Cell-stroma interactions in monocytopoiesis   总被引:2,自引:0,他引:2  
Abstract We have used immunohistochemistry to distinguish monocytes from early granulocyte precursors in trephine biopsies, in order to determine the distribution of monocytopoiesis within bone marrow. Developing granulocytes and monocytes have extensively overlapping immunophenotypes, but differential expression of calgranulin by monocytes and granulocytes during their maturation permitted the use of this antigen as a marker of bone marrow monocytes. In addition to morphologically normal bone marrow biopsies, in which monocyte numbers are relatively low, we studied pathological conditions in which either monocytopoiesis or granulopoiesis is selectively increased. By contrast with the highly zonal distribution of developing granulocytes, we found that monocytes were dispersed singly throughout the bone marrow. There was no evidence of preferential localisation of monocytes to particular stromal compartments. We hypothesise that developing monocytes are highly mobile within the bone marrow stroma and are relatively independent of physical stromal contacts for differentiation signals.  相似文献

This report investigates physical changes associated with the short-term inhibition of root elongation in intact maize seedlings (Zea mays L. vs. Halamish) by exogenous auxin. Movement of root tips was assayed by video microscopy in control roots, roots grown for 45 min in 10–6 M indole3-acetic acid (IAA), or roots chilled for 3 min at 11°C. IAA and chilling treatments similarly reduced root elongation rates (from 29 ± 6 m min–1 to 6 ± 2 m min–1). Initial rates of root tip contraction induced by 300 mOsmol mannitol were used to calculate tissue contractibility values. These allowed a comparison of effects of IAA and chilling treatments on apparent rates of water transport out of the root tip tissues. Chilling treatment reduced root tip contractibility by 66%, whereas IAA had much less effect (26% reduction). Roots were also exposed to an osmotic jump treatment; the initial osmotically induced increase in elongation rate was used to determine root tip extensibility values. Both IAA and chilling treatments reduced root tip extensibilities by 57%. Inhibition of wall-yielding properties, rather than hydraulic limitations, appeared to be primarily associated with inhibition of intact root tip elongation by exogenous IAA.  相似文献

Milk is a complex bio-colloid which presents some unique problems for the protein isolation chemist, but the majority of the processing criteria for purifying recombinant proteins are the same as with any complex biological mixture. The casein micelles and fat globules behave as separate phases; they prevent filtration of the milk and interfere with the usual separation methods. The usual first step is to centrifuge the milk to remove the fat and precipitate the casein micelles with low pH or precipitating agents. Some recombinant proteins may associate to some degree with the micelles which may necessitate solubilizing them with chelating agents. If the majority of the product protein associates with either the fat or micelles, this can be used to advantage. Once the casein micelles have been removed or disrupted, the clarified milk can be processed by the usual separation methods. There also are proteases in milk which can degrade recombinant proteins. The greatest advantage of producing recombinant proteins in milk is the high concentration which can be obtained. The high levels of product protein can alleviate many problems associated with the application of classical purification strategies to transgenic milk proteins.  相似文献

An ion-selective vibrating-microelectrode system, which was originally used to measure extracellular Ca2+ gradients generated by Ca2+ currents, was used to study K+, H+ and Ca2+ transport in intact maize (Zea mays L.) roots and individual maize suspension cells. Comparisons were made between the vibrating ion-selective microelectrode, and a technique using stationary ion-selective microelectrodes to measure ionic gradients in the unstirred layer at the surface of plant roots. The vibrating-microelectrode system was shown to be a major improvement over stationary ion-selective microelectrodes, in terms of sensitivity and temporal resolution. With the vibrating ion microelectrode, it was easy to monitor K+ influxes into maize roots in a background K+ concentration of 10 mM or more, while stationary K+ electrodes were limited to measurements in a background K+ concentration of 0.3 mM or less. Also, with this system it was possible to conduct a detailed study of root Ca2+ transport, which was previously not possible because of the small fluxes involved. For example, we were able to investigate the effect of the excision of maize roots on Ca2+ influx. When an intact maize root was excised from the seedling at a position 3 cm from the site of measurement of Ca2+ transport, a rapid fourfold stimulation of Ca2+ influx was observed followed by dramatic oscillations in Ca2+ flux, oscillating between Ca2+ influx and efflux. These results clearly demonstrate that wound or perturbation responses of plant organs involve transient alterations in Ca2+ transport, which had previously been inferred by demonstrations of touch-induced changes in cytoplasmic calcium. The sensitivity of this system allows for the measurement of ion fluxes in individual plant cells. Using vibrating K+ and H+electrodes, it was possible to measure H+efflux and both K+ influx and efflux in individual maize suspension cells under different conditions. The availability of this technique will greatly improve our ability to study ion transport at the cellular level, in intact plant tissues and organs, and in specialized cells, such as root hairs or guard cells.Symbol X amplitude of vibration The authors would like to thank Richard Sanger for his invaluable work on the design and improvement of the ion-selective vibratingmicroelectrode system. Lemon plants (Citrus limonum L. cv. Verna) were grown in the field under two different flood irrigation treatments. The dry treatment received four irrigations per year (March, July, September and November) and the wet treatment one monthly. The amounts of water applied per year for dry and wet treatments were 340.0 mm and 1020.0 mm, respectively. The effects of the two treatments on certain aspects of the plant water relations during the period between flowering and the end of rapid fruit growth (critical period) were studied. Soil matric potential (ψm) and leaf water potential (ψi) values in the dry treatment revealed development of water stress during the experimental period. The water supply in the wet treatment seems sufficient to achieve the crop water requirements. The g1 values in July were higher in the wet than dry treatments. Pronounced oscillations in g1 from sunrise to afternoon were found especially in the dry treatment.  相似文献

