共生在水生蕨类植物满江红(又称“红萍”:Azolla)叶腔中的固氮兰细菌曾被认为是一个种满江红鱼腥藻(Anabaena azollae Strasb.).但由于现存的满江红在分类学上有2个亚属,7个种1,对不同满江红叶腔中鱼腥藻的鉴别,多年来引起人们的重视.80年代,鱼腥藻的单克隆抗体和RFLP的研究,发现了它与宿主分类上一定程度的对应关系2,3.90年代以后,Van Coppenolle和Eskew分别以RAPD-PCR和DAF-PCR对从满江红共生体中提取的DNA进行了分析4,5,但后者发现了共生体中藻的DNA对整个PCR产物的干扰作用会影响满江红属本身系统分类.

胃伤害性刺激诱导大鼠脑干星形胶质细胞GFAP蛋白表达及其与神经元的关系 总被引:7,自引:0,他引:7
研究大鼠在福尔马林诱发胃伤害性刺激时脑干内星形胶质细胞及神经元的变化。应用免疫组织化学三重标记法在脑原位切片同时显示脑干内Fos蛋白,胶质原纤维酸性蛋白(GFAP),酪氨酸羟化酶(TH)的表达,结果显示:1、在福尔马林诱发胃伤害性刺激后,脑干胶质细胞GFAP表达阳性,并表现出明显的核团或亚核定位特点,在延髓内脏带(MVZ0,中缝大核(RMg),蓝斑(LC),臂旁外侧核(LPB),中缝背核(DR),中脑导水管周围灰质腹外侧区(vlPAG),上丘中灰层(IngSC)等脑区有较多的Fos阳性细胞,而且Fos阳性表达的分布与上述GFAP阳性分布基本一致;2、MVZ,LC,DR,vlPAG等部位有大量Fos及TH双标阳性神经元,周围有密集的GFAP阳性细胞;3、随着刺激后存活时间的变化,GFAP与Fos阳性细胞的反应均经历逐渐升高后又渐降低直至消失的变化。结果表明:上述核团的神经元和星形胶质细胞可能同时参与了内脏痛及其调节过程。 相似文献

目的:比较连续性血液净化(continuous blood purification,CBP)和血液透析(haemodialysis,HD)治疗尿毒症脑病的疗效.方法:70例尿毒症脑病(uremic encephalopathy,UE)患者随机分成连续性血液净化(CBP)组(n=35)和血液透析(HD)组(n=35),分别给予相应治疗.观察缓解率、平均动脉压、肾功能、电解质及血气指标等.结果:1周缓解率分别为100%(CBP组)和65.7%(HD组)(P<0.05);两组的BUN,Scr,K+,Na+,Cl-和β2-MG均显著降低(P<0.05),但治疗后CBP组的BUN,Scr和β2-MG显著低于HD组(P<0.05);CBP组的HC03-和pH较治疗前显著降低(P<0.05),而HD组中HC03-和pH治疗前后无显著改变(P>0.05).CBP组治疗前后平均动脉压变化无统计学差异(P>0.05),HD组低血压发生率为13.7%,高血压发生率为15.4%.结论:CBP治疗能很好清除BUN、Scr和β2-MG,纠正电解质及酸碱紊乱,维持患者血液动力学,具有较好的临床疗效. 相似文献

