To determine whether a rat heart model can provide load-insensitive measurements of cardiac function, a recently developed biventricular perfused preparation was tested. Using 29 Sprague-Dawley rat hearts perfused with modified Krebs-Henseleit buffer, ventricles functioned simultaneously with adjustable independent preload (venous reservoirs) and afterload (compliance chambers). Ultrasonic crystal pairs provided continuous left (LV) and right ventricular (RV) short-axis dimensions. LV and RV pressure-length loops (loop area = work) were generated from paired intraventricular pressure and short-axis dimensions. Load-insensitive measurements were obtained from the slopes (elastance) and x-intercepts (L0) of regression lines generated from the end-systolic coordinates of these pressure-length loops over ranges of RV and LV preloads. Measurements were made after 15 min of stable function and after 20 min of warm (37°C) ischemia. During perturbations in LV afterload, there were linear changes in dP/dt, but loop work remained relatively unchanged. RV dP/dt and work varied little with physiologic ranges of afterload. Increased RV afterload had little effect on LV function. Ischemia affected LV function more than RV function using these measurements. Elastance, however, increased after ischemia with diastolic creep (increased L0) for both ventricles. Load-insensitive and other sophisticated hemodynamic measurements are possible with this new preparation.  相似文献

Within the last 30 years, researchers have explored what role hypoxia might play in causing permeability changes in the pulmonary microvasculature. Since the data accumulated thus far are unclear, the effects of hypoxia on microvascular transport in the isolated, Ringer's perfused adult rabbit lung was observed and the following parameters were measured or computed for both oxygenated and hypoxic perfusates: pulmonary arterial (ra) and pulmonary venous (rv) resistances, pulmonary capillary filtration coefficients (Kf), and pulmonary capillary endothelial reflection coefficients () for NaCl and inulin. Separate reservoir bottles were used to create the desired oxygenated (aeration of solution with 95% O2-5% CO2) gas mixture or hypoxic (aeration of solution with 95% N2-5% CO2) gas mixture. A higher, but not significant, resistance value was found during the oxygenated state. A significant increase in the pulmonary capillary filtration coefficient during hypoxia (10.72 × 10–4±0.446 × 10–4 cm3/s cm H2O for the hypoxic perfusate and 8.80 × 10–4±0.384 × 10–4 cm3/s cm H2O for the oxygenated perfusate) was found and a significant difference between oxygenated and hypoxic pulmonary capillary reflection coefficients for inulin was computed (oxygenated solution revealed a finding of 0.120±0.003 and the hypoxic solution revealed 0.105±0.002). These findings imply a change in the microvascular permeability during hypoxia. According to the pore theory, a change in pore number, pore size, or both could have occurred. However, from the reflection coefficient data, a change in pore radius seems most likely.  相似文献

Incubation of (20R)-20-phenyl-5-pregnene-3beta,20-diol, an aromatic analog of (23S)-20-hydroxycholesterol, with an adrenal mitochondrial preparation leads to the formation of four compounds: pregnenolone, phenol, a C8 ketone, acetophenone, and a nonpolar C19 compound. This latter compound has now been identified by reverse isotope dilution analysis and by gas chromatography/mass spectrometry as 17-methyl-18-norandrosta-5,13(17)-dien-3beta-ol. From these results it is evident that enzymatic fission of the C-17,20 bond of this synthetic derivative occurs. On the other hand, when (20S)-20-hydroxy[21-14C]cholesterol was used as substrate, the analogous cleavage did not take place. Thus, substitution of an aromatic group on C-20 facilitates side chain cleavage between that carbon atom and the nucleus whereas neither of the naturally occuring precursors, cholesterol or its 20-hydroxylated counterpart, are metabolized to a C8 fragment.  相似文献

Summary To determine if harmala alkaloids affect transport systems other than (Na +K)-ATPase, effects of harmaline on Na and water fluxes were studied in amphibian skins. Net Na flux was evaluated from short-circuit current, and water flux monitored with automatic, volumetric methods. At 2 to 5mm, harmaline consistently inhibited SCC and prevented the natriferic effects of oxytocin and norepinephrine. However, at 0.1 to 0.5mm, harmaline produced an increase in SCC inhibitable with amiloride. The stimulatory effects of harmaline and oxytocin were either nonadditive or additive depending on whether the hallucinogen was present in the inner solution or in the outer solution bathing the skin, respectively. Water flow was not modified by harmaline on the outer medium. In contrast, addition of the drug to the inner medium elicited a conspicuous, sustained, vasopressin-like, hydrosmotic effect, comparable to and competitive with those of vasopressin and norepinephrine. The ensemble of these results suggests that harmaline may affect three distinct transport systems: (i) the Na pump; (ii) the cyclic nucleotide system; (iii) the Na entry pathway at the outer membrane of the skin that is also activated by agents such as diphenylhydantoin, lanthanides and propranolol.  相似文献

Daily turnover of cholesterol obtained by the balance method was compared to daily input rates calculated by input-output analysis in 43 experiments. The mean value of input rates for kinetic data of 10.1-16.4 weeks' duration (14 experiments) was 1.05 g/day vs. the chemical turnover of 0.94 g/day (difference 10.9 percent). For decay curves of 4.8-9.9 weeks' duration (29 experiments) the mean results were 1.67 g/day vs. 1.31 g/day, respectively (difference 20.1 percent). A combination of the balance method with input-output analysis is proposed to estimate the size of M (minimum value of the total exchangeable mass of cholesterol) in short-term experiments. Using this method, the analysis of curves of 10-12 weeks' duration showed a mean difference of 7.5 percent with the analysis of curves of 50-66 weeks' duration in 17 patients. However, because of considerable variations that can occur in individual cases, it is urged that a standard correction factor not be used, either in estimating turnover data or M from 10-12 weeks' kinetic data; rather, the proposed combined method will alert the investigator to the occurrence of discrepant results.  相似文献

Cortex tissue from postclimacteric `Golden Delicious' apples (Malus domestica, Borkh.) stored at 0 C for 9 months after harvest were induced to form callus in vitro. Cell suspension cultures were subsequently formed from calli. Of five media tested, only the medium of Schenk and Hildebrandt (Can J Bot 1972, 50: 192) and that of Uchimiya and Murashige (Plant Physiol 1974, 54: 936) allowed callus formation. During growth both the callus and cell cultures produced ethylene in a pattern which showed a rapid rise and then a fall as the culture grew. 14C-Labeled methionine was converted to labeled ethylene by the cell suspension cultures, which also could be inhibited from producing ethylene by a rhizobitoxine analog or free radical scavengers. Ethylene production in these cultures, like that in intact fruit tissue slices, could be stimulated by IAA or suppressed by N6-(γ,γ-dimethylallyl) adenosine and GA3.  相似文献
