COLLIER, GREG R, KEN WALDER, PAUL LEWAN DOWSJCI, ANDREW SANIGORSKI, PAUL ZIMMET. Leptin and the development of obesity and diabetes in Psammomys obesus. The recently discovered ob gene and its circulating product, leptin, may be critical factors in the control of energy balance. Recent studies in ob/ob mice, which lack circulating leptin, have shown dramatic reductions in food intake and bodyweight after leptin treatment. In addition, studies in both humans with obesity and animal models of obesity have demonstrated hyperleptinemia. Here, we report a longitudinal study examining changes in circulating leptin during the development of obesity and diabetes in Psammomys obesus. Over the 8 weeks of the study, lean animals increased their bodyweight by 154% and leptin levels remained essentially unchanged. In contrast, animals that developed obesity (223 % increase in bodyweight), hyperglycemia, and hyperinsulinemia also developed hyperleptinemia between 4 weeks and 8 weeks of age. These results demonstrate that the development of hyperleptinemia is associated with the development of obesity and subsequent metabolic abnormalities.  相似文献

A model biofilm consisting of Pseudomonas aeruginosa, Pseudomonas fluorescens, and Klebsiella pneumoniae was developed to study the relationships between structural heterogeneity and hydrodynamics. Local fluid velocity in the biofilm system was measured by a noninvasive method of particle image velocimetry, using confocal scanning laser microscopy. Velocity profiles were measured in conduit and porous medium reactors in the presence and absence of biofilm. Liquid flow was observed within biofilm channels; simultaneous imaging of the biofilm allowed the liquid velocity to be related to the physical structure of the biofilm.  相似文献

A temporal study of gene expression in peripheral blood leukocytes (PBLs) from a Mycobacterium tuberculosis primary, pulmonary challenge model Macaca fascicularis has been conducted. PBL samples were taken prior to challenge and at one, two, four and six weeks post-challenge and labelled, purified RNAs hybridised to Operon Human Genome AROS V4.0 slides. Data analyses revealed a large number of differentially regulated gene entities, which exhibited temporal profiles of expression across the time course study. Further data refinements identified groups of key markers showing group-specific expression patterns, with a substantial reprogramming event evident at the four to six week interval. Selected statistically-significant gene entities from this study and other immune and apoptotic markers were validated using qPCR, which confirmed many of the results obtained using microarray hybridisation. These showed evidence of a step-change in gene expression from an ‘early’ FOS-associated response, to a ‘late’ predominantly type I interferon-driven response, with coincident reduction of expression of other markers. Loss of T-cell-associate marker expression was observed in responsive animals, with concordant elevation of markers which may be associated with a myeloid suppressor cell phenotype e.g. CD163. The animals in the study were of different lineages and these Chinese and Mauritian cynomolgous macaque lines showed clear evidence of differing susceptibilities to Tuberculosis challenge. We determined a number of key differences in response profiles between the groups, particularly in expression of T-cell and apoptotic makers, amongst others. These have provided interesting insights into innate susceptibility related to different host `phenotypes. Using a combination of parametric and non-parametric artificial neural network analyses we have identified key genes and regulatory pathways which may be important in early and adaptive responses to TB. Using comparisons between data outputs of each analytical pipeline and comparisons with previously published Human TB datasets, we have delineated a subset of gene entities which may be of use for biomarker diagnostic test development.  相似文献

