While the microbiota resident in the human gut is now known to provide a range of functions relevant to host health, many of the microbial members of the community have not yet been cultured or are represented by a limited number of isolates. Clostridium difficile is the commonest cause of hospital-acquired infection in the United Kingdom. We characterized the abilities of 21 clinical isolates to form spores; to adhere to inorganic and organic surfaces, including stainless steel and human adenocarcinoma cells; and to germinate. The composition of culture media had a significant effect on spore formation, as significantly more spores were produced in brain heart infusion broth (Student''s t test; P = 0.018). The spore surface relative hydrophobicity (RH) varied markedly (14 to 77%) and was correlated with the ability to adhere to stainless steel. We observed no correlation between the ribotype and the ability to adhere to steel. When the binding of hydrophobic (DS1813; ribotype 027; RH, 77%) and hydrophilic (DS1748; ribotype 002; RH, 14%) spores to human gut epithelial cells at different stages of cell development was examined, DS1813 spores adhered more strongly, suggesting the presence of surface properties that aid attachment to human cells. Electron microscopy studies revealed the presence of an exosporium surrounding DS1813 spores that was absent from spores of DS1748. Finally, the ability of spores to germinate was found to be strain and medium dependent. While the significance of these findings to the disease process has yet to be determined, this study has highlighted the importance of analyzing multiple isolates when attempting to characterize the behavior of a bacterial species.  相似文献

The Australian ghost bat is a large, opportunistic carnivorous species that has undergone a marked range contraction toward more mesic, tropical sites over the past century. Comparison of mitochondrial DNA (mtDNA) control region sequences and six nuclear microsatellite loci in 217 ghost bats from nine populations across subtropical and tropical Australia revealed strong population subdivision (mtDNA φST = 0.80; microsatellites URST = 0.337). Low-latitude (tropical) populations had higher heterozygosity and less marked phylogeographic structure and lower subdivision among sites within regions (within Northern Territory [NT] and within North Queensland [NQ]) than did populations at higher latitudes (subtropical sites; central Queensland [CQ]), although sampling of geographically proximal breeding sites is unavoidably restricted for the latter. Gene flow among populations within each of the northern regions appears to be male biased in that the difference in population subdivision for mtDNA and microsatellites (NT φST = 0.39, URST = 0.02; NQ φST = 0.60, URST = ?0.03) is greater than expected from differences in the effective population size of haploid versus diploid loci. The high level of population subdivision across the range of the ghost bat contrasts with evidence for high gene flow in other chiropteran species and may be due to narrow physiological tolerances and consequent limited availability of roosts for ghost bats, particularly across the subtropical and relatively arid regions. This observation is consistent with the hypothesis that the contraction of the species' range is associated with late Holocene climate change. The extreme isolation among higher-latitude populations may predispose them to additional local extinctions if the processes responsible for the range contraction continue to operate.  相似文献

New Zealand ephemeral wetlands are ecologically important, containing up to 12% of threatened native plant species and frequently exhibiting conspicuous cyanobacterial growth. In such environments, cyanobacteria and associated heterotrophs can influence primary production and nutrient cycling. Wetland communities, including bacteria, can be altered by increased nitrate and phosphate due to agricultural practices. We have characterized cyanobacteria from the Wairepo Kettleholes Conservation Area and their associated bacteria. Use of 16S rRNA amplicon sequencing identified several operational taxonomic units (OTUs) representing filamentous heterocystous and non‐heterocystous cyanobacterial taxa. One Nostoc OTU that formed macroscopic colonies dominated the cyanobacterial community. A diverse bacterial community was associated with the Nostoc colonies, including a core microbiome of 39 OTUs. Identity of the core microbiome associated with macroscopic Nostoc colonies was not changed by the addition of nutrients. One OTU was highly represented in all Nostoc colonies (27.6%–42.6% of reads) and phylogenetic analyses identified this OTU as belonging to the genus Sphingomonas. Scanning electron microscopy showed the absence of heterotrophic bacteria within the Nostoc colony but revealed a diverse community associated with the colonies on the external surface.  相似文献

