributed species occur in communities of fewer individuals and fewer species compared to widely distributed species irrespective of locale. Within the foraging guild, dominance was also lower at the periphery than at the center of focal species’ ranges. However, guild abundance was also lower at the periphery of ranges while guild richness did not vary with locale. Specifically, focal species abundance decreased at a greater rate than abundance of other members of the foraging guild. Contrary to overall community data, widely distributed species were more dominant members of their guild at the center of their ranges. The phosphatidylinositol 3-kinase (PI3K)/AKT pathway is frequently upregulated in human cancer. Activation of this pathway
has been reported to be associated with resistance to various chemotherapeutical agents. We here used a chemical biology/chemical
informatic approach to identify apoptotic mechanisms that are insensitive to activation of the PI3K/AKT pathway. The National
Cancer Institute (NCI) Mechanistic Set drug library was screened for agents that induce apoptosis in colon carcinoma cells
expressing a constitutively active form of AKT1. The cytotoxicity screening data available as self-organized maps at the Developmental
Therapeutics Program (DTP) of the NCI was then used to classify the identified compounds according to mechanism of action.
The results showed that drugs that interfere with the mitotic process induce apoptosis which is comparatively insensitive
to constitutive AKT1 activity. The conditional screening approach described here is expected to be useful for identifying
relationships between the state of activation of signaling pathways and sensitivity to anticancer agents. 相似文献

Magnetic circular dichroism (MCD) spectroscopy has been used to explore the connection between optical spectra and the high spin population of several hemoglobins under various conditions. It is found that the effectiveness of IHP in inducing spectral changes can be markedly affected by solvent. For example, the IHP-induced spectral changes in the visible region for nitritomethemoglobin-A in mixed buffer solvent systems (glycerol or polyethylene glycol (PEG), mw 190–210) are more than double those observed in aqueous buffers. We estimate that IHP induces a mix of R/T forms in bis-tris phosphate buffers, for NO2?metHb that is only about 50% T form. While PEG and glycerol both lead to enhanced IHP-induced spectral differences, they behave differently in two aspects. PEG shifts the visible MCD and absorption spectra of F?metHb-A. supposedly already biased towards the T form by ligand, in the same direction that IHP does. PEG also maximizes the spin state changes with IHP for three R form hemoglobins and N3?metHb-A, and so appears to stabilize the T form in all cases. Glycerol does not. In addition, the apparent binding constant for NO2? to H2OmetHb-A differs between these two solvents. Comparison of the data from several hemoglobins leads to the conclusion that the changes in spin state distributions induced by IHP correlate well with quarternary structure for a given hemoglobin. We continued our effort to make a comprehensive database (LISTA) for the yeast Saccharomyces cerevisiae. As in previous editions the genetic names are consistently associated to each sequence with a known and confirmed ORF. If necessary, synonyms are given in the case of allelic duplicated sequences. Although the first publication of a sequence gives-according to our rules-the genetic name of a gene, in some instances more commonly used names are given to avoid nomenclature problems and the use of ancient designations which are no longer used. In these cases the old designation is given as synonym. Thus sequences can be found either by the name or by synonyms given in LISTA. Each entry contains the genetic name, the mnemonic from the EMBL data bank, the codon bias, reference of the publication of the sequence, Chromosomal location as far as known, SWISSPROT and EMBL accession numbers. New entries will also contain the name from the systematic sequencing efforts. Since the release of LISTA4.1 we update the database continuously. To obtain more information on the included sequences, each entry has been screened against non-redundant nucleotide and protein data bank collections resulting in LISTA-HON and LISTA-HOP. This release includes reports from full Smith and Watermann peptide-level searches against a non-redundant protein sequence database. The LISTA data base can be linked to the associated data sets or to nucleotide and protein banks by the Sequence Retrieval System (SRS). The database is available by FTP and on World Wide Web. 相似文献

Cyto- and histological analysis of the whole immunogenic system at the development of an acute experimental appendicitis in rabbits has demonstrated that all the constituent parts of the system are involved into the reaction. In the central organ of immunity--thymus the main change is its progressive exhaustion, while peripheral organs of immunity are subjected to rather essential cytological rearrangements and activation of both cellular and humoral immunity. Accordingly to the type of the reaction, two groups of peripheral organs of immunity can be distinguish: the first--the appendix and the iliocecal lymph node which predominantly participate in the immunoblastic reaction and in the reaction of young plasmatic cells and demonstrate increased signs of immature cells emigration; the second--the axillary lymph node, as a representative of somatic nodes, and the spleen--the main carriers of the reaction in mature plasmic cells. Structural peculiarities of the complex appendix--iliocecal lymph node in intact animals and in the experiment make it possible to consider the complex as a peculiar functional link of the immune peripheral organs. 相似文献

An unusual Tc(III) boron-capped imine-oxime complex has been isolated from the reaction of 99TcCl3(CH3CN)(PPh3)2, dimethyl glyoxime (DMG) and ethyl boronic acid (EtB(OH)2). A single crystal X-ray structure analysis of this molecule 99TcCl(DMG)2(BDI)BEt (BDI=butane-2, 3-dione imine-oxime) shows it to be seven coordinate: TcClC14H25N6O5B, a=9.073(2), b=23.686(5), c=19.539(6) Å; β=93.77(2)°, P21/n, Z=8. Its structure is very similar to that of previously reported Tc(III) complexes 99TcCl(dioxime)3BR, except that one dioxime ligand on the molecule has been reduced to an imineoxime. 相似文献

Human clinical and psychophysical observations suggest that the taste
system is able to compensate for losses in peripheral nerve input, since
patients do not commonly report decrements in whole mouth taste following
chorda tympani nerve damage or anesthesia. Indeed, neurophysiological data
from the rat nucleus of the solitary tract (NST) suggests that a release of
inhibition (disinhibition) may occur centrally following chorda tympani
nerve anesthesia. Our purpose was to study this possibility further. We
recorded from 59 multi- and single- unit taste-responsive sites in the rat
NST before, during and after recovery from chorda tympani nerve anesthesia.
During anesthesia, average anterior tongue responses were eliminated but no
compensatory increases in palatal or posterior tongue responses were
observed. However, six individual sites displayed increased taste
responsiveness during anesthesia. The average increase was 32.9%.
Therefore, disinhibition of taste responses was observed, but infrequently
and to a small degree in the NST At a subset of sites, chorda
tympani-mediated responses decreased while greater superficial
petrosal-mediated responses remained the same during anesthesia. Since this
effect was accompanied by a decrease in spontaneous activity, we propose
that taste compensation may result in part by a change in signal-to-noise
ratio at a subset of sites.
