Preparation of gradually componential metal electrode on solution-casted Nafion membrane

A typical ionic polymer-metal composite consists of a thin perfluorinated ionomer membrane and noble metal electrodes plated on both surfaces. It undergoes a large bending motion when an electric field is applied hence exhibits deformation by a certain amount of cation. With proper arrangement and package, a great number of "smart devices" are anticipated. In this study, a solution-cast route was used to prepare the electro-active polymer membrane and platinum electrodes were fabricated onto the membrane through electro-less plating. The ionic polymer used is the commercial Nafion, the perfluoro-sulfonated ionomer membrane, developed by DuPont Co. Nafion membrane was cast by the solution-casting route and then loaded with different weights simultaneously. The optimized processing conditions, membrane properties and electrodes behaviors were investigated. The results of shift in WAXD peaks showed that weights delineated the crystallinity of the solution-cast membranes. The number and size of the crystalline domains of solution-cast membrane decrease as studied by SAXS. The Young's modulus of solution-cast membrane decreases as increasing weight because of the loss of crystallinity (180-140 MPa). A finely dispersed platinum particle deeper and gradient penetrating within the near-boundary region with a smaller average particle size and more uniform distribution could be obtained through a reverse electro-less plating. Its surface roughness is 3 nm comparing to 52 nm of a typical process. But its surface resistance is too high (3.5 Omega) to activate the bending motion. To solve this problem, we coated the second Pt electrodes by a typical electro-less plating, and the resistance decreased to 0.7 Omega. The results depicted that the fabricated IPMC shows longer bending lifetime than typical IPMC. In a 0.09% NaCl solution, the device was able to vibrate for 8h under a 5 V, 0.1 Hz actuation.     

Effects of CDB-4022 on Leydig cell function in adult male rats

CDB-4022, an indenopryridine, suppresses spermatogenesis and decreases inhibin secretion in adult male rats. In the present study, we investigated the effects of CDB-4022 on Leydig cell function. A single oral dose of CDB-4022 (2.5 mg/kg) resulted in a 2-fold decrease in serum testosterone levels after 7 days that was paralleled by a decrease in Cyp17a1 mRNA and protein levels and 17alpha hydroxylase enzymatic activity compared with vehicle-treated rats. Consistent with the lower serum testosterone levels, pituitary Lhb and Fshb mRNA levels were increased 3.2- and 2.3-fold, respectively, by CDB-4022 treatment. Ultrastructural analysis of pituitary gonadotrophs showed distended endoplasmic reticulum (ER) and fewer secretory granules in CDB-4022-treated rats, characteristic of enhanced secretory activity. Conversely, CDB-4022 increased serum progesterone levels, testicular Star mRNA and protein expression, and the number of Leydig cells per testis. Serum inhibin B levels were undetectable in CDB-4022-treated rats, while serum activin A levels were similar to controls, indicating that the CDB-4022-treated rats have an elevated activin A:inhibin B ratio. In the presence of hCG stimulation, activin A directly suppressed testosterone secretion but enhanced progesterone secretion from rat Leydig cell primary cultures. Likewise, treatment of MA-10 cells with activin A was found to enhance cAMP-stimulated progesterone secretion and STAR expression. Together, our data indicate that CDB-4022 treatment inhibits CYP17A1 and stimulates STAR expression, thereby decreasing testosterone but increasing progesterone production. We propose that unopposed actions of activin A most likely contribute to the steroid profile in rats after CDB-4022 treatment. Our findings establish CDB-4022 as a new model to examine intratesticular control mechanisms that modulate Leydig cell gene expression and function.     

Gain of C-Ala enables AlaRS to target the L-shaped tRNAAla

Unlike many other aminoacyl-tRNA synthetases, alanyl-tRNA synthetase (AlaRS) retains a conserved prototype structure throughout biology. While Caenorhabditis elegans cytoplasmic AlaRS (CeAlaRS_c) retains the prototype structure, its mitochondrial counterpart (CeAlaRS_m) contains only a residual C-terminal domain (C-Ala). We demonstrated herein that the C-Ala domain from CeAlaRS_c robustly binds both tRNA and DNA. It bound different tRNAs but preferred tRNA^Ala. Deletion of this domain from CeAlaRS_c sharply reduced its aminoacylation activity, while fusion of this domain to CeAlaRS_m selectively and distinctly enhanced its aminoacylation activity toward the elbow-containing (or L-shaped) tRNA^Ala. Phylogenetic analysis showed that CeAlaRS_m once possessed the C-Ala domain but later lost most of it during evolution, perhaps in response to the deletion of the T-arm (part of the elbow) from its cognate tRNA. This study underscores the evolutionary gain of C-Ala for docking AlaRS to the L-shaped tRNA^Ala.     

Lack of inhibitory effect of aprotinin and bacitracin on the spinal release of Met-enkephalin induced by intraventricular beta-endorphin

We have previously reported that administration of beta-endorphin intraventricularly in the rat increases the release of immunoreactive Met-enkephalin from the spinal cord. To further eliminate the possibility that the increase in Met-enkephalin might arise from the degradation of beta-endorphin injected, the effect of peptidase inhibitors, aprotinin and bacitracin, on the spinal fluid content of Met-enkephalin released by intraventricular beta-endorphin was studied using an intrathecal perfusion technique in urethane anesthetized rats. Inhibition of peptidases by intraventricular aprotinin and bacitracin did not decrease nor enhance the increased content of Met-enkephalin in the spinal perfusate produced by intraventricular beta-endorphin. The result indicates that the Met-enkephalin arises from neuronal release in the spinal cord rather than from degradation of the beta-endorphin injected intraventricularly.     

Induction and growth of mammary tumors after superior cervical ganglionectomy in sighted and blinded-anosmic rats

Female rats were subjected to superior cervical ganglionectomy (Gx), blinding and anosmia (BAs) or combined procedures (BAsGx). Onset and growth of dimethylbenz(a)anthracene (DBMA)-induced mammary tumors was studied in these animals and compared to tumorigenesis in intact control rats. Carcinostatic effects were present in all surgically altered animals, as evidenced by a trend toward reduced tumor incidence, reduced final tumor mass, and a significant reduction in mean number of tumors in Gx and BAsGx rats, and increased regression of tumors in BAs rats compared to intact group. Reduced tumorigenesis was paralleled by a trend toward either an increase (BAs) or a decrease (Gx and BAsGx) in the activity of pineal hydroxyindole-O-methyltransferase (HIOMT) compared to intact group. In addition, BAs and BAsGx animals showed a significant reduction in body weight. These results suggest that Gx reduces mammary tumorigenesis in both sighted and BAs rats. They further confirm the findings of others on reduced mammary tumorigenesis in BAs rats. Possible involvement of multiple carcinostatic mechanisms in different animal models is discussed.     

A Supplement to the Chloranthaceae of China

In the present paper the longstanding confusion of three uncertain species of Chloranthus, C. monostachys R. Br., C. pernyanus Solms-Laub. and C. kiangsiensis Metcalf is clarified. In addition to the new record of C. nervosus Coll. et Hemsl. to the flora of China, the distributional ranges of Sarcandra hainanensis (Pei) Swamy et Bailey and Hedyosmum orientale Merr. et Chun are also found to be much bigger than previously known.