Indigenous fire knowledge offers significant benefits for ecosystem management and human livelihoods, but is threatened worldwide because of disruption of customary practices. In Australia, the historical prevalence and characteristics of Aboriginal burning are intensely debated, including arguments that Aboriginal burning was frequent across the continent. Frequent burning is supported by contemporary Aboriginal knowledge and practice in some regions, but in southern Australia evidence is typically limited to historical and ecological records. Towards characterizing Aboriginal fire regimes in southern Australia, we collaborated with Ngadju people from the globally significant Great Western Woodlands in south‐western Australia to document their fire knowledge. We used workshops, site visits, interviews and occupation mapping to aid knowledge sharing. Consistent with the established significance of Aboriginal fire in Australia, planned fires were important in Ngadju daily life and land management. However, Ngadju use of fire was characterized by its selectivity rather than its ubiquity. Specifically, Ngadju described only highly targeted planned burning across extensive eucalypt woodlands and sandplain shrublands. By contrast, frequent planned burning was described for resource‐rich landscape elements of more restricted extent (granite outcrop vegetation, grasslands and coastal scrub). Overall, Ngadju fires are likely to have resulted in subtle but purposeful direct effects on the vegetation and biota. However the extent to which they collectively constrained large, intense wildfires remains unclear. Ngadju demonstrated a predictive knowledge of the ecological consequences of burning, including attention to fine‐scale needs of target organisms, and application of diverse fire regimes. These are consistent with the recently proposed concept that Aboriginal burning was guided by ‘templates’ targeting different resources, although diverse regimes predominantly reflect edaphically driven vegetation patterns rather than template‐driven use of fire to create resource diversity. We conclude that Ngadju fire knowledge fills an important gap in understanding Aboriginal fire regimes in southern Australia, highlighting a novel balance between frequent and constrained use of fire.  相似文献

We studied prefledging growth, energy expenditure and time budgets of African Black Oystercatcher, Haematopus moquini, chicks on Robben Island, Western Cape, South Africa. The aim of the study was to investigate the effect of parental feeding on the growth and energetics of semi-precocial shorebird chicks. Chicks reached mean fledging mass, 463 g, in 40 days. The growth rate coefficient of African Black Oystercatcher chicks was 2% below the predicted value for a shorebird species of their body mass, but it was smaller than that of other precocial and semi-precocial shorebirds to date. Resting metabolic rate (RMR, measured through respirometry), daily metabolisable energy (DME), defined as daily energy expenditure (DEE, measured with doubly labelled water) plus energy deposited into tissue (E(tis)), and total metabolisable energy (TME) of African Black Oystercatcher chicks were similar to those expected for a species of their body size. DEE was not influenced by weather (ambient temperature, operative temperature and wind speed), therefore, variations in DEE may be explained by body mass alone. The relative RMR of the African Black Oystercatcher was greater, their TME was approximately the same, their average daily metabolisable energy (ADME) was less, and they spent less time foraging (short periods of parental feeding) and more time inactive than three precocial species in the Western Cape. Therefore, the semi-precocial mode of development of African Black Oystercatcher chicks reduced energy costs from thermoregulation and activity, and they were able to grow relatively faster than precocial, self-feeding shorebird species in similar climatic conditions. The growth rate coefficient of African Black Oystercatcher chicks was smaller than that of Eurasian Oystercatcher, Haematopus ostralegus, chicks, which may be a consequence of differences in body size and latitudinal effects.  相似文献

An optimal Yurchenko layout vault of an elite female gymnast was identified by Koh et al. [2003. A predicted optimal performance of the Yurchenko layout vault in women's artistic gymnastics. Journal of Applied Biomechanics 19, 187-204] to require a combination of an increased body angle at horse contact and increased angular momentum for postflight than was recorded experimentally. However, the individual effect of each of these variables to producing the optimal vault is not known. The purpose of the study was to determine an appropriate strategy to teaching the optimal Yurchenko layout vault. Separate optimisations were carried out to investigate how each of these variables would change in order to produce the optimal vault identified by Koh et al. (2003). A combined optimal parameter selection and optimal control approach was used. The results suggest that when the body angle of attack at horse impact was kept low, pre-flight angular momentum had to be increased, with further gains during horse impact, to produce an optimal vault. This strategy of increasing solely the level of angular momentum needed for optimum postflight may not be attainable realistically. On the other hand, employing a larger body angle of attack required an increase in angular momentum during impact but which was attainable. Both optimisations show that increasing the vertical CM horse takeoff velocity is essential for postflight height and distance. The strategy to enhance performance should thus focus on maintaining an appropriate CM pre-flight velocity, a high level of angular momentum during pre-flight and to contact the horse with a large body angle of attack.  相似文献
