To improve ethanol production in Saccharomyces cerevisiae, two yeast strains were constructed. In the mutant, KAM-4, the GPD1 gene, which encodes a glycerol 3-phosphate dehydrogenase of S. cerevisiae to synthesize glycerol, was deleted. The mutant KAM-12 had the GLT1 gene (encodes glutamate synthase) placed under the PGK1 promoter while harboring the GPD1 deletion. Notably, overexpression of GLT1 by the PGK1 promoter along with GPD1 deletion resulted in a 10.8% higher ethanol production and a 25.0% lower glycerol formation compared to the wild type in anaerobic fermentations. The growth rate of KAM-4 was slightly lower than that of the wild type under the exponential phase whereas KAM-12 and the wild type were indistinguishable in the biomass concentration at the end of growth period. Meanwhile, dramatic reduction of formation of acetate and pyruvic acid was observed in all the mutants compared to the wild type.  相似文献

目的 探讨左归丸含药血清对化疗损伤性颗粒细胞和膜细胞的影响及作用机制。方法 制备左归丸含药血清,培养大鼠卵巢颗粒细胞和膜细胞,使用磷酰胺氮芥造模分组后给药。CCK-8法测定颗粒细胞和膜细胞存活率,实时荧光定量PCR法(RT-PCR)及蛋白质免疫印迹法(Western blot)分别检测卵巢自噬启动因子Beclin-1、微管结合蛋白轻链3(LC3B)、自噬受体蛋白p62、凋亡蛋白Bax、Caspase3在转录水平和翻译水平上的表达。结果 10%左归丸含药血清对于细胞存活的挽救率最高。Beclin-1、LC3B、Bax、Caspase3在磷酰胺氮芥作用的颗粒细胞和膜细胞中,相对于空白对照组有高表达(P<0.05),10%左归丸含药血清可下调上述蛋白质在模型组中的表达(P<0.05);然而受体蛋白p62较空白对照组升高(P<0.05),10%左归丸含药血清可上调模型组p62的表达(P<0.05)。此外,在颗粒细胞实验组中,激活或抑制自噬途径后,自噬相关蛋白的表达在发生相应改变的同时,凋亡相关蛋白的表达也会发生相应改变。结论 磷酰胺氮芥可通过促进凋亡、激活自噬/溶酶体降解途径的机制损伤颗粒细胞和膜细胞。10%左归丸含药血清能缓解由此带来的损伤,同时影响了颗粒细胞和膜细胞自噬和凋亡过程。在磷酰胺氮芥损伤颗粒细胞的过程和10%左归丸含药血清缓解其损伤过程中均存在自噬与凋亡串流(cross-talk)。  相似文献

Background

The aim of this study was to evaluate a possible association between malignancy and anxiety disorders (AD) in Taiwan.

Methods

We employed data from the National Health Insurance system of Taiwan. The AD cohort contained 24,066 patients with each patient randomly frequency matched according to age and sex with 4 individuals from the general population without AD. Cox''s proportional hazard regression analysis was conducted to estimate the influence of AD on the risk of cancer.

Results

Among patients with AD, the overall risk of developing cancer was only 1% higher than among subjects without AD, and the difference was not significant (hazard ratio [HR] = 1.01, 95% confidence interval [95% CI] = 0.95–1.07). With regard to individual types of cancer, the risk of developing prostate cancer among male patients with AD was significantly higher (HR = 1.32, 95% CI = 1.02–1.71). On the other hand, the risk of cervical cancer among female patients with AD was marginally significantly lower than among female subjects without AD (HR = 0.72, 95% CI = 0.51–1.03).

Limitations

One major limitation is the lack of information regarding the life style or behavior of patients in the NHI database, such as smoking and alcohol consumption.

Conclusions

Despite the failure to identify a relationship between AD and the overall risk of cancer, we found that Taiwanese patients with AD had a higher risk of developing prostate cancer and a lower risk of developing cervical cancer.  相似文献

Background

Controversy still exists regarding whether alendronate (ALN) use increases the risk of esophageal cancer or breast cancer.

Methods

This paper explores the possible association between the use of oral ALN in osteoporosis patients and subsequent cancer risk using the National Health Insurance (NHI) system database of Taiwan with a Cox proportional-hazard regression analysis. The exposure cohort contained 5,624 osteoporosis patients used ALN and randomly frequency-matched by age and gender of 3 osteoporosis patients without any kind of anti-osteoporosis drugs in the same period.

Results

For a dose ≥1.0 g/year, the risk of developing overall cancer was significantly higher (hazard ratio: 1.69, 95% confidence ratio: 1.39–2.04) than in osteoporosis patients without any anti-osteoporosis drugs. The risks for developing liver, lung, and prostate cancers and lymphoma were also significantly higher than in the control group.

