Recombinant histidine-tagged v-Ha-ras (his-ras) was purified to homogeneity from extracts ofE. coli M15 using a one-step procedure which involved immobilised metal ion chromatography on Ni2+-nitriloacetic acid agarose (Ni-NTA). The optimal pH for elution by imidazole was 6.6 and the yield of his-ras protein (greater than 95% pure) was about 4 mg/litreE. coli culture. Chromatography of a mixture of purified his-ras and rat brain cytosol on Ni-NTA together with SDS-PAGE and silver staining of proteins were employed to search forras-binding proteins present in rat brain cytosol. Chromatography of rat brain cytosol alone on Ni-NTA revealed several protein species which were not readily eluted with imidazole. These are likely to be low-abundance brain metal ion binding proteins. Pre-treatment of rat brain cytosol with Ni-NTA before a second round of chromatography on Ni-NTA removed most of these proteins. Chromatography of a mixture of pre-treated rat brain cytosol and purified his-ras protein revealed four new protein bands with molecular weights of 250, 90, 80 and 70 kDa. These were considered to be candidateras-binding proteins. Summary The temperatures at which chlorophyll fluorescence yield is substantially increased and the temperatures at which the quantum yield for CO2 uptake is irreversibly inhibited were measured for three shortgrass prairie species. The experimental taxa include, a cool season species (Agropyron smithii), a warm season species (Bouteloua gracilis), and a species which grows throughout the cool and warm seasons (Carex stenophylla). Agropyron smithii exhibited lower high temperature damage thresholds (43°C in cool grown plants, 46°C in warm grown plants), relative to the other two species. Bouteloua gracilis exhibited the highest tolerance to high temperature, with threshold values being 44–49°C for cool grown plants and 53–55°C for warm grown plants. Carex stenophylla exhibited threshold values which were intermediate to the other two species (43–47°C for cool grown plants, and 51–53°C for warm grown plants). Seasonal patterns in the fluorescence rise temperatures of field grown plants indicated acclimation to increased temperatures in all three species. The results demonstrate a correlation between the high temperature thresholds for damage to the photosynthetic apparatus, and in situ seasonal phenology patterns for the three species.  相似文献

Summary Plant population dynamics and life cycle characteristics are presented for two genetically related, yet ecologically distinct, chromosomal races of Machaeranthera gracilis (Nutt.) Shinners. Seedling mortality was very high for a field population of the desert race (n=2) as only 8% of the maximum number of seedlings produced did survive to the period of peak reproduction. Lower mortality occurred in the foothills race (n=4) as 15% of the maximum number of seedlings produced did survive until the period of peak reproduction. The desert race also exhibited the capacity to assume the perennial growth habit. Perennial plants of the desert race produced 4.4 heads/plant (116 heads/m2) versus 0.2 heads/plant (4 heads/m2) for annual plants of the desert race under field conditions. Annual plants of the foothills race produced 2.7 heads/plant (75 heads/m2). The perennial habit did not occur in field populations of the foothills race. When grown in identical glasshouse conditions the desert race reached anthesis of the first four flower heads ca. 25 days prior to the foothills race. This acceleration to anthesis was accompanied by a greater potential reproductive allocation. The latter response was due in part to greater biomass allocation to reproductive structures at the expense of vegetative structures, and the uncoupling of net photosynthesis from phenological events.  相似文献

