人工食物对高原鼠兔稳定性碳和氮同位素组成的影响 总被引:6,自引:1,他引:6
以玉米面、小米面和绿豆粉 (质量比为 5∶3∶2 )的混合食物饲喂青藏高原地区特有种高原鼠兔 ,4 0d后断头杀死 ,取其后腿肌肉测定稳定性碳和氮同位素组成。结果表明 ,用混合食物饲喂后 ,高原鼠兔的稳定性碳同位素比值显著升高 ,由对照的 (- 2 4 6 6± 0 2 5 )‰上升为 (- 19 0 5± 0 0 9)‰ ;稳定性氮同位素的组成变化较小 ,仅由对照的 (3 2 8± 0 13)‰增加为饲喂后的 (3 6 1± 0 32 )‰。经人工食物饲喂后 ,高原鼠兔与其食物间稳定性碳同位素的分馏效应为 4 73‰ ,稳定性氮同位素的分馏效应为 2 79‰。C4组分食物的加入影响高原鼠兔的稳定性同位素 (特别是氮同位素 )代谢模式 ,其稳定性同位素代谢周转率可能高于原来预测的 4 0d。 相似文献

The combination of a circular dichroism detector and nonchiral liquid chromatography was used for the chiral analysis of unresolved enantiomers to determine the enantioselectivity of a molybdenum-catalyzed asymmetric allylic alkylation reaction. The CD/UV peak area ratio of the unresolved enantiomers was calculated and compared with that of a reference standard to determine the enantiomeric purity. The limit of quantitation (LOQ) is 20 ng for the chiral ligand and 1 microg for the branched chiral product. The viability of the system depends on the limit of quantitation of the CD response and the linearity range of the CD and UV response. 相似文献

Background
We have previously reported a novel fungal galectin Agrocybe aegerita lectin (AAL) with apoptosis-induced activity and nuclear migration activity. The importance of nuclear localization for AAL's apoptosis-induced activity has been established by mutant study. However, the mechanism remains unclear.Methods
We further investigated the mechanism using a previously reported carbohydrate recognition domain (CRD) mutant protein H59Q, which retained its nuclear localization activity but lost most of its apoptotic activity. The cell membrane-binding ability of recombinant AAL (rAAL) and H59Q was analyzed by FACS, and their cellular partners were identified by affinity chromatography and mass spectroscopy. Furthermore, the interaction of AAL and ligand was proved by mammalian two-hybrid and pull down assays. A knockdown assay was used to confirm the role of the ligand.Results
The apoptotic activity of AAL could be blocked by lactose. Mutant H59Q retained comparable cell membrane-binding ability to rAAL. Four cellular binding partners of AAL in HeLa cells were identified: glucose-regulated protein 78 (GRP78); mortality factor 4-like protein 1 (MRG15); elongation factor 2 (EEF2); and heat shock protein 70 (Hsp70). CRD region of AAL was required for the interaction between AAL/mutant AAL and MRG15. MRG15 knockdown increased the cells' resistance to AAL treatment.Conclusion
MRG15 was a nuclear ligand for AAL in HeLa cells. These data implied the existence of a novel nuclear pathway for the antitumor activity of fungal galectin AAL.General significance
These findings provide a novel explanation of AAL bioactivity and contribute to the understanding of mushroom lectins' antitumor activity. 相似文献994.
Correlation between spike trains or neurons sometimes indicates certain neural coding rules in the visual system. In this
paper, the relationship between spike timing correlation and pattern correlation is discussed, and their ability to represent
stimulus features is compared to examine their coding strategies not only in individual neurons but also in population. Two
kinds of stimuli, natural movies and checkerboard, are used to arouse firing activities in chicken retinal ganglion cells.
The spike timing correlation and pattern correlation are calculated by cross-correlation function and Lempel–Ziv distance
respectively. According to the correlation values, it is demonstrated that spike trains with similar spike patterns are not
necessarily concerted in firing time. Moreover, spike pattern correlation values between individual neurons’ responses reflect
the difference of natural movies and checkerboard; neurons cooperate with each other with higher pattern correlation values
which represent spatiotemporal correlations during response to natural movies. Spike timing does not reflect stimulus features
as obvious as spike patterns, caused by their particular coding properties or physiological foundation. As a result, separating

