We investigated the effects of the predatory backswimmer, Anisops sardea, on oviposition habitat selection of mosquitoes and other dipterans, and on community structure in experimental pools. We predicted that those dipteran species whose larvae were shown to be highly vulnerable to predation by Anisops would avoid Anisops pools when choosing an oviposition site. We established the following treatments in plastic tubs: (1) Control (without Anisops); (2) Free Anisops (ten Anisops within pool but not caged); (3) Caged Anisops (ten Anisops in cage). The pools were open to colonization by insects. We added resting stages of crustaceans and first instar larvae of the mosquitoes Culiseta longiareolata and Culex laticinctus. Among the dipteran species, Culiseta longiareolata, Culex laticinctus, Chironomus riparius (Chironomidae) and Forcipomyia sp. (Ceratopogonidae), only Culisetalarvae were highly vulnerable to predation. As predicted, based on larval vulnerability, Culiseta, but not the other species, avoided Anisops pools when ovipositing. Free Anisops reduced taxon richness. This reduction resulted largely from the elimination of the cladoceran Ceriodaphniasp. and Culiseta in most free Anisops pools. Thus, Anisops sardea structures the community, both by a behavioral response of prey to its presence and by consumption of prey.  相似文献

Abstract. Stimulation of floating peat by the introduction of poorly humified peat from four cut‐over bogs in The Netherlands was studied in a one‐year outdoor experiment. The pH of the various peat substrates was increased by adding different amounts of lime: 0–2 to 4–8 g‐CaCO3.kg‐1 fresh peat. Both peat type and lime addition were found to affect the buoyancy of the peat substrates. Low nutrient concentrations in the peat, together with a high bulk density, proved to be unfavourable for creating floating peat. Three of the peat types proved to be too acidic (pH < 4.5) to produce sufficient methane (ca. 400–600 μmol.L‐1), and buoyancy was only achieved if lime was mixed in with the peat. The smallest amount of lime added (2 g.kg‐1 fresh peat) was sufficient to maintain buoyancy for at least one year. Lime addition did not stimulate nutrient mobilization by mineralization, although P, N and K concentrations in the peat water were relatively high. It is concluded that floating peat can be initiated by the introduction of poorly humified peat. If the peat substrates are too acidic, lime can be added to stimulate buoyancy of the peat. Results are discussed in relation to restoration prospects.  相似文献

The Mouse Lymphoma Assay (MLA) Workgroup of the International Workshop on Genotoxicity Tests (IWGT) met on June 28th and 29th, 2002, in Plymouth, England. This meeting of the MLA group was devoted to discussing the criteria for assay acceptance and appropriate approaches to data evaluation. Prior to the meeting, the group conducted an extensive analysis of data from both the microwell and soft agar versions of the assay. For the establishment of criteria for assay acceptance, 10 laboratories (6 using the microwell method and 4 using soft agar) provided data on their background mutant frequencies, plating efficiencies of the negative/vehicle control, cell suspension growth, and positive control mutant frequencies. Using the distribution curves generated from this data, the Workgroup reached consensus on the range of values that should be used to determine whether an individual experiment is acceptable. In order to establish appropriate approaches for data evaluation, the group used a number of statistical methods to evaluate approximately 400 experimental data sets from 10 laboratories entered into a database created for the earlier MLA Workshop held in New Orleans [Environ. Mol. Mutagen. 40 (2002) 292]. While the Workgroup could not, during this meeting, make a final recommendation for the evaluation of data, a general strategy was developed and the Workgroup members agreed to evaluate this new proposed approach using their own laboratory data. This evaluation should lead to a consensus global approach for data evaluation in the near future.  相似文献

The existence of metal hyperaccumulator species demonstrates that plants have the genetic potential to remove toxic metals from contaminated soil. Possibly, one of the best-known hyperaccumulators is Thlaspi caerulescens. This species has been shown to accumulate very high Zn concentrations without manifesting any sign of toxicity. Thus, T. caerulescens represents an excellent experimental system for studying metal hyperaccumulation in plants as it relates to phytoremediation. In this article, we review the results of an investigation into the physiology, biochemistry, and molecular regulation of Zn transport and accumulation in T. caerulescens compared with a nonaccumulator relative T. arvense. Physiological studies focused on the use of 65Zn radiotracer flux techniques to characterize zinc transport and compartmentation in the root, and translocation to the shoot. Transport studies indicated that a number of Zn transport sites were stimulated in T. caerulescens, contributing to the hyperaccumulation trait. Thus, Zn influx into root and leaf cells, and Zn loading into the xylem was greater in T. caerulescens compared with the nonaccumulator T. arvense. The 4.5-fold stimulation of Zn influx into the roots of T. caerulescens was hypothesized to be due to an overexpression of Zn transporters in this species. Additionally, compartmental analysis (radiotracer wash out or efflux techniques) was used to show that Zn was sequestered in the root vacuole of T. arvense inhibiting Zn translocation to the shoot in this nonaccumulator species. Molecular studies focused on the cloning and characterization of Zn transport genes in T. caerulescens. Functional complementation of a yeast Zn transport-defective mutant with a T. caerulescens cDNA library constructed in a yeast expression vector resulted in the cloning of a Zn transport cDNA, ZNT1. Expression of ZNT1 in yeast allowed for a physiological characterization of this transporter. ZNT1 was shown to encode a high-affinity Zn transporter that can also mediate low-affinity Cd transport. Biochemical analyses indicated that enhanced Zn transport in T. caerulescens results from a constitutively high expression of ZNT1 in roots and shoots. These results suggest that overexpression of ZNT1 may be linked to an alteration of the Zn tolerance mechanism in this species.  相似文献

