In greenhouse and field trials, a bacterial mixture of Collimonas arenae Cal35 and Bacillus velezensis FZB42, but not Cal35 alone or FZB42 alone, was able to protect tomato plants from challenge with the soilborne fungal pathogen Fusarium oxysporum f.sp. lycopersici (Fol). To identify genes and mechanisms underlying this property in Cal35, we screened a random transposon insertion library for loss of function and identified two mutants that were impaired completely or partially in their ability to halt the growth of a wide range of fungal species. In mutant 46A06, the transposon insertion was located in a biosynthetic gene cluster that was predicted to code for a hybrid polyketide synthase–non-ribosomal peptide synthetase, while mutant 60C09 was impacted in a gene cluster for the synthesis and secretion of sugar repeat units. Our data are consistent with a model in which both gene clusters are necessary for the production of an antifungal compound we refer to as carenaemins. We also show that the ability to produce carenaemin contributed significantly to the observed synergy between Cal35 and FZB42 in protecting tomato plants from Fol. We discuss the potential for supplementing Bacillus-based biocontrol products with Collimonas bacteria to boost efficacy of such products.  相似文献

Background

Camouflage patterns that hinder detection and/or recognition by antagonists are widely studied in both human and animal contexts. Patterns of contrasting stripes that purportedly degrade an observer's ability to judge the speed and direction of moving prey ('motion dazzle') are, however, rarely investigated. This is despite motion dazzle having been fundamental to the appearance of warships in both world wars and often postulated as the selective agent leading to repeated patterns on many animals (such as zebra and many fish, snake, and invertebrate species). Such patterns often appear conspicuous, suggesting that protection while moving by motion dazzle might impair camouflage when stationary. However, the relationship between motion dazzle and camouflage is unclear because disruptive camouflage relies on high-contrast markings. In this study, we used a computer game with human subjects detecting and capturing either moving or stationary targets with different patterns, in order to provide the first empirical exploration of the interaction of these two protective coloration mechanisms.

Results

Moving targets with stripes were caught significantly less often and missed more often than targets with camouflage patterns. However, when stationary, targets with camouflage markings were captured less often and caused more false detections than those with striped patterns, which were readily detected.

Conclusions

Our study provides the clearest evidence to date that some patterns inhibit the capture of moving targets, but that camouflage and motion dazzle are not complementary strategies. Therefore, the specific coloration that evolves in animals will depend on how the life history and ontogeny of each species influence the trade-off between the costs and benefits of motion dazzle and camouflage.  相似文献

Introduction

The goal of this study is to investigate whether the -308G > A promoter polymorphism in the tumor necrosis factor alpha (TNFA) gene is associated with disease severity and radiologic joint damage in a large cohort of patients with rheumatoid arthritis (RA).

Methods

A long-term observational early RA inception cohort (n = 208) with detailed information about disease activity and radiologic damage after 3, 6 and 9 years of disease was genotyped for the TNFA -308G > A promoter polymorphism (rs1800629). A longitudinal regression analysis was performed to assess the effect of genotype on RA disease severity and joint damage. Subsequently, a meta-analysis, including all publically available data, was performed to further test the association between joint erosions and the TNFA polymorphism. To learn more about the mechanism behind the effect of the polymorphism, RNA isolated from peripheral blood from RA patients (n = 66) was used for TNFA gene expression analysis by quantitative PCR.

Results

Longitudinal regression analysis with correction for gender and disease activity showed a significant difference in total joint damage between GG and GA+AA genotype groups (P = 0.002), which was stable over time. The meta-analysis, which included 2,053 patients, confirmed an association of the genetic variant with the development of erosions (odds ratio 0.78, 95% CI 0.62, 0.98). No significant differences in TNFA gene expression were observed for the different genotypes, confirming earlier findings in healthy individuals.

Conclusions

Our data confirm that the TNFA -308G > A promoter polymorphism is associated with joint damage in patients with RA. This is not mediated by differences in TNFA gene expression between genotypes.  相似文献

Since 1998, a group of archaeologists have been studying the long-term history of human settlement and activity in the mid to high altitude (1800m and above) in theParc National des Ecrins (southern French Alps). This research has identified a number of different phases of settlement since the end of the last Ice Age (10,000 years ago). Whilst we accept that it is impossible to present the variation in actual number of people present in these marginal milieus, we can present an overview of waxing and waning of human activity in our study area. Our research demonstrates that poeple moved into these high altitude zones as soon as the glaciers retreated. The first phase of extensive and relatively intensive activity dates to the Bronze Age (c. 2000 BC). During the Iron Age and Roman period there appears to have been a relative reduction in the level of activity. However, from the early medieval period onwards there is incontrovertible evidence for a substantial increase in activity. This culminates with the emergence of a “busy” landscape during the post-medieval period when mining and pastoral activities were at their peak.  相似文献