Background
Human height is a classical example of a polygenic quantitative trait. Recent large-scale genome-wide association studies (GWAS) have identified more than 200 height-associated loci, though these variants explain only 2∼10% of overall variability of normal height. The objective of this study was to investigate the variance explained by these loci in a relatively isolated population of European descent with limited admixture and homogeneous genetic background from the Adriatic coast of Croatia.Methodology/Principal Findings
In a sample of 1304 individuals from the island population of Hvar, Croatia, we performed genome-wide SNP typing and assessed the variance explained by genetic scores constructed from different panels of height-associated SNPs extracted from five published studies. The combined information of the 180 SNPs reported by Lango Allen el al. explained 7.94% of phenotypic variation in our sample. Genetic scores based on 20∼50 SNPs reported by the remaining individual GWA studies explained 3∼5% of height variance. These percentages of variance explained were within ranges comparable to the original studies and heterogeneity tests did not detect significant differences in effect size estimates between our study and the original reports, if the estimates were obtained from populations of European descent.Conclusions/Significance
We have evaluated the portability of height-associated loci and the overall fitting of estimated effect sizes reported in large cohorts to an isolated population. We found proportions of explained height variability were comparable to multiple reference GWAS in cohorts of European descent. 在根癌农杆菌介导的T-DNA(携带有除草剂Basta抗性基因bar和Ds因子)转化中花11水稻群体中,获得了一个叶片发生明显内卷的突变体R1-A。经过连续三代的分离鉴定,获得突变体的纯合株(R1-A2),并与中花11号进行杂交,在调查的36个F_1植株中,全部表现为卷叶,并对Basta除草剂都表现为抗性。在852个F_2单株中,卷叶为645株,正常叶207株,卷叶和正常叶的比例为3:1,其中,卷叶株均对Basta表现抗性,正常叶株均对Basta表现敏感,表明卷叶性状和Basta抗性存在着共分离关系。用扩增Ds因子的引物,对F_2中45个卷叶抗性株进行PCR鉴定,都获得预期长度的Ds因子片段,进一步表明在这些卷叶的植株中都有T-DNA的插入;而30个正常叶敏感株都不能检测到Ds的特征片段。在以卷叶突变(R1-A2)为回交亲本的F_1B_1植株中,全部植株表现卷叶;在以中花11号为回交亲本的F_1B_1植株中,卷叶和正常叶植株的分离比为1:1。上述结果表明该卷叶突变是个显性突变,受一个基因所控制,且该基因的突变与T-DNA的插入有关。 相似文献

Between July 1975 and April 1980, 71 patients were admitted to the Second Attached Hospital of Hubei Provincial Medical College in Wuchang with the diagnosis of epidemic hemorrhagic fever (EHF). The clinical course among these patients was similar to that described for patients with Korean hemorrhagic fever, and hemorrhagic fever with renal syndrome of the U.S.S.R. The overall mortality was 11.2 percent. Sera obtained from some of these patients as well as from patients admitted to the First Attached Hospital of Hubei Provincial Medical College were tested against an antigen associated with Korean hemorrhagic fever and showed exceedingly high antibody titers. We conclude that EHF in Central China represents the same or a closely related disease process as Korean hemorrhagic fever. 相似文献

Despite the widespread use of antiplatelets and anticoagulants, women with antiphospholipid syndrome (APS) may face pregnancy complications associated with placental dysplasia. Neutrophil extracellular traps (NETs) are involved in the pathogenesis of many autoimmune diseases, including vascular APS; however, their role in obstetric APS is unclear. Herein, we investigated the role of NETs by quantifying cell‐free DNA and NET marker levels. Live‐cell imaging was used to visualize NET formation, and MAPK signalling pathway proteins were analysed. Cell migration, invasion and tube formation assays were performed to observe the effects of NETs on trophoblasts and human umbilical vein endothelial cells (HUVECs). The concentrations of cell‐free DNA and NETs in sera of pregnant patients with APS were elevated compared with that of healthy controls (HCs) matched to gestational week. APS neutrophils were predisposed to spontaneous NET release and IgG purified from the patients (APS‐IgG) induced neutrophils from HCs to release NETs. Additionally, APS‐IgG NET induction was abolished with inhibitors of reactive oxygen species, AKT, p38 MAPK and ERK1/2. Moreover, NETs were detrimental to trophoblasts and HUVECs. In summary, APS‐IgG‐induced NET formation deserves further investigation as a potential novel therapeutic target in obstetrical APS. 相似文献
