本文用脑室灌注和Fura2测定细胞内游离钙技术观察了地塞米松(dexamethasone,DEX)对家兔乙二醇双(2氨基乙醚)四乙酸性发热效应和下丘脑细胞内游离钙浓度([Ca2+]i)的影响,借此深入探讨地塞米松解热作用的中枢机制。结果发现:脑室灌注乙二醇双(2氨基乙醚)四乙酸(06nmol)引起家兔结肠温度明显升高,静脉注射地塞米松(5mg/kg)显著抑制家兔乙二醇双(2氨基乙醚)四乙酸性发热,地塞米松(60~120μmol/L)并不影响下丘脑细胞内[Ca2+]i,而事先脑室灌注抑制基因转录的放线菌素D(3nmol)则完全取消了地塞米松对乙二醇双(2氨基乙醚)四乙酸性发热的解热作用。这些结果提示:地塞米松显著抑制家兔乙二醇双(2氨基乙醚)四乙酸性发热,其机制与地塞米松激活脑内某些基因的表达有关,而与下丘脑神经细胞跨膜钙离子流无关。 相似文献

Inclination for hyperinsulinemia that was formed in the human population at early stages of development and was fixed genetically in the process of evolution is suggested to underlie pathogenesis of the currently most widely spread diseases of the cardiovascular system (atherosclerosis and hypertension) as well as diabetes mellitus of the 2 type and obesity that are concomitant with a high frequency. Under conditions of civilization, chronic hyperinsulinemia can develop in response to the regularly excessive nutrition. The consequence of the excessive food consumption and the chronic hyperinsulinemia can be overcrowding with lipids (triglycerides) of adipose tissue—the organism lipid store. The natural protective reaction of the cell that contains the limitedly possible amount of lipids is a decrease of the number of insulin receptors and development of insulin resistance. The insulin resistance, in turn, provides the appearance of hyper- and dislipoproteinemia alongside with hyperglycemia. One of the most clinically significant ways of achievement of homeostasis is storage of lipids in the arterial wall. Atherogenic and other concomitant metabolic disorders, specifically changes of blood coagulation properties providing susceptibility to hypercoagulation, affect the blood rheological properties and seem to lead to pathology of the entire vascular system: chronic venous insufficiency, disturbance of microcirculation, and arterial atherosclerosis. DAP5 is an eIF4G protein previously implicated in mediating cap-independent translation in response to cellular stresses. Here we report that DAP5 is crucial for continuous cell survival in nonstressed cells. The knockdown of endogenous DAP5 induced M phase-specific caspase-dependent apoptosis. Bcl-2 and CDK1 were identified by two independent screens as DAP5 translation targets. Notably, the activity of the Bcl-2 IRES was reduced in DAP5 knockdown cells and a selective shift of Bcl-2 mRNA toward light polysomal fractions was detected. Furthermore, a functional IRES was identified in the 5'UTR of CDK1. At the cellular level, attenuated translation of CDK1 by DAP5 knockdown decreased the phosphorylation of its M phase substrates. Ectopic expression of Bcl-2 or CDK1 proteins partially reduced the extent of caspase activation caused by DAP5 knockdown. Thus, DAP5 is necessary for maintaining cell survival during mitosis by promoting cap-independent translation of at least two prosurvival proteins. 相似文献

Influence of different metabolic poisons on cAMP-evoked neuron membrane permeability is investigated. Drugs preventing cAMP binding with R subunits of protein kinase decrease the cAMP-evoked current, but the inhibitor of the C subunit. H8, has no effect. Recovery of the histomorphometric indices of rat skin at the period from 38 to 150 days following local X-irradiation with doses of 2.5-20 Gy has been studied. It is shown that the rate of restitution of the epidermis and dermis thickness is 0.0021 and 0.0037 days-1, respectively. However, the dermis thickness is less than in the control. The hair follicle density not only fails to restore after irradiation with a dose of above 10 Gy but continues decreasing in time. 相似文献

In the course of a study on the early events of cambial derivative differentiation in Populus × euramericana, seasonal changes in the pattern of pectin methylesterase (PME, EC 3.1.1.11) isoforms were followed. During the resting season,
cell wall extracts contained mainly alkaline isoforms with an Mr around 55 kDa and optimal pH between 5.6 and 6.0. Neutral isoforms with an Mr around 35 kDa and optimal pH between 6.0 and 6.6 predominated in the extracts during the period of high meristematic activity.
In the active cambial initials and in their immediate derivatives, the enzymes were immunolocalized exclusively in the dictyosomes.
In older cells, they were present both in dictyosomes and in wall junctions. These results indicate that exportation of neutral
PMEs towards the walls might be considered as an early marker of differentiation in cambial derivatives.