The process of restenosis after a stenting procedure is related to local biomechanical environment. Arterial wall stresses caused by the interaction of the stent with the vascular wall and possibly stress induced stent strut fracture are two important parameters. The knowledge of these parameters after stent deployment in a patient derived 3D reconstruction of a diseased coronary artery might give insights in the understanding of the process of restenosis.  相似文献

Numerous microorganisms, including bacteria, yeasts, and molds, constitute the complex ecosystem present in milk and fermented dairy products. Our aim was to describe the bacterial ecosystem of various cheeses that differ by production technology and therefore by their bacterial content. For this purpose, we developed a rapid, semisystematic approach based on genetic profiling by temporal temperature gradient electrophoresis (TTGE) for bacteria with low-G+C-content genomes and denaturing gradient gel electrophoresis (DGGE) for those with medium- and high-G+C-content genomes. Bacteria in the unknown ecosystems were assigned an identity by comparison with a comprehensive bacterial reference database of ~150 species that included useful dairy microorganisms (lactic acid bacteria), spoilage bacteria (e.g., Pseudomonas and Enterobacteriaceae), and pathogenic bacteria (e.g., Listeria monocytogenes and Staphylococcus aureus). Our analyses provide a high resolution of bacteria comprising the ecosystems of different commercial cheeses and identify species that could not be discerned by conventional methods; at least two species, belonging to the Halomonas and Pseudoalteromonas genera, are identified for the first time in a dairy ecosystem. Our analyses also reveal a surprising difference in ecosystems of the cheese surface versus those of the interior; the aerobic surface bacteria are generally G+C rich and represent diverse species, while the cheese interior comprises fewer species that are generally low in G+C content. TTGE and DGGE have proven here to be powerful methods to rapidly identify a broad range of bacterial species within dairy products.  相似文献

Chloroplast thylakoids contain three classes of glycolipids, monogalactosyldiacylglycerol (MGDG), digalactosyldiacylglycerol (DGDG), and sulfoquinovosyldiacylglycerol (SQDG). We have investigated the stability of large unilamellar vesicles made from egg phosphatidylcholine (EPC) and different chloroplast glycolipids during freezing to -18 degreesC, as a function of the presence of three sugars: glucose, sucrose, or trehalose. Contrary to the situation in thylakoids, where cryoprotection increases from glucose < sucrose < trehalose, liposomes containing 50% DGDG showed the opposite behavior. In fact, carboxyfluorescein leakage increased over the control values (freezing in the absence of sugar) in the presence of trehalose. This effect was not seen in vesicles made from pure EPC, or a mixture of EPC and MGDG, or EPC and SQDG. Liposomes made from mixtures of all three glycolipids, however, showed even more leakage in the presence of trehalose than liposomes containing only DGDG and EPC. Transfer processes from a eutrophic ecosystem, the river Rhône, to an oligotrophic environment, the north-west Mediterranean Sea, were studied during the winter period, when inputs of dissolved and particulate matter are at their highest.During a cruise carried out in February 1987, salinity, temperature, turbidity, nutrients, pigments, suspended particulate matter and particulate carbon and nitrogen were measured in surface and subsurface waters near the Rhône outlet.For most of the parameters the results show that during this period a simple dilution process occurs in the river plume of the Rhône without adsorption, desorption, precipitation or biological uptake, contrary to recordings for some other rivers. All the nutrients discharged into the sea by the Rhône are available for utilization by marine phytoplankters.  相似文献

Hydrological and chemical structures off the Rhône River estuary resulting from the introduction of the river flow into the Mediterranean Sea are described. The effect of the fresh-water/sea-water interface on the distribution of inorganic and organic matter off the Rhône river is investigated. Strong vertical gradients of inorganic and dissolved organic matter such as lipids characterized the first few meters in this area (from 83.7 to 0.6 N-NO3 µgat l–1, from 6.39 to 0.92 N-NH4 µgat l–1 and from 299 to 73 µg l–1 of total dissolved lipids). At the interface, substantial increases of particulate organic (PON: from 45 µg l–1 at surface to 118 µg l–1 at the interface, POC: from 462 to 876 µg l–1, total particulate lipids: from 33 to 648 µg l–1) and suspended matter in general (from 18 to 22.2 mg l–1) were observed. High phytoplanktonic production may account for some of this enrichment, although passive accumulation might also be involved.  相似文献
