A series of nucleoside transport inhibitors has been tested for their ability to displace [3H]diazepam binding to CNS membranes. No correlation between their potency as [3H]adenosine uptake blockers and as inhibitors of [3H]diazepam binding was found, either in rat or guinea-pig brain tissue. Dipyridamole, a potent adenosine transport inhibitor interacted strongly (Ki = 54 nM) with peripheral-type benzodiazepine binding sites (“acceptor sites”) and was 4–5 fold weaker in displacing [3H]methylclonazepam and [3H]Ro15-1788, ligands selective for the specific central benzodiazepine “receptor”. Unlike the benzodiazepines, dipyridamole had no anticonvulsant action against metrazole-induced convulsions in mice. Ro5-4864, a benzodiazepine which selectively interacts with the peripheral-type benzodiazepine binding site, was approximately equipotent with diazepam in inhibiting [3H]adenosine uptake in brain tissue. These results do not support the idea of a very close link between high-affinity central binding sites for clinically-active benzodiazepines and the adenosine uptake site. The possibility of a connection between benzodiazepine “acceptor” sites and the membrane nucleoside transporter is discussed.  相似文献

Aim Cuckoo‐shrikes and allies (Campephagidae) form a radiation of birds widely distributed in the Indo‐Pacific and Africa. Recent studies on the group have been hampered by poor taxon sampling, causing inferences about systematics and biogeography to be rather speculative. With improved taxon sampling and analyses within an explicit spatiotemporal framework, we elucidate biogeographical patterns of dispersal and diversification within this diverse clade of passerine birds. Location Africa, Asia, Australo‐Papua, the Pacific, the Philippines and Wallacea. Methods We use model‐based phylogenetic methods (Mr Bayes and garli ) to construct a phylogenetic hypothesis of the core Campephagidae (Campephagidae with the exclusion of Pericrocotus). The phylogeny is used to assess the biogeographical history of the group with a newly developed Bayesian approach to dispersal–vicariance analysis (Bayes‐diva) . We also made use of a partitioned beast analysis, with several calibration points taken from island ages, passerine mitochondrial substitution rates and secondary calibration points for passerine birds, to assess the timing of diversification and dispersal. Results We present a robust molecular phylogeny that includes all genera and 84% of the species within the core Campephagidae. Furthermore, we estimate divergence dates and ancestral area relationships. We demonstrate that Campephagidae originated in Australo‐Papua with a single lineage (Pericrocotus) dispersing to Asia early. Later, there was further extensive transoceanic dispersal from Australo‐Papua to Africa involving lineages within the core Campephagidae radiation. Main conclusions The phylogenetic relationships, along with the results of the ancestral area analysis and the timing of dispersal events, support a transoceanic dispersal scenario from Australo‐Papua to Africa by the core Campephagidae. The sister group to core Campephagidae, Pericrocotus, dispersed to mainland Asia in the late Oligocene. Asia remained uncolonized by the core Campephagidae until the Pliocene. Transoceanic dispersal is by no means an unknown phenomenon, but our results represent a convincing case of colonization over a significant water gap of thousands of kilometres from Australo‐Papua to Africa.  相似文献

Like most of Madagascar's endemic primates, ring‐tailed lemurs (Lemur catta) face a number of threats to their survival. Although habitat loss is of greatest concern, other anthropogenic factors including environmental contamination may also affect lemur health and survival. In this study, we examined ring‐tailed lemurs from the Beza Mahafaly Special Reserve (BMSR), southern Madagascar for exposure to organochlorine (OC) pesticides and metals and examined differences in contaminant concentrations between sexes and among age groups, troops, and habitats. A total of 14 pesticides and 13 metals was detected in lemur blood (24 individuals) and hair (65 individuals) samples, respectively. p,p′‐DDT, heptachlor, aldrin, heptachlor epoxide, endrin aldehyde, and endrin were among the most prevalent pesticides detected. Surprisingly, the persistent metabolite of p,p′‐DDT, p,p′‐DDE, was not detected. The most commonly detected metals were aluminum, zinc, boron, phosphorus, silicon, and copper, whereas metals considered more hazardous to wildlife (e.g. arsenic, cadmium, lead, selenium, vanadium) were not found above detection limits. Overall, concentrations of OC pesticides and metals were low and similar to those considered to be background concentrations in other studies examining the ecotoxicology of wild mammals. Few inter‐sex, ‐age, ‐troop, and ‐habitat differences in contaminant concentrations were observed, suggesting a uniform distribution of contaminants within the reserve. Several statistically significant relationships between lemur body size and contaminant concentrations were observed, but owing to the lack of supportive data regarding contaminant exposure in wild primates, the biological significance of these findings remains uncertain. Results of this study document exposure of ring‐tailed lemurs at BMSR to multiple OC pesticides and metals and provide essential baseline data for future health and toxicological evaluations of lemurs and other wild primates, especially those in regions with expanding agricultural and mining operations. Morphological and genetic variation were studied in Plantago cordata Lam., an imperiled aquatic plant. The existence of distinct seasonal morphs was confirmed by numerical morphological analyses of specimens from 53 populations. Plants from two North Carolina populations possessed leaves that were significantly shorter, narrower, and fewer-veined than populations in other portions of the species range. Genetic variation within and among ten populations in seven states/provinces (Illinois, Missouri, New York, North Carolina, Ohio, Ontario, and Wisconsin) was examined electrophoretically. The species is an apparent allopolyploid with fixed heterozygosity observed at 67% of the loci surveyed. Electrophoretic variation was mostly partitioned among populations. The genetic identity among populations was high except for North Carolina populations which differed at 27% of the loci surveyed.  相似文献

