Rap1 Functions as a Key Regulator of T-Cell and Antigen-Presenting Cell Interactions and Modulates T-Cell Responses 下载免费PDF全文

Activation of T cells by antigen requires adhesive interactions with antigen-presenting cells (APC) in which leukocyte function-associated antigen 1 (LFA-1) and intercellular adhesion molecules (ICAMs) are important. However, it is not well understood what signaling molecules regulate this process and how the modulation of adhesive events influences T-cell activation. Here we show that Rap1 is activated in T cells in an antigen-dependent manner and accumulated at the contact site of T-cell and antigen-loaded APC. Inhibition of Rap1 activation by a dominant-negative Rap1 or SPA-1, a Rap1 GTPase-activating protein, abrogates LFA-1-ICAM-1-mediated adhesive interactions with antigen-pulsed APC and the subsequent T-cell-receptor triggering and interleukin-2 production. Conversely, augmented antigen-dependent Rap1 activation by the expression of wild-type Rap1 enhances these responses but culminates in apoptosis by Fas and FasL. Thus, Rap1 functions as a key regulator of T-cell and APC interactions and modulates T-cell responses from productive activation to activation-induced cell death by regulating the strength of adhesive interactions. Moreover, constitutive Rap1 activation rendered T cells unresponsive with accumulation of p27(Kip1). Transmission and reception of mobile telephony signals take place through electromagnetic wave radiation, or electromagnetic radiofrequency fields, between the mobile terminal and the radio base station. Based on reports in the literature on adverse effects from exposure to this type of radiation, the objective of this study was to evaluate the genotoxic and cytotoxic potential of such exposure, by means of the micronucleus test on exfoliated cells from the oral epithelium. The sample included 45 individuals distributed in 3 groups according to the amount of time in hours per week (t) spent using mobile phones: group I, t?>?5?h; group II, t?>?1?h and ≤?5?h; and group III, t?≤?1?h. Cells from the oral mucosa were analyzed to assess the numbers of micronuclei, broken egg structures and degenerative nuclear abnormalities indicative of apoptosis (condensed chromatin, karyorrhexis and pyknosis) or necrosis (karyolysis in addition to these changes). The occurrences of micronuclei and degenerative nuclear abnormalities did not differ between the groups, but the number of broken egg (structures that may be associated with gene amplification) was significantly greater in the individuals in group I (p?0.05). 相似文献

Pseudomonas avellanae (Pav) has been reported as the causal agent of bacterial decline and bacterial canker of hazelnut in Italy and Greece, respectively. Both hazelnut diseases were reported to be similar in terms of symptoms, severity and persistence. In this study, we found that both symptomatic and asymptomatic trees in the field were colonized by Pav. Multilocus Sequence Typing (MLST) analysis showed that Pav strains isolated during this study in Italy belong to the P. syringae phylogroup 1 and they are closely related to Pav strains previously isolated in Greece from hazelnut bacterial canker. On the other hand, strains isolated in earlier studies from hazelnut decline in Italy belong to both phylogroup 1 and 2 of P. syringae. Both phylogroup 1 strains of P. syringae from Greece and Italy are different than strains isolated in this study in terms of their capacity to excrete fluorescent pigments on different media. Despite the same plant genotype and cropping practices adopted, the incidence of hazelnut decline ranged from nearly 0 to 91% across our study sites. No disease developed on plants inoculated with Pav through wounding while leaf scar inoculations produced only mild disease symptoms. Based on our results and the previously reported correlation between pedo-climatic conditions and hazelnut decline, we conclude that hazelnut decline in central Italy could be incited by a combination of predisposing (adverse pedo-climatic conditions) and contributing factors (Pav). Because this is a true decline different from “bacterial canker” described in Greece, we refer to it as hazelnut decline (HD). 相似文献

