草菇培养物中粗三萜和黄酮含量及抗氧化抗肿瘤活性研究

采用液体摇瓶法培养草菇菌丝体,并用不同有机溶剂对培养液及菌丝体中的代谢成分分别进行分离提取,获得了不同来源的次生代谢提取物。对各提取物的成分分析表明,石油醚相、乙酸乙酯相和乙醇相提取物中均含有粗三萜和黄酮类物质,但菌丝体提取物中粗三萜和黄酮含量明显高于培养液提取物中的含量。石油醚抽提菌丝体获得的提取物中粗三萜含量最高,达17%,而乙酸乙酯抽提菌丝体获得的提取物中黄酮含量最高,达9.31%。抗氧化活性检测结果显示,石油醚相、乙酸乙酯相、乙醇相中的代谢提取物均有较高的抗氧化活性,但乙酸乙酯相提取物的抗氧化活性明显高于石油醚相提取物,具最高抗氧化活性的提取物分别来自乙酸乙酯、乙醇对菌丝体的抽提物。MTT法检测各提取物对胃癌细胞增殖的抑制作用,结果显示各组分均有较高的抗肿瘤活性,且抗肿瘤活性与提取物浓度存在明显的量效关系。     

悉生蚕的研究与应用

悉生蚕是指蚕体内外栖息的其他生物体是已知的蚕,不仅培育方法较哺乳动物简单,且彻底克服蚕的季节性和地域性饲育限制,为蚕在实验动物科学中的广泛应用打下基础。随着现代生物技术的发展,悉生蚕作为生物反应发生器或转基因工程动物的应用已逐步进入实用化研究阶段。     

Immortalization and characterization of mouse floxed Bmp2/4 osteoblasts

Generation of a floxed Bmp2/4 osteoblast cell line is a valuable tool for studying the modulatory effects of Bmp2 and Bmp4 on osteoblast differentiation as well as relevant molecular events. In this study, primary floxed Bmp2/4 mouse osteoblasts were cultured and transfected with simian virus 40 large T-antigen. Transfection was verified by polymerase chain reaction (PCR) and immunohistochemistry. To examine the characteristics of the transfected cells, morphology, proliferation and mineralization were analyzed, expression of cell-specific genes including Runx2, ATF4, Dlx3, Osx, dentin matrix protein 1, bone sialoprotein, osteopontin, osteocalcin, osteonectin and collagen type I was detected. These results show that transfected floxed Bmp2/4 osteoblasts bypassed senescence with a higher proliferation rate, but retain the genotypic and phenotypic characteristics similar to the primary cells. Thus, we for the first time demonstrate the establishment of an immortalized mouse floxed Bmp2/4 osteoblast cell line.     

淋洗后生活垃圾堆肥建植高羊茅草坪生理生态响应

以不同淋洗方式处理后的生活垃圾堆肥为基质进行草坪植物培植,研究了高羊茅的生理生态响应。结果表明,淋洗处理堆肥中高羊茅的发芽率及株高等生长指标均优于未淋洗的原堆肥对照组,其中水肥淋洗比例为1∶1的处理中,高羊茅的最大发芽率比对照提高89.6%。通过对超氧化物歧化酶（SOD）、过氧化物酶（POD）、过氧化氢酶（CAT）3种保护酶的活性及脯氨酸和丙二醛（MDA）含量等指标的测定可以看出,经淋洗处理后的堆肥基质对草坪植物的胁迫程度均小于原堆肥,更有利于草坪植物的正常生长。综合高羊茅生长指标及生理指标来考虑,堆肥处理的最佳淋洗比例为水∶堆肥的质量比为2∶1。     

肠道病毒71型（EV71）对ICR小鼠的感染

目的研究肠道病毒71型经不同途径感染不同日龄ICR小鼠后的感染状况,了解肠道病毒71型的感染特点,为了解EV71小鼠感染机制和模型制备提供实验信息和技术支撑。方法分别通过口腔途径、颅腔途径、肌肉途径及腹腔途径感染1日龄、7日龄及3~4周龄SPF级ICR小鼠,定期安乐动物,采集各器官组织进行病原学诊断,确定EV71病毒感染情况;同时建立一步RT-PCR、病毒分离、IFA及IEA等方法。结果经腹腔途径感染成年鼠出现竖毛、弓背、消瘦症状,其他各途径感染小鼠感染后未见竖毛、弓背、觅食减少、体重减轻、精神呆滞及神经系统症状。颅腔注射3~4周龄ICR小鼠能在脑组织检测到病毒RNA,腹腔注射和肌肉注射1日龄乳鼠能在肌肉组织和肠道检测到病毒RNA,其中,肌肉组织病毒分离可检测到活病毒。本研究同时建立了分子生物学、血清学方法,为今后研究其它适合EV71的动物模型奠定了基础。结论临床分离的EV71毒株通过口腔接种、颅腔、肌肉、腹腔注射途径感染1日龄、7日龄及3~4周龄SPF级ICR小鼠的疾病程度和病毒检出不同,ICR乳鼠及成年鼠可作为该病毒感染机制、病毒体内分布等基础研究,但用作EV71动物模型应用,感染程度尚不十分理想。     