Linkage disequilibrium (LD) analysis has been promoted as a method of mapping disease genes, particularly in isolated populations, but has not yet been used for genome-screening studies of complex disorders. We present results of a study to investigate the feasibility of LD methods for genome screening using a sample of individuals affected with severe bipolar mood disorder (BP-I), from an isolated population of the Costa Rican central valley. Forty-eight patients with BP-I were genotyped for markers spaced at approximately 6-cM intervals across chromosome 18. Chromosome 18 was chosen because a previous genome-screening linkage study of two Costa Rican families had suggested a BP-I locus on this chromosome. Results of the current study suggest that LD methods will be useful for mapping BP-I in a larger sample. The results also support previously reported possible localizations (obtained from a separate collection of patients) of BP-I-susceptibility genes at two distinct sites on this chromosome. Current limitations of LD screening for identifying loci for complex traits are discussed, and recommendations are made for future research with these methods.  相似文献

Restoration projects are increasingly widespread and many promote habitat succession and the diversity and abundance of faunal communities. These positive effects on biodiversity and abundance may extend to enhancing the ecological functioning and resilience of previously degraded ecosystems, but this is rarely quantified. This study surveyed a 200-ha restoring coastal wetland and three control wetlands in the Maroochy River, eastern Australia to compare the effects of wetland restoration on the consumption of carrion and the biodiversity, abundance, and functional diversity of functionally important fish and crustaceans. Carrion consumption by fish and crustaceans was measured every 6 months from spring 2017 until spring 2021 for nine events using a combination of baited cameras and scavenging assays. We found restoration improved rates of carrion consumption and the biodiversity, functional diversity, and abundance of scavenger species. Despite positive effects on the diversity of scavengers and carrion consumption, the abundance of two species, longfin eels (Anguilla reinhardtii) and mud crabs (Scylla serrata), was the most important predictors of carrion consumption rates. The spatial distribution of carrion consumption was concentrated in areas with high saltmarsh extent, moderate to high mangrove extent, and high salinity, which also resembled the distribution of both longfin eels and mud crabs. We show that restoration can promote the rates of key ecological functions but that increases to functions are likely to be characterized by low functional redundancy and greater complementarity. Therefore, maintaining or increasing the abundance of functionally important species should become a key objective in future restoration projects.  相似文献

Staphylococcus aureus is frequently detected in patients with sepsis and thus represents a major health burden worldwide. CD4+ T helper cells are involved in the immune response to S. aureus by supporting antibody production and phagocytosis. In particular, Th1 and Th17 cells secreting IFN-γ and IL-17A, are involved in the control of systemic S. aureus infections in humans and mice.To investigate the role of T cells in severe S. aureus infections, we established a mouse sepsis model in which the kidney was identified to be the organ with the highest bacterial load and abundance of Th17 cells. In this model, IL-17A but not IFN-γ was required for bacterial control. Using Il17aCre × R26YFP mice we could show that Th17 fate cells produce Th17 and Th1 cytokines, indicating a high degree of Th17 cell plasticity. Single cell RNA-sequencing of renal Th17 fate cells uncovered their heterogeneity and identified a cluster with a Th1 expression profile within the Th17 cell population, which was absent in mice with T-bet/Tbx21-deficiency in Th17 cells (Il17aCre x R26eYFP x Tbx21-flox). Blocking Th17 to Th1 transdifferentiation in Th17 fate cells in these mice resulted in increased S. aureus tissue loads.In summary, we highlight the impact of Th17 cells in controlling systemic S. aureus infections and show that T-bet expression by Th17 cells is required for bacterial clearance. While targeting the Th17 cell immune response is an important therapeutic option in autoimmunity, silencing Th17 cells might have detrimental effects in bacterial infections.  相似文献

D-Psicose is a hexoketose monosaccharide sweetener, which is a C-3 epimer of D-fructose and is rarely found in nature. It has 70 % relative sweetness but 0.3 % energy of sucrose, and is suggested as an ideal sucrose substitute for food products. It shows important physiological functions, such as blood glucose suppressive effect, reactive oxygen species scavenging activity, and neuroprotective effect. It also improves the gelling behavior and produces good flavor during food process. This article presents a review of recent studies on the properties, physiological functions, and food application of D-psicose. In addition, the biochemical properties of D-tagatose 3-epimerase family enzymes and the D-psicose-producing enzyme are compared, and the biotechnological production of D-psicose from D-fructose is reviewed.  相似文献