Conclusions

This population-based retrospective cohort study did not find a relationship between ALN use and either esophageal or breast cancer, but unexpectedly discovered that use of ALN with dose ≥1.0 g/year significantly increased risks of overall cancer incidence, as well as liver, lung, and prostate cancers and lymphoma. Further large population-based unbiased studies to enforce our findings are required before any confirmatory conclusion can be made.  相似文献

The 2009 influenza pandemic provided an opportunity to observe dynamic changes of the hemagglutinin (HA) and neuraminidase (NA) of pH1N1 strains that spread in two metropolitan areas -Taipei and Kaohsiung. We observed cumulative increases of amino acid substitutions of both HA and NA that were higher in the post–peak than in the pre-peak period of the epidemic. About 14.94% and 3.44% of 174 isolates had one and two amino acids changes, respective, in the four antigenic sites. One unique adaptive mutation of HA2 (E374K) was first detected three weeks before the epidemic peak. This mutation evolved through the epidemic, and finally emerged as the major circulated strain, with significantly higher frequency in the post-peak period than in the pre-peak (64.65% vs 9.28%, p<0.0001). E374K persisted until ten months post-nationwide vaccination without further antigenic changes (e.g. prior to the highest selective pressure). In public health measures, the epidemic peaked at seven weeks after oseltamivir treatment was initiated. The emerging E374K mutants spread before the first peak of school class suspension, extended their survival in high-density population areas before vaccination, dominated in the second wave of class suspension, and were fixed as herd immunity developed. The tempo-spatial spreading of E374K mutants was more concentrated during the post–peak (p = 0.000004) in seven districts with higher spatial clusters (p<0.001). This is the first study examining viral changes during the naïve phase of a pandemic of influenza through integrated virological/serological/clinical surveillance, tempo-spatial analysis, and intervention policies. The vaccination increased the percentage of E374K mutants (22.86% vs 72.34%, p<0.001) and significantly elevated the frequency of mutations in Sa antigenic site (2.36% vs 23.40%, p<0.001). Future pre-vaccination public health efforts should monitor amino acids of HA and NA of pandemic influenza viruses isolated at exponential and peak phases in areas with high cluster cases.  相似文献

主要嗅觉表皮(main olfactory epithelium, MOE)是哺乳动物感知气味分子的主要嗅觉器官。在MOE组织内,大多数嗅觉神经元通过cAMP信号传导通路感知气味信息。作为嗅觉cAMP信号通路的主要成员之一,腺苷酸环化酶3(adenylyl cyclase 3, ac3)基因敲除小鼠嗅觉探测功能丧失。除cAMP信号传导通路外,MOE内AC3相关因子AC2和AC4,以及肌醇1,4,5-三磷酸(inositol 1,4,5-trisphosphate,IP3)信号通路和Sonic Hedgehog(Shh)信号通路均有表达。然而,敲除ac3是否会对ac2和ac4以及IP3和Shh信号通路成员产生影响,尚不清楚。本文以AC3缺失(AC3-/-)及其野生型小鼠(AC3+/+)MOE为材料,采用实时荧光定量PCR(qRT-PCR)和免疫荧光组织化学方法,发现AC3缺失后,MOE内的ac2和ac4,以及IP3信号通路中的IP3受体ip3r1及钙调蛋白calm1和calm2表达水平均明显降低。Shh信号通路中的受体patched(ptch)与smoothened(smo)、以及核转录因子gli1与gli2的表达也受到了影响。总之,AC3基因缺失不但导致小鼠MOE组织中cAMP信号通路受损,同时AC3相关因子,IP3信号通路和Shh信号通路的传导也受到抑制。本文对于阐明AC3基因敲除小鼠嗅觉丧失的原因及其嗅觉探测机制具有重要启示作用。  相似文献

动物对植物的采食会刺激植物进行补偿性更新生长, 克隆整合效应能够通过分株之间的物质传输增强克隆植物的这种补偿生长。现今对克隆整合效应在箭竹(Fargesia)补偿更新中的作用仍未得到全面认识。2011年10月到2012年11月, 设立了糙花箭竹(Fargesia scabrida)和缺苞箭竹(F. denudata)各40个样方, 分别进行不剪除样方内分株和剪除样方内分株数量的25%、50%、75%四种模拟采食干扰处理, 并将样方四周的根状茎切断或保持连接。从2012年6月起观测并统计了箭竹分株种群的累积出笋率、总出笋率、补充率, 以及新生分株的株高、基径和单株生物量。结果表明: (1)在不剪除分株的样方, 切断根状茎连接显著增加了糙花箭竹的出笋率和补充率, 但降低了新生分株的株高和单株生物量, 也显著降低了缺苞箭竹的出笋率和补充率; (2)保持根状茎连接时, 25%的剪除强度仅仅降低了糙花箭竹新生分株的单株生物量; 同样在保持根状茎连接的条件下, 25%、50%的剪除强度使缺苞箭竹种群的补充率有所降低, 而切断根状茎后缺苞箭竹在25%的剪除强度下的分株补充率反而升高; (3) 75%的剪除强度并未影响两种箭竹新生分株数量更新, 但造成新生分株质量显著下降; 切断根状茎连接显著降低了糙花箭竹的新生分株的株高和基径, 对缺苞箭竹影响不显著。实验证明克隆整合影响了两种箭竹新生分株的萌发、存活和生长, 但不是两种箭竹进行补偿更新的主要机制, 仅在糙花箭竹分株种群受到重度采食干扰后的更新中才起到明显的促进作用; 两种箭竹均能在50%的剪除强度下通过补偿生长恢复种群的稳定, 75%的剪除强度则会造成箭竹新生分株质量的下降。  相似文献