灵芝多糖对顺铂引起的呕吐具抑制作用

以雄性昆明小鼠为实验材料,研究灵芝多糖对顺铂引起呕吐的抑制作用。实验分为5组,生理盐水+生理盐水组:腹腔注射0.9% NaCl两次,格拉司琼+顺铂组:腹腔注射格拉司琼和顺铂,生理盐水+顺铂组:腹腔注射0.9% NaCl和顺铂,灵芝多糖+生理盐水组:腹腔注射灵芝多糖和0.9% NaCl,灵芝多糖+顺铂组:腹腔注射灵芝多糖和顺铂,两种成分的注射间隔为30min。上述处理每天1次,连续5d,比较各组小鼠对高岭土的摄取量和脑内Fos蛋白表达水平。结果显示:顺铂可增加小鼠高岭土摄取量和脑内Fos蛋白表达水平,而格拉司琼和灵芝多糖可减少小鼠对高岭土的摄取量并降低脑内Fos蛋白表达水平。因此认为灵芝多糖可有效抑制顺铂引起的恶心呕吐。     

BOD: a customizable bioinformatics on demand system accommodating multiple steps and parallel tasks

The integration of bioinformatics resources worldwide is one of the major concerns of the biological community. We herein established the BOD (Bioinformatics on demand) system to use Grid computing technology to set up a virtual workbench via a web-based platform, to assist researchers performing customized comprehensive bioinformatics work. Users will be able to submit entire search queries and computation requests, e.g. from DNA assembly to gene prediction and finally protein folding, from their own office using the BOD end-user web interface. The BOD web portal parses the user's job requests into steps, each of which may contain multiple tasks in parallel. The BOD task scheduler takes an entire task, or splits it into multiple subtasks, and dispatches the task or subtasks proportionally to computation node(s) associated with the BOD portal server. A node may further split and distribute an assigned task to its sub-nodes using a similar strategy. In the end, the BOD portal server receives and collates all results and returns them to the user. BOD uses a pipeline model to describe the user's submitted data and stores the job requests/status/results in a relational database. In addition, an XML criterion is established to capture task computation program details.     

Effects of a verbenachalcone derivative on neurite outgrowth, inhibition of caspase induction and gene expression

A verbenachalcone derivative was synthesized and shown to protect N2a cells from caspase induction caused by serum starvation and to enhance the effect of NGF on neurite outgrowth in PC12 cells. As an initial investigation of the compound's mechanism(s) of action, we performed differential gene expression profiling in PC12 cells using oligonucleotide ( approximately 10,000 gene probes) microarrays. Gene expression patterns were compared in the presence of NGF (2 and 50 ng/mL) and NGF (2 ng/mL) plus the verbenachalcone derivative. Ten genes were significantly (2-fold; p0.05) up-regulated and seven genes were significantly down-regulated in the presence of the compound. These results were independently validated by quantitative real-time PCR for a subset of genes (cathepsin L, sigma-1 receptor and protein tyrosine phosphatase receptor type R). These genes or their protein products may represent useful therapeutic targets for treating neurodegeneration, such as Alzheimer's disease.     

内蒙古5个民族12对性状的基因频率

报道了内蒙古地区鄂温克、鄂伦春、达斡尔、蒙古族和汉族12对遗传性状的基因频率,并进行民族间基因频率的比较。结果显示：内眦褶性状的民族间差异较大,叠舌性状次之,利手和鼻梁侧面观性状的民族间差异较小。 Abstract:The gene frequency of 12 characters was reported in Ewenki,Oroqen,Daur,Mongol and Han nationalities in Inner Mongolia,and compared among these nationalities.The result indicated that the difference of Mongoloid fold among nationalities was significant,followed by the Folding tongue,while the difference of Handedness and Nasal profile was relatively insignificant.     

Comparative genome mapping among Populus adenopoda, P. alba, P. deltoides, P. euramericana and P. trichocarpa

Among the genus Populus, the sections Populus (white poplar), Aigeiros Duby (black poplar) and Tacamahaca Spach contain many tree species of economical and ecological important properties. Two parental maps for the inter-specific hybrid population of Populus adenopoda × P. alba (two species of Populus section) were constructed based on SSR and SRAP markers by means of a two-way pseudo-test cross mapping strategy. The same set of SSR markers developed from the P. trichocarpa (belonging to Tacamahaca section) genome which were used to construct the maps of P. deltoides and P. euramericana (two species of Aigeiros section) was chosen to analyze the genotype of the experimental population of P. adenopoda × P. alba. Using the mapped SSR markers as allelic bridges, the alignment of the white and black poplar maps to each other and to the P. trichocarpa physical map was conducted. The alignment showed high degree of marker synteny and colinearity and the closer relationship between Aigeiros and Tacamahaca sections than that of Populus and Tacamahaca. Moreover, there was evidence for the chromosomal duplication and inter-chromosomal reorganization involving some poplar linkage groups, suggesting a complicated course of fission or fusion in one of the lineages. A poplar consensus map based on the comparisons could be constructed will be useful in practical applications including marker assisted selection.
