Thermophilic sulfate-reducing bacteria were isolated from oil field waters from oil production platforms in the Norwegian sector of the North Sea. Spore-forming rods dominated in the enrichments when lactate, propionate, butyrate, or a mixture of aliphatic fatty acids (C(4) through C(6)) was added as a carbon source and electron donor. Representative strains were isolated and characterized. The isolates grew autotrophically on H(2)-CO(2) and heterotrophically on fatty acids such as formate, propionate, butyrate, caproate, valerate, pyruvate, and lactate and on alcohols such as methanol, ethanol, and propanol. Sulfate, sulfite, and thiosulfate but not nitrate could be used as an electron acceptor. The temperature range for growth was 43 to 78 degrees C; the spores were extremely heat resistant and survived 131 degrees C for 20 min. The optimum pH was 7.0. The isolates grew well in salt concentrations ranging from 0 to 800 mmol of NaCl per liter. Sulfite reductase P582 was present, but cytochrome c and desulfoviridin were not found. Electron micrographs revealed a gram-positive cell organization. The isolates were classified as a Desulfotomaculum sp. on the basis of spore formation, general physiological characteristics, and submicroscopic organization. To detect thermophilic spore-forming sulfate-reducing bacteria in oil field water, polyvalent antisera raised against antigens from two isolates were used. These bacteria were shown to be widespread in oil field water from different platforms. The origin of thermophilic sulfate-reducing bacteria in the pore water of oil reservoirs is discussed.  相似文献

One of the earliest events leading to cell activation and growth is the hydrolysis of inositol phospholipids producing various membrane signals induced by an interaction between growth factors or hormones with their respective receptors on the cell membrane [1].To demonstrate the mitogenic action of transferrin,our results show that an addition of transferrin to “serum-deprived” rat hepatoma cells produced a rapid but transient rise in inositol 1,4,5-trisphosphate(IP3) level,and at the same time,an increased intracellular Ca^2 activity and a cytoplasmic alkalinization were observed.These signal transductions further lend support to the mitogenic nature of transferrin.In addition,a possible link between the receptor-mediated endocytosis of transferrin with the generation of intracellular signals is discussed herewith.  相似文献

By using the fluorescence enhancement of ethidium bromide bound to nuclei acid, a very rapid, simple and sensitive assay of DNA in the green alga Chlamydomonas has been devised. Total fluorescence (DNA + RNA) was determined by complex formation with ethidium bromide in a cell lysate made by mixing cell samples with lauroyl sarcosinate, EDTA and NaOH and incubating the mixture for 5 min at room temperature followed by neutralization. For determination of DNA the RNA was digested by incubating the cell sample in te alkaline lysis solution for 45 min at 60 degrees C followed by neutralization, and complex formation with ethidium bromide. Quenching of the fluorescence due to cellular pigments was corrected for using an internal DNA standard.  相似文献

The α adrenergic agonistic activity of a number of imidazolines was measured on the α adrenergic receptors of the isolated rabbit intestine. A number of imidazolines were full agonists having an intrinsic activity constant equal to 1 in relation to noradrenaline as a standard. Some other derivatives had only partial agonist activity. The affinities of the imidazolines for these adrenergic receptors was assessed using pD2 values. The pD2 values were correlated with a number of physicochemical parameters of the drugs, i.e. pKa, the molar volume and the apparent and true partition coefficients (APC and TPC respectively) by stepwise regression analysis.Results indicate that pD2 was correlated with pKa primarily, whereas molar volume also was a factor determining agonistic activity at the peripheral α adrenergic receptor.Central α adrenergic receptor stimulating properties of the imidazolines were determined by measuring bradycardia induced by these agents after intrahypothalamic injection into rats. Although it was not possible to correlate central α adrenergic activity with any of the physicochemical parameters due to the lack of action of a number of imidazolines, results show that structural requirements for central α adrenergic activity clearly differ from those for peripheral α adrenergic activity.These data are discussed in view of the mechanism of action of the antihypertensive imidazoline clonidine.  相似文献

Arylsulfatase (aryl-sulfate sulfohdydrolase, EC 3.1.6.1) has been purified from SO4-2-minus-starved cells of Chlamydomonas reinhardti. The enzyme was isolated from acetone-powder extract by (NH4)2SO4 precipitation, Sephadex G-200 filtration and ion-exchange chromatography. Only one fraction of aryl-sulfatase was found. The final preparation was homogenous by the criteria of sedimentation, diffusion and polyacrylamide gel electrophoresis. The purified enzyme had a molecular weight of about 150 000, estimated by ultracentrifugation and gel filtration, and an isoelectric point of 9.0. The properties of the enzyme as investigated in intact cells and in the purified state were found to be very similar except for the temperature optimum. Imidazole strongly increased the enzyme by increasing the V, but reduced the affinity for the substrate. The enzyme activity was competitively inhibited by borate with a greater affinity for borate than for the substrate. The Chlamydomonas enzyme is a Type I arylsulfatase since it was inhibited by CN-minus, but not SO4-2-minus and phosphate.  相似文献

The external surface of P. conostomum is characterized by relatively large ridges encircling the anterior part of the worm at regular intervals. On the posterior part depressions on the ventral side at regular intervals and relatively small ridges on the dorsal side are present. Ventroposteriorally cobblestone-like protuberances observed are arranged in longitudinal rows. No corresponding arrangement was found dorsally. Only domed papillae (with and without a central knob) were observed, tentatively identified as sensory organs. The regular pattern of these papillae on the ventral and oral sucker is described, in addition to their arrangement ventrally and at the anterior end. A frontal pit anterior of oral suckers and a notch at the posterior end are figured and briefly described. No spines were observed on the body tegument.  相似文献
