Solubility of sickle hemoglobin measured by a kinetic micromethod.

We have developed a photolytic method to determine the concentration of reactive hemes in a solution in the presence of a trace amount of CO. By measurement of the bimolecular rate of CO binding, and by calibration of the rate constant under equivalent conditions, the concentration of the reactive hemes can be determined. In a solution of sickle hemoglobin, the molecules in the gel contribute negligibly to the recombination rate, allowing the concentration of the molecules in the solution phase to be determined. To optimize signal to noise, modulated excitation methods were employed, although the method could also be used with pulse techniques and suitable signal averaging. Because the optical method employs a microspectrophotometer, only a few microliters of concentrated Hb solution is required to reproduce the entire temperature dependence of the solubility previously determined by centrifugation using milliliter quantities of solutions of the same concentration. This should be especially useful for studies of site-directed mutants, and we present results obtained on one such HbS in which Leu 88 beta has been replaced by Ala. The free energy difference in the polymerization of the Leu 88 beta double mutant is consistent with known differences in the amino acid hydrophobicities. The calibration required for these experiments also provides an excellent determination of the activation energy for binding the first CO to deoxy Hb.     

FLIGHT ACTIVITY AND FATTY ACID UTILIZATION IN MYTHIMNA SEPARATA (WALKER) MOTHS*

Abstract The fatty acid (FA) compositions for total lipids from fat body, hemolymph and flight muscle of the armyworm moths, Mythirnna separata, at rest and after tethered flight for 1 h were determined by GC and GC-MS. The composition in these tissues comprises myristic acid (1%-2%), palmitic acid (more than 35%1, palmitoleic acid (9%-11%), stearic acid (less than 1%), oleic acid (about 32%), linoleic acid (12%-17%) and linolenic acid (3%-6%). After flight, FA level in the fat body, compared to that at rest, shows a significant decline at about 20 μg/mg tissue.h^-1; the concentration of FAs in hemolymph rises evidently, but change of FA content in flight muscle appears to be small. From the changes of proportional composition of FAs in fat body, hemolymph and flight muscle, it is found that the FAs selectively utilized for flight in flight muscle are predominantly the palmitic acid and oleic acid.

系统研究了杉木、火力楠纯林及混交林细根生物量、生产力及年周转率。结果表明,杉木、火力楠纯林及混交林活细根生物量分别为８８０、３０３５和１５６０ｋｇ．ｈａ＾－１;死细根生物量为２３５、３９８和５６５ｋｇ．ｈａ＾－１;细根年周转率为１．２９、１．４２和１．４０;年生产量为１１３７、４３１８和２１７９ｋｇ．ｈａ＾－１;年死亡量为４９７．５９５和１１４９ｋｇ．ｈａ＾－１,分别相当于林分枯枝落叶年凋落量为３     

慢性缺氧对大鼠肺内皮素表达的影响

本文以ＡＢＣ法和原位杂交技术,观察了慢性缺氧时大鼠肺组织内内皮素－１（ＥＴ－１）的表达情况,结果发现：①正常肺血管内皮细胞有少许ＥＴ－１样阳性染色物质呈现。②缺氧ＩＷ后,肺内ＥＴ－１含量增加,主要位于肺血管内皮细胞和支气管粘膜上皮细胞。③缺氧２Ｗ和３Ｗ后,ＥＴ１阳性免疫物质进一步增加,于肺泡细胞内也见到阳性染色。④缺氧１Ｗ后肺内ＥＴ－１ｍＲＮＡ表达增加,缺氧２Ｗ和３Ｗ后,ＥＴ－１ｍＲＮＡ的表达进一步加强。提示缺氧可刺激肺内ＥＴ－１ｍＲＮＡ的表达,慢性缺氧时肺内ＥＴ－１持续分泌增加,这可能是缺氧性肺动脉高压发生的重要因素之一。     

酸水解蚕蛹制备复合氨基酸的研究

采用硫酸水解法,以蚕蛹制取复合氨基酸产品,得到氨基酸态氮分别为９．０５％和１３．４５％的食用复合氨基粉和精制复合氨基酸粉。食用复合氨基酸粉含有１８种氨基酸,其中必需氨基酸含量为３９．２％。食用复合氨基酸粉的制备方法经工厂小批量生产证实,其工艺简单易行,适合于中小企业采用,该产品的质量优良,生产成本低廉,具有市场竞争力。     

青霉素和NAA与6—BA配合在离体草莓器官建成中的作用

本实验研究了青霉素和ＮＡＡ与６－ＢＡ配合对离体草莓器官建成以及体内过氧化物同工酶的影响。结果表明：青霉素促进不定根的分化,但抑制不定根的伸长和不定芽的分化,并且不同程度地影响器官建成过程中过氧化和抽工酶的谱带和活性。青霉素与植物激素一样,参与了植物体内的生理代谢而引起植物器官建成。     

t-PA cDNA在CHO细胞中的高效稳定表达

我们曾报道t-PA mRNA非翻译区序列对其表达有明显的抑制作用,在此基础上,通过对5′-UTR及3′-UTR的改造,使t-PA在COS-7细胞中的表达水平提高30倍左右。将t-PA表达质粒用电击法转染中国仓鼠卵巢细胞二氢叶酸还原酶缺陷株(CHO-dhfr),经过混合加压及筛选,在CHO细胞中高效表达了t-PA,表达水平达到5000～6000 IU/10~6细胞/24hr。重组t-PA具有与天然t-PA相同的分子量及酶活性。经过8个月连续传代,表达水平未下降,表明细胞株是稳定的,其主要指标均符合工程细胞株的要求。     

Determination of base specificity of multiple ribonucleases from crude samples

Ribonucleases are widely found in the tissues of living organisms, but the functions of individual ribonucleases are not clear. To facilitate characterization of individual ribonucleases, I have developed a rapid method to separate and identify each ribonuclease from a crude sample by gel electrophoresis instead of by time-consuming purification steps. The ribonucleases in a crude sample are first separated by RNA-cast SDS-polyacrylamide gel electrophoresis and then eluted from the gel after ethidium bromide staining. To determine the base specificity of each ribonuclease, a 5 labelled oligonucleotide with known sequence is added to the enzyme eluate and the digested products are analyzed by denaturing gel electrophoresis. The base specificity of bovine pancreatic ribonuclease (RNase A), bullfrog oocyte-specific ribonuclease (RC-RNase), human serum ribonucleases and sweet potato leaf ribonucleases were determined by this method. Other properties of individual ribonucleases, e.g. substrate preference, may also be determined from crude samples by this method without further purification steps.Abbreviations RNase ribonuclease - SDS sodium dodecyl sulfate