Isoprene emission in relation to stomatal distribution and conductance was determined for the hypostomatous species, aspen and white oak, and the amphistomatous species, cottonwood. For aspen and oak, isoprene emission from the adaxial (nonstomatal) surface was <2% of that from the abaxial (stomatal) surface, even when stomata were closed by addition of abscisic acid (ABA). When treated with ABA, the total flux rate of isoprene emission from leaves of these two hypostomatous species was unchanged, despite decreases in stomatal conductance of over 90%. The lack of control over isoprene emission rate by stomatal conductance, despite the apparent movement of isoprene through the stomatal pores, was due to increases in the intercellular isoprene concentration that compensated for the decreased stomatal conductance and restored the equilibrium between the isoprene synthesis rate and emission rate. This relationship was demonstrated by (a) an experiment in which the decrease in the internal isoprene pool following the imposition of darkness took longer in the presence of ABA than in its absence, and (b) direct measurements of the internal isoprene concentration through vacuum extraction, which revealed substantially higher values in the presence of ABA than in its absence. In the amphistomatous species, cottonwood, isoprene was emitted from both surfaces and addition of ABA caused an increase in isoprene emission from one surface coupled with a decrease from the other surface. The specific surface exhibiting an increase varied among leaves, with some leaves exhibiting an increase from the adaxial surface and other leaves from the abaxial surface. We interpret this as indicating nonuniform stomatal closure with concomitant emission of isoprene at the greatest rate from the surface with the highest stomatal conductance. We also observed an increase in the total isoprene emission rate from cottonwood leaves following treatment with ABA. We interpret this as indicating a stimulation of isoprene synthesis in response to ABA or stomatal closure, with unknown cause.  相似文献

Lactation is the most energetically demanding stage of reproduction in female mammals. Increased energetic allocation toward current reproduction may result in fitness costs, although the mechanisms underlying these trade‐offs are not well understood. Trade‐offs during lactation may include reduced energetic allocation to cellular maintenance, immune response, and survival and may be influenced by resource limitation. As the smallest marine mammal, sea otters (Enhydra lutris) have the highest mass‐specific metabolic rate necessitating substantial energetic requirements for survival. To provide the increased energy needed for lactation, female sea otters significantly increase foraging effort, especially during late‐lactation. Caloric insufficiency during lactation is reflected in the high numbers of maternal deaths due to End‐Lactation Syndrome in the California subpopulation. We investigated the effects of lactation and resource limitation on maternal stress responses, metabolic regulation, immune function, and antioxidant capacity in two subspecies of wild sea otters (northern: E. l. nereis and southern: E. l. kenyoni) within the California, Washington, and Alaska subpopulations. Lactation and resource limitation were associated with reduced glucocorticoid responses to acute capture stress. Corticosterone release was lower in lactating otters. Cortisol release was lower under resource limitation and suppression during lactation was only evident under resource limitation. Lactation and resource limitation were associated with alterations in thyroid hormones. Immune responses and total antioxidant capacity were not reduced by lactation or resource limitation. Southern sea otters exhibited higher concentrations of antioxidants, immunoglobulins, and thyroid hormones than northern sea otters. These data provide evidence for allocation trade‐offs during reproduction and in response to nutrient limitation but suggest self‐maintenance of immune function and antioxidant defenses despite energetic constraints. Income‐breeding strategists may be especially vulnerable to the consequences of stress and modulation of thyroid function when food resources are insufficient to support successful reproduction and may come at a cost to survival, and thereby influence population trends.  相似文献

Abstract: Isoprene is emitted from the leaves of some plants. It was recently reported that exogenous isoprene delays the onset of leaf damage during controlled increases in leaf temperature (Singsaas et al. Plant Physiology 115: 1413–1420 [1997[17). Thylakoid membranes are presumed to be the site of action based upon isoprene's hydrophobicity, production in chloroplasts, and effect upon chlorophyll fluorescence at high temperatures. In an attempt to discern the mechanistic basis for isoprene's thermoprotective role, we studied the effect of exogenous isoprene on the peroxidation, permeability, and stability of spinach thylakoids and phosphatidylcholine liposomes. Isoprene, supplied at either 18 or 21 μ1 L1, had no effect upon the rate of liposome peroxidation in the presence of a hydroxyl radical-generating system. Isoprene also did not affect liposome peroxidation at high temperatures. Neither the proton permeability of thylakoids nor the leakage of a fluorescent probe from liposomes was influenced by exogenous isoprene, when measured at several temperatures. Isoprene did not affect the stability of thylakoid membrane proteins during a temperature increase, as shown by differential scanning calorimetry. Therefore, despite the use of a variety of techniques to investigate fundamental membrane parameters, we were unable to demonstrate an effect of isoprene.  相似文献