Immunofluorescence and microinjection experiments have shown that vinculin (molecular weight 130,000) is localized at adhesion plaques of fibroblasts spread on a solid substrate. We found that this protein affects actin filament assembly and interactions in vitro at substoichiometric levels. Vinculin inhibits the rate of actin polymerization under conditions that limit nuclei formation, indicating an effect on the filament elongation step of the reaction. Vinculin also reduces actin filament-filament interaction measured with a low-shear viscometer. Scatchard plot analysis of the binding of 3H-labeled vinculin to actin filaments showed that there is one high-affinity binding site (dissociation constant = 20 nM) for every 1,500–2,000 actin monomers. These results suggest that vinculin interacts with a specific site located at the growing ends of actin filaments in a cytochalasin-like manner, a property consistent with its proposed function as a linkage protein between filaments and the plasma membrane.  相似文献

A mathematical model for the perturbation of a biological oscillator by single and periodic impulses is analyzed. In response to a single stimulus the phase of the oscillator is changed. If the new phase following a stimulus is plotted against the old phase the resulting curve is called the phase transition curve or PTC (Pavlidis, 1973). There are two qualitatively different types of phase resetting. Using the terminology of Winfree (1977, 1980), large perturbations give a type 0 PTC (average slope of the PTC equals zero), whereas small perturbations give a type 1 PTC. The effects of periodic inputs can be analyzed by using the PTC to construct the Poincaré or phase advance map. Over a limited range of stimulation frequency and amplitude, the Poincaré map can be reduced to an interval map possessing a single maximum. Over this range there are period doubling bifurcations as well as chaotic dynamics. Numerical and analytical studies of the Poincaré map show that both phase locked and non-phase locked dynamics occur. We propose that cardiac dysrhythmias may arise from desynchronization of two or more spontaneously oscillating regions of the heart. This hypothesis serves to account for the various forms of atrioventricular (AV) block clinically observed. In particular 22 and 42 AV block can arise by period doubling bifurcations, and intermittent or variable AV block may be due to the complex irregular behavior associated with chaotic dynamics.  相似文献

The exact sites at which a number of drugs inhibit the nick translation of DNA by E.coli DNA polymerase-I have been pinpointed. In order to do this, a method has been developed for sequencing double-stranded plasmid DNA from the site of a specifically induced nick. The initial experiments have concentrated on analysis of drug inhibition of nick translation in a 200 nucleotide region near the Eco Rl origin of pBR313. Many drugs were found to inhibit nick translation in a highly sequence specific manner. For actinomycin D, significant inhibition occurred at just four sites in the nucleotide sequence under test and only one sequence (pGpCpGpCpGpGp) gave really strong inhibition. Distamycin A gave a different pattern of inhibition with particularly strong stops in just two of the many A-T rich regions in the DNA. Experiments with caffeine suggest that factors in addition to primary sequence are important in determining where major inhibition occurs.  相似文献

We present a general neural model for supervised learning of pattern categories which can resolve pattern classes separated by nonlinear, essentially arbitrary boundaries. The concept of a pattern class develops from storing in memory a limited number of class elements (prototypes). Associated with each prototype is a modifiable scalar weighting factor () which effectively defines the threshold for categorization of an input with the class of the given prototype. Learning involves (1) commitment of prototypes to memory and (2) adjustment of the various factors to eliminate classification errors. In tests, the model ably defined classification boundaries that largely separated complicated pattern regions. We discuss the role which divisive inhibition might play in a possible implementation of the model by a network of neurons.This work was supported in part by the Alfred P. Four different cleansing regimens were assessed in a prospective survey of 435 patients referred for barium enema. A regimen using simple dietary instructions and laxatives is as effective as a preliminary cleansing enema. It is suggested that routine cleansing enemas in outpatients represent an unnecessary and uncomfortable ritual that might usefully be abandoned.  相似文献
