Regulation of the supramolecular structure and catalytic activity of the heterodimeric enzyme gamma-glutamyltransferase in the system of Aerosol OT reversed micelles in octane was studied. Variation of the hydration degree causes a reversible dissociation of the enzyme to the light and heavy subunits, both possessing the catalytic activity. The subunits were separated on the preparative scale in the reversed micelle system using ultracentrifugation. The active centres of gamma-glutamyltransferase were studied using the enzyme's irreversible inhibitor AT-125 (L-(alpha S, 5S)-alpha-amino-3-chloro-4,5-dihydro-5-isoxazoleacetic acid). It is shown that the separation of the gamma-glutamyltransferase subunits results in "opening" of a new active centre in the heavy subunit, whereas in the enzyme's dimeric form this centre is masked and not accessible to the inhibitor's molecule. The kinetic and inhibitor analysis data indicate that the active centres in the light and heavy subunits are similar. 相似文献

Therapeutic efficiency of artificial Alpine climate was studied in a group of 318 patients with chronic nonspecific pulmonary diseases (CNDP). The Alpine climate was modelled on device "Orotron". The base, progradient and prolonged regimes of therapy by artificial Alpine climate (orhotherapy) were used in work. It is shown that application of optimal conditions of orhotherapy promotes normalization of the external respiration function in patients with CNPD due to a decrease in expression of broncho-obstructive and bronchitic syndromes and in nonuniformity of regional ventilation of lungs. Therapy by artificial Alpine climate is recommended to be used in wide clinical practice for treatment and rehabilitation of patients with CNDP. 相似文献

Micellar solutions of surfactant in organic solvents with rubber additions are proposed for determination of active enzyme concentration. A kinetic theory of enzymatic reactions in reversed micellar systems is developed, suggesting the intermicellar transport of the substrate to be the limiting step in viscous medium. Under these conditions, it is shown that fraction of the product formed after quick transformation of the substrate located in the enzyme-containing micelles depends upon active enzyme concentration and aggregation number of surfactant molecules. The proposed approach is used for the active-site titration of trypsin and cellobiase and for the determination of the aggregation number of Aerosol OT (AOT) molecules in the ternary system AOT/water/cyclohexane. 相似文献

In cell suspension of Desulfovibrio desulfuricans B-1388, oxidation of CO as the only energy source is associated with reduction of SO42-. After a 2-h incubation of cells in 8% CO, 81% of the gas is converted. Oxidation of 1 mole CO results in formation of 0.23 mole H2S. Intracellular ATP content increases from 2.5 (control) to 8.3 nmoles/mg (during CO conversion). Dinitrophenol inhibits sulfate reduction and CO oxidation. Equilibrium maintenance during standing in humans was investigated with a 3-joint (ankle, knee and hip) sagittal model of
body movement. The experimental paradigm consisted of sudden perturbations of humans in quiet stance by backward displacements
of the support platform. Data analysis was performed using eigenvectors of motion equation. The results supported three conclusions.
First, independent feedback control of movements along eigenvectors (eigenmovements) can adequately describe human postural
responses to stance perturbations. This conclusion is consistent with previous observations (Alexandrov et al., 2001b) that
these same eigenmovements are also independently controlled in a feed-forward manner during voluntary upper-trunk bending.
Second, independent feedback control of each eigenmovement is sufficient to provide its stability. Third, the feedback loop

The catalytic activity and quaternary structure of soluble (s) and membrane (m) forms of angiotensin-converting enzyme (ACE) were studied in reversed micelles of ternary system Aerosol OT--water--octane. The profile of the dependence of the catalytic activity of the two enzyme forms on the degree of surfactant hydration (micellar size) had several optima corresponding to the function of various active oligomeric enzyme forms; the curves for the s- and m-forms of ACE were different. Data of sedimentation analysis prove that in reversed micelles, s-ACE can exist as monomers, dimers, or tetramers depending on the hydration degree, and the m-form is present as dimers and tetramers only. The values of the kinetic parameters for the hydrolysis of the substrate furylacryloyl-Phe-Gly-Gly by all the enzyme forms were determined, and the data indicate that the activity of the m-form is enhanced by oligomerization. Thermal unfolding parameters were determined for a two-domain tetrameric enzyme, phosphorylating D-glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and for its isolated NAD(+)-binding domain. At pH 8.0, the transition temperatures (t(max)) for the apoforms of the native Bacillus stearothermophilus GAPDH and the isolated domain were 78.3 degrees C and 61.9 degrees C, with calorimetric enthalpies (DeltaH(cal)) of 4415 and 437 kJ/mol (or 30.7 and 22.1 J/g), respectively. In the presence of nearly saturating NAD(+) concentrations, the t(max) and the DeltaH(cal) increased by 13.6 degrees C and by 2365 kJ/mol, respectively, for the native apoenzyme, and by 2.8 degrees C and 109 kJ/mol for the isolated domain. These results indicate that interdomain interactions are essential for NAD(+) to produce its stabilizing effect on the structure of the native enzyme. The thermal stability of the isolated NAD(+)-binding domain increased considerably upon transition from pH 6.0 to 8.0. By contrast, native GAPDH exhibited greater stability at pH 6.0; similar pH-dependencies of thermal stability were displayed by GAPDHs isolated from rabbit muscle and Escherichia coli. The binding of NAD(+) to rabbit muscle apoenzyme increased t(max) and DeltaH(cal) and diminished the widths of the DSC curves; the effect was found to grow progressively with increasing coenzyme concentrations. Alkylation of the essential Cys149 with iodoacetamide destabilized the apoenzyme and altered the effect of NAD(+). Replacement of Cys149 by Ser or by Ala in the B. stearothermophilus GAPDH produced some stabilization, the effect of added NAD(+) being basically similar to that observed with the wild-type enzyme. These data indicate that neither the ion pairing between Cys149 and His176 nor the charge transfer interaction between Cys149 and NAD(+) make any significant contribution to the stabilization of the enzyme's native tertiary structure and the accomplishment of NAD(+)-induced conformational changes. The H176N mutant exhibited dramatically lower heat stability, as reflected in the values of both DeltaH(cal) and t(max). Interestingly, NAD(+) binding resulted in much wider heat capacity curves, suggesting diminished cooperativity of the unfolding transition. 相似文献

Partially purified delta 5-3-ketosteroid isomerase (KSI) from Pseudomonas testosteroni was studied kinetically after solubilization in reverse micelles of aerosol OT (AOT) in isooctane and water, as regards its application to biotechnology. With delta 5,10-estren-17 beta-ol-3-one as a substrate, KSI displays an enzyme activity in the micellar system but a low stability. In the presence of urea, the enzyme is, however, stable. Kinetic parameters of the stabilized enzyme are highly sensitive to both the hydration degree of the surfactant and its concentration.