In vitro and in vivo developmental competence of fresh and cryopreserved in vitro produced (IVP) bovine embryos was evaluated up to birth. Three experiments were done. The objective in the first experiment was to develop an optimal vitrification procedure for IVP bovine embryos by determining effects of exposure time (2, 5, 10, 20 min) and temperature (4, 22, 27 degrees C) in cryoprotective agents prior to vitrification on their post-thaw viability. The best combination was used in Experiments 2 and 3. In the second experiment, the importance of post-thaw morphologic selection on pregnancy rates was determined by transferring either selected or unselected single embryos. In the third experiment, pregnancy initiation, maintenance and calving results of vitrified embryos were compared with fresh and conventionally frozen embryos. Fetal losses, birth weights, gestation lengths and frequency of dystocia in the third experiment were monitored. The interaction of exposure time and temperature on both post-thaw re-expansion and hatching rates was significant (P < 0.01). Five minute exposure at 27 degrees C was optimal. In the second experiment, post-thaw selected vitrified embryos had higher pregnancy rates than unselected embryos (P < 0.05). In the third experiment, the pregnancy rate of vitrified embryos did not differ from that of fresh embryos (P > 0.05). However, pregnancy rate of conventionally frozen embryos was lower than that of fresh or vitrified embryos (P < 0.05). Of 92 calves born, 53 were male and 39 were female. Birth weights and dystocia scores of single-born calves did not differ between sexes (P > 0.05). Twin-born calves were lighter than single-born calves (P < 0.05). Overall, the data demonstrate that the transfer of vitrified IVP bovine embryos can result in healthy, apparently normal calves similar to those derived from transfer of fresh and conventionally frozen IVP bovine embryos.  相似文献

Although isoprene synthesis is closely coupled to photosynthesis, both via ATP requirements and carbon substrate availability, control of isoprene emission is not always closely linked to photosynthetic processes. In this study we grew velvet bean (Mucuna sp.) under different levels of photon flux density (PFD) and nitrogen availability in an effort to understand better the degree to which these two processes are linked. As has been observed in past studies, we found that during early leaf ontogeny the onset of positive rates of net photosynthesis precedes that of isoprene emission by 3 to 4 d. Other studies have shown that this lag is correlated with the induction of isoprene synthase activity, indicating that overall control of the process is under control of that enzyme. During leaf senescence, photosynthesis rate and isoprene emission rate declined in parallel, suggesting similar controls over the two processes. This coordinated decline was accelerated when plants were grown with high PFD and high nitrogen availability. The latter effect included declines in the photon yield of photosynthesis, suggesting that an unexplained stress arose during growth under these conditions, triggering a premature decline in photosynthesis and isoprene emission rate. In mature leaves, growth PFD and nitrogen nutrition affected photosynthesis and isoprene emission in qualitatively similar, but quantitatively different, ways. This resulted in a significant shift in the percentage of fixed carbon that was re-emitted as isoprene. In the case of increasing growth PFD, isoprene emission rate was more strongly affected than photosynthesis rate, and more carbon was lost as isoprene. In the case of increasing nitrogen, photosynthesis rate increased more than isoprene emission rate, and leaves containing high amounts of nitrogen lost a lower percentage of their assimilated carbon as isoprene. Taken together, our results demonstrate that, although the general correlation between isoprene emission rate and photosynthesis rate is consistently expressed, there is evidence that both processes are capable of independent responses to plant growth environment.  相似文献

Recent studies of allocation to defensive chemicals in plants have provided insights into the ecological controls over plant defensive chemicals. Both developmental and ecological studies now suggest that we can understand the factors influencing allocation to defense by examining the relative availability of resources, external needs for chemical defense, and the internal demands for growth that plants face. These studies have also shed light on one of the more popular theories in plant evolutionary ecology, the growth-differentiation balance hypothesis of plant resource allocation.  相似文献