Phenotypic adaptations may be the result of changes in gene structure or gene regulation, but little is known about the evolution of gene expression. In addition, it is unclear whether the same selective forces may operate at both levels simultaneously. Reproductive proteins evolve rapidly, but the underlying selective forces promoting such rapid changes are still a matter of debate. In particular, the role of sexual selection in driving positive selection among reproductive proteins remains controversial, whereas its potential influence on changes in promoter regions has not been explored. Protamines are responsible for maintaining DNA in a compacted form in chromosomes in sperm and the available evidence suggests that they evolve rapidly. Because protamines condense DNA within the sperm nucleus, they influence sperm head shape. Here, we examine the influence of sperm competition upon protamine 1 and protamine 2 genes and their promoters, by comparing closely related species of Mus that differ in relative testes size, a reliable indicator of levels of sperm competition. We find evidence of positive selection in the protamine 2 gene in the species with the highest inferred levels of sperm competition. In addition, sperm competition levels across all species are strongly associated with high divergence in protamine 2 promoters that, in turn, are associated with sperm swimming speed. We suggest that changes in protamine 2 promoters are likely to enhance sperm swimming speed by making sperm heads more hydrodynamic. Such phenotypic changes are adaptive because sperm swimming speed may be a major determinant of fertilization success under sperm competition. Thus, when species have diverged recently, few changes in gene-coding sequences are found, while high divergence in promoters seems to be associated with the intensity of sexual selection. 相似文献

Non-human primates use various communicative means in interactions with others. While primate gestures are commonly considered to be intentionally and flexibly used signals, facial expressions are often referred to as inflexible, automatic expressions of affective internal states. To explore whether and how non-human primates use facial expressions in specific communicative interactions, we studied five species of small apes (gibbons) by employing a newly established Facial Action Coding System for hylobatid species (GibbonFACS). We found that, despite individuals often being in close proximity to each other, in social (as opposed to non-social contexts) the duration of facial expressions was significantly longer when gibbons were facing another individual compared to non-facing situations. Social contexts included grooming, agonistic interactions and play, whereas non-social contexts included resting and self-grooming. Additionally, gibbons used facial expressions while facing another individual more often in social contexts than non-social contexts where facial expressions were produced regardless of the attentional state of the partner. Also, facial expressions were more likely ‘responded to’ by the partner’s facial expressions when facing another individual than non-facing. Taken together, our results indicate that gibbons use their facial expressions differentially depending on the social context and are able to use them in a directed way in communicative interactions with other conspecifics. 相似文献

- Waterbird‐mediated endozoochory is an essential mechanism for the dispersal of sessile organisms in freshwater ecosystems. However, in the neotropics there are no previous studies of how different waterbird species vary in the dispersal functions they perform, and how seasonality influences endozoochory. In this study, we identified plant diaspores dispersed in faeces of five South American waterfowl (Brazilian teal Amazonetta brasiliensis, yellow‐billed teal Anas flavirostris, ringed teal Callonetta leucophrys, coscoroba swan Coscoroba coscoroba, and white‐faced whistling‐duck Dendrocygna viduata).
- We collected 165 faecal samples from five wetlands in southern Brazil surrounded by pasture and rice fields, then separated and measured intact seeds and other diaspores. Using generalised linear models, we tested how diaspore abundance and taxonomic richness differed among bird species and between cold (April–September) and warm (October–March) periods. We also analysed bird‐specific and seasonal variations in diaspore composition through principal coordinates analysis and permutational multivariate analysis of variance. We used indicator species analysis to determine which diaspore species discriminated between bird species and seasons. Finally, we measured diaspore length in order to analyse differences among waterfowl species in the size of diaspores dispersed.
- We found 2,066 intact diaspores from 40 different plant taxa, including seeds of 37 angiosperms and diaspores of Lycophyta (Isoetes cf. maxima), Pteridophyta (Azolla filiculoides), and Charophyceae. There was at least one diaspore in 65% of all faecal samples. Diaspores of native amphibious and emergent plants were dominant. We found 1,835 diaspores (from 33 taxa) in the cold period but only 231 (23 taxa) in the warm period. Seeds of the grass Zizaniopsis bonariensis and of the sedge Rynchospora sp. were the most abundant taxa. A strong interaction between bird species and season was the most important predictor of variation in both taxonomic richness and abundance of diaspores. The taxonomic composition of diaspores differed among waterfowl species and season. Indicator species analysis identified 12 plant taxa associated with particular bird species and seasons. Coscoroba swan, the largest bodied species in our study dispersed a higher proportion (8.2%) of large (length >2 mm) seeds.
- Despite considerable overlap, there are important differences in the plants dispersed by each species, and the smallest (ringed teal) and largest (coscoroba swan) birds are particularly different. All five waterfowl species are distributed over wide areas of South America and here we demonstrated that they are likely to be important plant vectors connecting wetland species at different geographical scales. Many of these plants have previously been assumed to lack mechanisms for long‐distance dispersal.
