The decrease in contact hip joint stress after Chiari osteotomy is studied using a mathematical model. In the model, additional coverage of the femoral head by the ala ossis ilei segment is taken into account. It is shown that this additional coverage significantly decreases stress, mostly by the indirect effect caused by the shift of the stress pole.  相似文献

In Xenopus laevis egg extracts, TPX2 is required for the Ran-GTP-dependent assembly of microtubules around chromosomes. Here we show that interfering with the function of the human homologue of TPX2 in HeLa cells causes defects in microtubule organization during mitosis. Suppressing the expression of human TPX2 by RNA interference leads to the formation of two microtubule asters that do not interact and do not form a spindle. Our results suggest that in vivo, even in the presence of duplicated centrosomes, spindle formation requires the function of TPX2 to generate a stable bipolar spindle with overlapping antiparallel microtubule arrays. This indicates that chromosome-induced microtubule production is a general requirement for the formation of functional spindles in animal cells.  相似文献

The high ingestion of soybean products in Asian countries has been suggested to be responsible for a reduced incidence of prostate cancer. The mechanism of action, however, is unknown. Our data demonstrate that genistein and some isoflavone metabolites reduce the activity of 25-D3-24-hydroxylase (CYP24) in the human prostate cancer-derived cell line DU-145. CYP24 is also responsible for degradation of the active vitamin D metabolite 1,25-dihydroxyvitamin D3 which is known to be antimitotic and prodifferentiating in prostate cancer cells. High levels of CYP24 frequently found in prostate cancer cells may thus degrade the active metabolite. This could be prevented by ingestion of genistein-containing food such as soybeans.  相似文献

To identify the receptor which may determine the macrophage tropism of porcine reproductive and respiratory syndrome virus (PRRSV), monoclonal antibodies (MAbs) against porcine alveolar macrophages (PAM) were produced. Two MAbs (41D3 and 41D5) which completely blocked PRRSV infection of PAM were further characterized. It was found that they reduce the attachment of PRRSV to PAM and immunoprecipitate a 210-kDa membrane protein from PAM. This protein was detected on the cell membranes of PAM but not of PRRSV-nonpermissive cells. A colocalization was found between the reactive sites of MAb 41D3 and PRRSV on PAM membranes. All PRRSV-infected cells in tissues of experimentally infected pigs reacted with MAb 41D3. Taken together, all these data suggest that the identified 210-kDa membrane protein is a putative receptor for PRRSV on porcine macrophages.  相似文献

Animal and clinical studies have shownrespiratory muscle dysfunction caused by treatment withglucocorticoids. The present study was designed to investigate whetheranabolic steroids are able to antagonize the loss of diaphragm forceinduced by long-term low-dose methylprednisolone (MP) administration.Male adult rats were randomized to receive saline or MP (0.2 mg · kg1 · day1sc) during 9 mo, with or without nandrolone decanoate (ND; 1 mg · kg1 · wk1im) during the last 3 mo. The ~10% reduction in force generation ofisolated diaphragm bundles induced by MP was completely abolished byaddition of ND. The MP-induced decrease in number of fibers expressingtype IIb myosin heavy chains was not reversed by ND. MP slightlyreduced type I, IIa, and IIx fiber cross-sectional areas(CSA), but not type IIb fiber CSA. Addition of ND abolished thereduction in IIa and IIx fiber CSA. The MP-induced alterations inglycogenolytic activity and fatty acid oxidation capacity were notreversed by ND. In conclusion, the marked reduction in diaphragm forcecaused by long-term low-dose MP was completely abolished by addition ofND. ND in part also antagonized the effects of MP on diaphragmmorphology but showed no beneficial effects on biochemical changes.

Gross oxygen evolution was compared with the electron transport rate (ETR), estimated from chl a fluorescence parameters on the common tropical green macro alga Ulva fasciata Delile with confirmatory carbon saturation curves from U. reticulata Forskål. Theoretically, the relationship between estimated ETR and gross oxygen evolution should be 4:1, that is, four electrons are transported through PSII for each molecule of oxygen evolved. However, deviations of the 4:1 relationship have previously been reported. Measurements were conducted with two commercially available and portable pulse amplitude modulated (PAM) chl fluorometers. We sought experimental approaches that minimize discrepancies between the two different measuring techniques of photosynthetic rates, both for in situ and laboratory conditions. Using fresh algal tissue for each of the different irradiances gave the best fit of gross oxygen evolution and ETR even at irradiances above light saturation, where large discrepancies between oxygen evolution and ETR are common. With increasing dissolved inorganic carbon (DIC) concentrations, there was a curvilinear response of gross oxygen evolution in relation to ETR. We therefore suggest to establish DIC saturation curves in the laboratory, oxygen evolution is probably the most relevant choice. Photorespiration could not readily explain a curvilinear response of O2 evolution and proportionally higher ETR at high irradiances. ETRs measured with the rapid light curve function of the PAM were compared with steady‐state rates of gross and net oxygen evolution, and the ETR was found to decrease at higher irradiances whereas oxygen evolution was constant.  相似文献

Despite the extensive use of photographic identification methods to investigate humpback whales in the North Pacific, few quantitative analyses have been conducted. We report on a comprehensive analysis of interchange in the North Pacific among three wintering regions (Mexico, Hawaii, and Japan) each with two to three subareas, and feeding areas that extended from southern California to the Aleutian Islands. Of the 6,413 identification photographs of humpback whales obtained by 16 independent research groups between 1990 and 1993 and examined for this study, 3,650 photographs were determined to be of suitable quality. A total of 1,241 matches was found by two independent matching teams, identifying 2,712 unique whales in the sample (seen one to five times). Site fidelity was greatest at feeding areas where there was a high rate of resightings in the same area in different years and a low rate of interchange among different areas. Migrations between winter regions and feeding areas did not follow a simple pattern, although highest match rates were found for whales that moved between Hawaii and southeastern Alaska, and between mainland and Baja Mexico and California. Interchange among subareas of the three primary wintering regions was extensive for Hawaii, variable (depending on subareas) for Mexico, and low for Japan and reflected the relative distances among subareas. Interchange among these primary wintering regions was rare. This study provides the first quantitative assessment of the migratory structure of humpback whales in the entire North Pacific basin.  相似文献

In this exploratory study of 15 families with a twin-pair, of which one has an intellectual disability, the Nijmegen Questionnaire on Childrearing Situations (NQCS) was used to ask mothers about their perceptions and experiences of parenting. An interview with the mothers was done to understand the initial stages of coping with this double problem: educating a twin-pair and a child with an intellectual disability. Four groups of mothers were identified, depending on whether they accepted the intellectual disability of their child and/or whether they perceived their children as a twin-pair or not. Finally, some critical questions for further research and management were formulated.  相似文献

Phytoplankton and zooplankton biomass distributions were calculated on a carbon basis for the inland part of the Oosterschelde, in the period before (1983), during (1984, 1986) and after (1987, 1988) the construction of the storm- surge barrier. In all years studied, both phytoplankton and zooplankton distributions are very irregular, and little consistent patterns emerge. The data were used to test the model of Sheldon et al. (1977). The observed standing stock ratios of zooplankton to phytoplankton agree with the model predictions in 1983, and are slightly higher during the period 1984–1987. In 1988, the model predictions are very different from the observed values, because of important changes in the zooplankton species abundance occurring in this year.  相似文献