Miscanthus species, which are C4perennial grasses, have a highbiomass potential but yields at many sites in Europe can belimited by insufficient water supply and plant survival is endangeredunder extreme summer drought. A pot experiment was conductedto measure the influence of reduced water supply on the wateruse efficiency (WUE) and biomass partitioning of three Miscanthusgenotypes (M. x giganteus, M. sacchariflorus, and a M. sinensishybrid) in a controlled environment. The experiment consistedof three phases (phase 1 = 0–20 d; phase 2 = 21–39d; phase 3 = 40–54 d) punctuated by destructive harvests.In phase 1, soil moisture was non-limiting. In the second andthird phases, lowered soil moisture contents induced water deficits.Air vapour pressure deficit (VPD) was 0.49 ± 0.05 kPa.Water deficits caused leaf senescence in M. x giganteus andM. sacchariflorus, but not in the M. sinensis hybrid. Greenleaf conductances were lowest in M. sinensis under water deficit,indicating stomatal regulation. Water use efficiency for wholeplants of each genotype ranged from 11.5 to 14.2 g dry matter(DM) kg-1H2O but did not differ significantly between genotypesor water treatments under the conditions of this experiment.However, differences in dry matter partitioning to the shoot(the harvestable component) resulted in genotypic differencesin WUE, calculated on a harvestable dry matter basis, whichranged from 4.1 g DM kg-1H2O for M. sacchariflorus to 2.2 gDM kg-1H2O for M. x giganteus. The physical properties (rheology) of biofilms will determine the shape and mechanical stability of the biofilm structure and consequently affect both mass transfer and detachment processes. Biofilm viscoelasticity is also thought to increase fluid energy losses in pipelines. Yet there is very little information on the rheology of intact biofilms. This is due in part to the difficulty in using conventional testing techniques. The size and nature of biofilms makes them difficult to handle, while removal from a surface destroys the integrity of the sample. We have developed a method which allowed us to conduct simple stress-strain and creep experiments on mixed and pure culture biofilms in situ by observing the structural deformations caused by changes in hydrodynamic shear stress (tau(w)). The biofilms were grown under turbulent pipe flow (flow velocity (u) = 1 m/s, Reynolds number (Re) = 3600, tau(w) = 5. 09 N/m(2)) for between 12 and 23 days. The resulting biofilms were heterogeneous and consisted of filamentous streamers that were readily deformed by changes in tau(w). At tau(w) of 10.11 N/m(2) the streamers were flattened so that the thickness was reduced by 25%. We estimated that the shear modulus (G) of the mixed culture biofilm was 27 N/m(2) and the apparent elastic modulus (E(app)) of both biofilms was in the range of 17 to 40 N/m(2). The biofilms behaved like elastic and viscoelastic solids below the tau(w) at which they were grown but behaved like viscoelastic fluids at elevated tau(w). The implications of these results for fluid energy losses and the processes of mass transfer and detachment are discussed.  相似文献

In the search for an effective DNA-typing technique for use in hospital epidemiology, the performance and convenience of a novel assay based on the fingerprinting of bacterial genomes by amplification of DNA fragments surrounding rare restriction sites (ADSRRS fingerprinting) was tested. A large number of vancomycin-resistant Enterococcus faecium (VREM) isolates from haematological ward patients of the Clinical Hospital in Gdańsk were examined. We found that ADSRRS fingerprinting analysis is a rapid method that offers good discriminatory power. The method demonstrated also excellent reproducibility. The usefulness of the ADSRRS fingerprinting method for molecular typing was compared with pulsed field gel electrophoresis (PFGE) method, which is currently considered the gold standard for molecular typing of isolates recovered from patients and the environment in the course of investigation and control of nosocomial outbreaks. Clustering of ADSRRS fingerprinting data matched pulsed field gel electrophoresis data.The features of ADSRRS fingerprinting technique is discussed in comparison with conventional methods. Data presented here demonstrate the complexity of the epidemiological situation concerning VREM that may occur in a single medical ward.  相似文献

BackgroundFeline injection-site sarcomas are malignant skin tumors of mesenchymal origin, the treatment of which is a challenge for veterinary practitioners. Methods of treatment include radical surgery, radiotherapy and chemotherapy. The most commonly used cytostatic drugs are cyclophosphamide, doxorubicin and vincristine. However, the use of cytostatics as adjunctive treatment is limited due to their adverse side-effects, low biodistribution after intravenous administration and multidrug resistance. Colloid gold nanoparticles are promising drug delivery systems to overcome multidrug resistance, which is a main cause of ineffective chemotherapy treatment. The use of colloid gold nanoparticles as building blocks for drug delivery systems is preferred due to ease of surface functionalization with various molecules, chemical stability and their low toxicity.MethodsStability and structure of the glutathione-stabilized gold nanoparticles non-covalently modified with doxorubicin (Au-GSH-Dox) was confirmed using XPS, TEM, FT-IR, SAXRD and SAXS analyses. MTT assay, Annexin V and Propidium Iodide Apoptosis assay and Rhodamine 123 and Verapamil assay were performed on 4 feline fibrosarcoma cell lines (FFS1WAW, FFS1, FFS3, FFS5). Statistical analyses were performed using Graph Pad Prism 5.0 (USA).ResultsA novel approach, glutathione-stabilized gold nanoparticles (4.3 +/- 1.1 nm in diameter) non-covalently modified with doxorubicin (Au-GSH-Dox) was designed and synthesized. A higher cytotoxic effect (p<0.01) of Au-GSH-Dox than that of free doxorubicin has been observed in 3 (FFS1, FFS3, FFS1WAW) out of 4 feline fibrosarcoma cell lines. The effect has been correlated to the activity of glycoprotein P (main efflux pump responsible for multidrug resistance).ConclusionsThe results indicate that Au-GSH-Dox may be a potent new therapeutic agent to increase the efficacy of the drug by overcoming the resistance to doxorubicin in feline fibrosarcoma cell lines. Moreover, as doxorubicin is non-covalently attached to glutathione coated nanoparticles the synthesized system is potentially suitable to a wealth of different drug molecules.  相似文献
