T cells have an important role during the development of autoimmune diseases. In adjuvant arthritis, a model for rheumatoid arthritis, we found that the percentage of CD4+ T cells expressing the activation marker CD134 (OX40 antigen) was elevated before disease onset. Moreover, these CD134+ T cells showed a specific proliferative response to the disease-associated epitope of mycobacterial heat shock protein 60, indicating that this subset contains auto-aggressive T cells. We studied the usefulness of CD134 as a molecular target for immune intervention in arthritis by using liposomes coated with a CD134-directed monoclonal antibody as a drug targeting system. Injection of anti-CD134 liposomes subcutaneously in the hind paws of pre-arthritic rats resulted in targeting of the majority of CD4+CD134+ T cells in the popliteal lymph nodes. Furthermore, we showed that anti-CD134 liposomes bound to activated T cells were not internalized. However, drug delivery by these liposomes could be established by loading anti-CD134 liposomes with the dipalmitate-derivatized cytostatic agent 5'-fluorodeoxyuridine. These liposomes specifically inhibited the proliferation of activated CD134+ T cells in vitro, and treatment with anti-CD134 liposomes containing 5'-fluorodeoxyuridine resulted in the amelioration of adjuvant arthritis. Thus, CD134 can be used as a marker for auto-aggressive CD4+ T cells early in arthritis, and specific liposomal targeting of drugs to these cells via CD134 can be employed to downregulate disease development.  相似文献

An efficient organic acid profiling and pattern recognition method is described for the correlation between urinary organic acid profiles and uterine cervical cancer. After methoximation of keto acids in alkalinized urine samples, all free organic acids were recovered by a dual solid-phase extraction procedure, followed by conversion to tert.-butyldimethylsilyl derivatives for the profiling analysis by dual-capillary column gas chromatography (GC) with subsequent screening for acids by retention index (I) library matching. A total of 50 organic acids were positively identified in urine samples (0.25 ml) from 12 uterine myoma (benign tumor group) and 14 uterine cervical cancer (malignant tumor group) patients studied. When the GC profiles were simplified to their corresponding organic acid I spectra in bar graphical form, characteristic patterns were obtained for each average of benign and malignant tumor groups. Stepwise discriminant analysis performed on the GC data selected 16 acids as the variables discriminating between the two groups. Canonical discriminant analysis applied to these 16 variables correctly classified 26 urine samples into two separate clusters according to tumor types in the canonical plot.  相似文献

By means of electron microscopy and ultrahistochemical research it has been attempted to detail the structure and explain the function of the pleural tufts of the parietal pleura present in the ventral mediastinum of dogs. This led to the discovery that the varions forms present for the surface-enlarging pleura protusions in actuality exhibit corresponding morphological characteristics. As a special formation of the serous lamina of parietal pleura, they are covered by an occasionally incomplete mesothel unit. The parenchyma of these organs is compinsed of a macrophage-rich outer border zone and of a vessel-canying central zone. The experimental in vivo application of cationized ferritin in the pleura-space clearly indicated a high resorptive metabolism of the mesothel and of the pluripotential mesenchym cells. The higher incidence of lymphoplasmatic cell-forms in the proximity of the central vessels along with leucocytic cell infiltrations fulfill an immunocellular and phagocytic function. The structure of the cell population of the pleural tufts partaking on the resorption of matter out of the pleural space has been discussed in detail. The in vivo application of a cationized ferritin solution for the purpose of studying these resorptive processes has been critically judged.  相似文献