The elygium is a darkly pigmented projection over the pupil of the tadpoles of certain species that live mostly at high altitudes. It has been suggested that this structure shades the retina, protecting it from high UV levels. In post‐metamorphic individuals, the elygium is replaced by a structure known as an umbraculum. Confusion arose in the past from the inconsistent use of terminology when referring to these two structures. While they may serve the same function, these structures differ fundamentally in structure and origin. Our investigation of the ultrastructure of the eye in Amietia vertebralis tadpoles, using electron and light microscopy, revealed that the elygium consists of an accumulation of melanophores situated within the inner cornea, whereas the umbraculum is a dorsal extension of the iris pigment epithelium, analogous to the ventral pupillary nodule, which is present in most frog species. In A. vertebralis, the umbraculum starts to develop on the iris of older tadpoles (Gosner [ 1960 ] stage 37 Herpetologica 16:183–190), medial to the overlying elygium. A smaller, ventral elygium is also present in this species and is similar in structure to the dorsal elygium. The development of the elygium over a six‐month period is also described. J. The African Great Lakes are important sources of fishes and water for domestic use, are used as avenues of transport, and receive agricultural, domestic and industrial effluents and atmospheric residues. Some of these lakes have speciose fish faunas of great interest to science. The catchment areas of some of the lakes are highly populated and user conflicts have increased the demands on the lakes' resources. There have been drastic reductions in fish stocks in most of the lakes due to overfishing. Introductions of new fish species, though followed by increases in fish catches, have been accompanied by a decline and in some cases extinction of native fish species. Some of the lakes have been invaded by the water hyacinth, Eichhornia crassipes. Agricultural activities, deforestation and devegetation of the catchment areas have increased siltation, and led to loss of suitable habitats and biodiversity. There are increased nutrient inputs from agriculture, sewage and industrial discharges and combustion processes which can cause eutrophication. There are also increased threats of toxic pollution from industrial waste discharge, mining, pesticides, and oil residues and spills. Climatic changes may also affect thermal stability of the lakes. These factors threaten availability of dietary protein, clean water and biodiversity. National and international efforts are required to manage the fisheries, guide the introduction of exotics, conserve biodiversity, control the water hyacinth, control eutrophication, reduce input of contaminants and manage climate change.  相似文献

Polyclonal antibodies raised against NAD-dependent glyceraldehyde-3-phosphate dehydrogenase (D-glyceraldehyde-3-phosphate:NAD+ oxidoreductase [phosphorylating], EC 1.2.1.12) from the plant cytosolic fraction of soybean [ Glycine max (L.) Merr. cv. Williams] nodules were used to study the subcellular location of the enzyme and its relative distribution between infected, interstitial and cortical cells of soybean (cv. Lincoln) nodules. Post-embedding immunogold labelling was carried out on nodules harvested 5, 12, 19 and 25 days after the first sign of nodulation. Labelling for glyceraldehyde-3-phosphate dehydrogenase was observed over the cytoplasm and nuclei of infected and uninfected cells, as well as over the nucleoid regions of bacteroids. In 5-day-old nodules, label also bound adjacent to the peribacteroid membranes. Statistical analysis of the number of gold particles per cell area indicated that in 5-day-old nodules, glyceraldehyde-3-phosphate dehydrogenase was distributed equally between infected, interstitial and cortical cells, but in older nodules the enzyme was more prominent in the interstitial and cortical cells than in infected cells.  相似文献

While poleward species migration in response to recent climatic warming is widely documented, few studies have examined entire range responses of broadly distributed sessile organisms, including changes on both the trailing (equatorward) and the leading (poleward) range edges. From a detailed population census throughout the entire geographical range of Aloe dichotoma Masson, a long-lived Namib Desert tree, together with data from repeat photographs, we present strong evidence that a developing range shift in this species is a 'fingerprint' of anthropogenic climate change. This is explained at a high level of statistical significance by population level impacts of observed regional warming and resulting water balance constraints. Generalized linear models suggest that greater mortalities and population declines in equatorward populations are virtually certainly the result, due to anthropogenic climate change, of the progressive exceedance of critical climate thresholds that are relatively closer to the species' tolerance limits in equatorward sites. Equatorward population declines are also broadly consistent with bioclimatically modelled projections under anticipated anthropogenic climate change but, as yet, there is no evidence of poleward range expansion into the area predicted to become suitable in future, despite good evidence for positive population growth trends in poleward populations. This study is among the first to show a marked lag between trailing edge population extinction and leading edge range expansion in a species experiencing anthropogenic climate change impacts, a pattern likely to apply to most sessile and poorly dispersed organisms. This provides support for conservative assumptions of species' migration rates when modelling climate change impacts for such species. Aloe dichotoma 's response to climate change suggests that desert ecosystems may be more sensitive to climate change than previously suspected.  相似文献
