Heparins/heparan sulfates modulate the function of proteins and cell membranes in numerous biological systems including normal and disease processes in humans. Heparin has been used for many years as an anticoagulant, and anticoagulant heparin-mimetics were developed several decades ago by chemical sulfation of non-mammalian polysaccharides, e.g., an antithrombotic sulfated xylan. This pharmaceutical, which comprises a mixture of sulfated oligoxylans, also mimics most other biological actions of natural heparins in vitro, including inhibition of the human immunodeficiency virus, but the molecular basis for these actions has been unclear. Here, numerous Components of the sulfated oligoxylan mixture were isolated and when bioassayed in the case of anti-HIV-1 infectivity revealed that a structural specificity underlines the capacity of sulfated xylan to inhibit HIV-1, rather than a non-specific mechanism. Components were isolated by chromatographic fractionation through Bio-Gel P10 in 0.5M ammonium bicarbonate. This fractionation revealed an elution range associated with apparent molecular weights of 22 000 to <1500 relative to standard heparin and heparan sulfates and newly prepared sulfated oligosaccharide standards. Components were characterized by metachromatic absorption spectroscopy, ultracentrifugation, GlcA analysis, and potency against HIV-1 infectivity, both in the tetrazolium cytotoxicity assay and in syncytium-forming assays, in CD4-lymphocytes. Structural specificity was indicated by the differential potencies exhibited by the Components: Highest activity (cytotoxicity) was exhibited by Components in the chromatographic region 5500 in mass (50% effective (inhibitory) concentration=0.5–0.7 g ml–1 in the first fractionation series, and 0.1–0.5 g ml–1 in a second series). The potency declined sharply below 5400 in mass, but with an exception; a second structure exhibiting relatively high potency eluted among low-mass oligosaccharides which had an average size of a nonomer. Components displayed differential potencies also against the syncytium-forming infectivity of HIV-1. The high potency against syncytium-formation was retained by Components down to a minimum size of about 4500 in mass, smaller than the 5400 required above. One in ten of the 1,4-linked xyloses in the native xylan are substituted with a monomeric 1,2 DGlcA branch. We have speculated that pharmaceutical actions of sulfated xylan might be related to structures involving the -D linked substituents and this was examined using a space-filling model of a sulfated octaxylan and by analyses of Components for GlcA content. Understanding structure/function relations in the heparin-like actions of these agents would be of general significance for the careful examination of their potential clinical usefulness in many human processes modulated by heparins, including AIDS. 相似文献

Plant mating systems are known to influence population genetic structure because pollen and seed dispersal are often spatially restricted. However, the reciprocal outcomes of population structure on the dynamics of polymorphic mating systems have received little attention. In gynodioecious sea beet (Beta vulgaris ssp. maritima), three sexual types co‐occur: females carrying a cytoplasmic male sterility (CMS) gene, hermaphrodites carrying a non‐CMS cytoplasm and restored hermaphrodites that carry CMS genes and nuclear restorer alleles. This study investigated the effects of fine‐scale genetic structure on male reproductive success of the two hermaphroditic forms. Our study population was strongly structured and characterized by contrasting local sex‐ratios. Pollen flow was constrained over short distances and depended on local plant density. Interestingly, restored hermaphrodites sired significantly more seedlings than non‐CMS hermaphrodites, despite the previous observation that the former produce pollen of lower quality than the latter. This result was explained by the higher frequency of females in the local vicinity of restored (CMS) hermaphrodites as compared to non‐CMS hermaphrodites. Population structure thus strongly influences individual fitness and may locally counteract the expected effects of selection, suggesting that understanding fine scale population processes is central to predicting the evolution of gender polymorphism in angiosperms. 相似文献

A multiyear study of pallid sturgeon distribution and relative abundance was conducted in the lower and middle Mississippi river (LMR and MMR, respectively). The LMR and MMR comprise the free‐flowing Mississippi River extending 1857 river kilometers (rkm) from its mouth at the Gulf of Mexico upstream to the mouth of the Missouri River. A total of 219 pallid sturgeon and 6018 shovelnose sturgeon was collected during the periods 1996–1997 and 2000–2006. Trotlines baited with worms were the primary collecting gear. The smallest pallid sturgeon captured on trotlines was 405 mm FL and the largest was 995 mm FL. Mean size of pallid sturgeon was statistically smaller in the Mississippi River below the Atchafalaya River near Baton Rouge, LA (621 mm FL). Mean abundance (catch per trotline night) of pallid sturgeon was highest at water temperatures around 10°C. There was a latitudinal trend in mean abundance of pallid and shovelnose sturgeon, but the pattern differed between species. Pallid sturgeon abundance was statistically (P < 0.05) higher (0.3 fish per trotline night) in the lower reach between the Atchafalaya River and New Orleans (rkm 154–507), and at the Chain of Rocks (COR), a low water dam near the mouth of the Missouri River. Pallid sturgeon abundance between these two locations was statistically the same (0.12–0.23). Shovelnose sturgeon abundance increased going upstream, but was disproportionally higher at the COR (22 fish per line compared with <6 fish per line in other reaches). Overall, the ratio between pallid and shovelnose sturgeon varied from a high of 1 : 6 at the lower reach, and gradually decreased upstream to a low of 1 : 77 at the COR. Based on differences in sturgeon abundance, size and habitat characteristics, the free‐flowing Mississippi River can be divided into two reaches in the MMR (i.e. COR is a separate location), and four reaches (i.e., including the Atchafalaya River) in the LMR where management goals may differ. 相似文献

Herbivory is a primary factor in determining the structure of coral reef communities. Spatial variation among reef habitats in the intensity of herbivory has been documented, but underlying variation in species composition and abundance within the herbivore guild has received little attention. The distribution and relative abundances of herbivorous fishes and sea urchins across several habitats were studied on the Belizean barrier reef off the Caribbean coast of Central America. Marked variation in total herbivore density as well as major changes in the composition of the herbivore guild were found across reef habitats. Acanthurids (surgeonfishes) predominated in shallow areas (< 5 m) while scarids (parrotfishes) were dominant in deeper habitats. Significant differences among habitats in an experimental assay of grazing intensity were strongly correlated with herbivore abundance. The spatial distribution of herbivorous fishes across reef habitats does not appear to be simply explained by differences in reef topography, but may depend on complex interactions among proximity to nearby shelter, predator abundance, density of territorial competitors, and local availability of food resources. 相似文献

Abstract 1. The response of dispersal towards evolution largely depends on its heritability for which upper limits are determined by the trait’s repeatability. 2. In the Linyphiid spider E. atra, we were able to separate long‐ and short‐distance dispersal behaviours (respectively ballooning and rappelling) under laboratory conditions. By performing repeated behavioural trials for females, we show that average dispersal trait values decrease with increasing testing days. By comparing mated and unmated individuals during two periods (before and after mating for the mated group, and the same two periods for the unmated group), we show that mating has no effect on the mean displayed dispersal behaviour or its within‐individual variation. Repeatabilities were high and consistent for ballooning motivation, but not for rappelling. 3. Ballooning motivation can be regarded as highly individual‐specific behaviour, while general pre‐dispersal and rappelling behaviours showed more individual variation. Such difference in repeatability between long‐ and short‐distance dispersal suggests that short‐ and long‐distance dispersal events are triggered by different ecological and evolutionary mechanisms. 相似文献