Background
It was well known that the clinical use of chemotherapeutic drugs is restricted by severe adverse reactions and drug resistances. Thus it is necessary to figure out a strategy to increase the specific anti-tumor efficiency of chemotherapeutic drugs. Apigenin, a kind of flavonoids, has been reported to possess anticancer activities with very low cytotoxicity to normal tissue.Methodology/Principal Findings
Our results from cell viability assay, western-blots and TdT-mediated dUTP-biotin nick end labeling (TUNEL) assay demonstrated the synergistic pro-apoptotic effects of a low dose of apigenin and paclitaxel in human cancer cell lines. To analyze the underlying mechanism, we examined reactive oxygen species (ROS) staining after cells were treated with a combination of apigenin and paclitaxel, or each of them alone. Data from flow-cytometry showed that superoxides but not reduction of peroxides accumulated in HeLa cells treated with apigenin or a combination of apigenin and paclitaxel. Apigenin and paclitaxel-induced HeLa cell apoptosis was related to the level of ROS in cells. We further evaluated activity and protein level of superoxide dismutase (SOD). Apigenin significantly inhibited SOD activity but did not alter the SOD protein level suggesting that apigenin promoted ROS accumulation through suppressing enzyme activity of SOD. Addition of Zn2+, Cu2+ and Mn2+ to cell lysates inhibited apigenin''s effects on SOD activity. At the same time, data from caspase-2 over-expression and knocked-down experiments demonstrated that caspase-2 participated in apigenin and paclitaxel-induced HeLa cell apoptosis.Conclusions/Significance
Taken together, our study demonstrated that apigenin can sensitize cancer cells to paclitaxel induced apoptosis through suppressing SOD activity, which then led to accumulation of ROS and cleavage of caspase-2, suggesting that the combined use of apigenin and paclitaxel was an effective way to decrease the dose of paclitaxel taken. 相似文献876.
Pathway enrichment analysis represents a key technique for analyzing high-throughput omic data, and it can help to link individual genes or proteins found to be differentially expressed under specific conditions to well-understood biological pathways. We present here a computational tool, SEAS, for pathway enrichment analysis over a given set of genes in a specified organism against the pathways (or subsystems) in the SEED database, a popular pathway database for bacteria. SEAS maps a given set of genes of a bacterium to pathway genes covered by SEED through gene ID and/or orthology mapping, and then calculates the statistical significance of the enrichment of each relevant SEED pathway by the mapped genes. Our evaluation of SEAS indicates that the program provides highly reliable pathway mapping results and identifies more organism-specific pathways than similar existing programs. Effects of ammonium on the photosynthetic recovery of Nostoc flagelliforme Berk. et M. A. Curtis were assayed when being rehydrated in low‐K+ or high‐K+ medium. Its photosynthetic recovery was K+ limited after 3 years of dry storage. The potassium absorption of N. flagelliforme reached the maximum after 3 h rehydration in low‐K+ medium but at 5 min in high‐K+ medium. The K+ content of N. flagelliforme rehydrated in high‐K+ medium was much higher than that in low‐K+ medium. The maximal PSII quantum yield (Fv/Fm) value of N. flagelliforme decreased significantly when samples were rehydrated in low‐K+ medium treated with 5 mM NH4Cl. However, the treatment of 20 mM NH4Cl had little effect on its Fv/Fm value in high‐K+ medium. The relative Fv/Fm 24 h EC50 (concentration at which 50% inhibition occurred) value of NH4+ in high‐K+ medium (64.35 mM) was much higher than that in low‐K+ medium (22.17 mM). This finding indicated that high K+ could alleviate the inhibitory action of NH4+ upon the photosynthetic recovery of N. flagelliforme during rehydration. In the presence of 10 mM tetraethylammonium chloride (TEACl), the relative Fv/Fm 24 h EC50 value of NH4+ was increased to 46.34 and 70.78 mM, respectively, in low‐K+ and high‐K+ media. This observation suggested that NH4+ entered into N. flagelliforme cells via the K+ channel. Furthermore, NH4+ could decrease K+ absorption in high‐K+ medium. 相似文献

首次采用反相中压制备色谱从芦荟中一次分离制备得到芦荟苷A、B以及异芦荟色苷D。以库拉索芦荟丙酮粗提物为原料,采用中压制备色谱系统:SCO色谱柱(40 cm×26 cm,30~50μm),流动相甲醇-0.5%乙酸水(33∶67,V/V),流速20 mL/min,等度洗脱方式,柱温室温,检测波长254 nm,收集波长356 nm对芦荟样品进行分离制备,得到三种化合物单体。经高效液相色谱、紫外、红外、质谱及核磁共振等方法分析表明所得到的三种化合物分别是异芦荟色苷D、芦荟苷A和芦荟苷B,其纯度分别达到了98.0%,96.0%和98.9%。该方法简便,产品质量高,一次制备可以得到多种单体,为芦荟成分的测定与药理活性的研究提供了条件。 相似文献

肠道病毒71型作为引起儿童群体常见传染性手足口病(HFMD)的主要病原,具有导致少量感染个体出现脑炎等神经系统病变以及相关心肺功能衰竭的病理学特性.因此其预防性疫苗的研发具有重要的公共卫生意义.在前期工作的基础上,一种EV71灭活病毒疫苗(人二倍体细胞)在本研究中基于恒河猴婴猴模型进行了相应的免疫保护性分析.以160EU剂量对2~3月龄婴猴进行0,4周免疫后,动物在第4周接受了剂量为10。~CCID50的病毒经呼吸道的攻击.对病毒攻击后动物在14天内的临床症状、血液生物学、器官病原学分布以及病理学检测的动态观察表明,经疫苗免疫的动物未出现对照动物所具有的特征性临床表现,其血液生物学及病理学检测均无异常.同时,器官病原学分布亦呈阴性.结合动物中和抗体的明确增长及对照动物的综合表现分析,本文的工作证实了该EV71灭活病毒疫苗(人二倍体细胞)在恒河猴婴猴体内的免疫保护性. 相似文献
