We studied the effects of tempol, an oxygen radical scavenger, on hydrosaline balance in rats with acute sodium overload. Male rats with free access to water were injected with isotonic (control group) or hypertonic saline solution (0.80 mol/l NaCl) either alone (Na group) or with tempol (Na-T group). Hydrosaline balance was determined during a 90 min experimental period. Protein expressions of aquaporin 1 (AQP1), aquaporin 2 (AQP2), angiotensin II (Ang II) and endothelial nitric oxide synthase (eNOS) were measured in renal tissue. Water intake, creatinine clearance, diuresis and natriuresis increased in the Na group. Under conditions of sodium overload, tempol increased plasma sodium and protein levels and increased diuresis, natriuresis and sodium excretion. Tempol also decreased water intake without affecting creatinine clearance. AQP1 and eNOS were increased and Ang II decreased in the renal cortex of the Na group, whereas AQP2 was increased in the renal medulla. Nonglycosylated AQP1 and eNOS were increased further in the renal cortex of the Na-T group, whereas AQP2 was decreased in the renal medulla and was localized mainly in the cell membrane. Moreover, p47-phox immunostaining was increased in the hypothalamus of Na group, and this increase was prevented by tempol. Our findings suggest that tempol causes hypernatremia after acute sodium overload by inhibiting the thirst mechanism and facilitating diuresis, despite increasing renal eNOS expression and natriuresis.  相似文献

We extend the concept of accessibility in temporal networks to model infections with a finite infectious period such as the susceptible-infected-recovered (SIR) model. This approach is entirely based on elementary matrix operations and unifies the disease and network dynamics within one algebraic framework. We demonstrate the potential of this formalism for three examples of networks with high temporal resolution: networks of social contacts, sexual contacts, and livestock-trade. Our investigations provide a new methodological framework that can be used, for instance, to estimate the epidemic threshold, a quantity that determines disease parameters, for which a large-scale outbreak can be expected.  相似文献

A simple and convenient synthesis of cholesterol esters from cholesterol and fatty acid anhydrides is described. The high, reproducible yield of cholesterol ester and the small excess of fatty acid used makes this method attractive for the preparation of cholesterol esters using difficultly obtained fatty acids.  相似文献

Dimethoxyethane, a good activator of phosphorylase b, has been used to study mechanisms of phosphorylase activation and the catalytic reaction. Activation can be explained best by an alteration of the allosteric equilibrium in favor of the active R conformation. Lesser effects are seen with phosphorylase a, and activation does not alter appreciably the equilibrium between the dimeric and tetrameric forms. With 20% 1,2-dimethoxyethane, the Vm value of phosphorylase b is 74% of that obtained in the presence of adenosine monophosphate. In the presence of 10% 1,2-dimethoxyethane, the Ki value for glucose inhibition is increased 3-fold, but inhibition by 1,5-gluconolactone is increased. The allosteric activation of glycogen phosphorylase results in a change in pK1 for the pH-activity profile. The formation of the dianionic form of the phosphoryl group of the coenzyme, pyridoxal phosphate, may account for this change. By analogy to the effects of anions and a change in dielectric on the acid hydroylsis of glucose 1-phosphate, it is suggested that the dianion of the coenzyme could stabilize the developing positive charge of an oxonium ion intermediate. Dimethoxyethane also affects the interaction of pyridoxal phosphate with phosphorylase. It influences the rates of both resolution and reconstitution. Good preparations of apophosphorylase a can be made by using 1,2-dimethoxyethane in the resolution medium.  相似文献

A fusion protein consisting of the TrpE protein and residues 166-211 of the Torpedo acetylcholine receptor alpha 1 subunit was produced in Escherichia coli using a pATH10 expression vector. Residues in the Torpedo sequence were changed by means of oligonucleotide-directed mutagenesis to residues present in snake alpha 1 subunit and rat nerve alpha 3 subunit which do not bind alpha-bungarotoxin. The fusion protein of the Torpedo sequence bound 125I-alpha-bungarotoxin with high affinity (IC50 = 2.5 x 10(-8) M from competition with unlabeled toxin, KD = 2.3 x 10(-8) M from equilibrium saturation binding data). Mutation of three Torpedo residues to snake residues, W184F, K185W, and W187S, had no effect on binding. Conversion of two additional Torpedo residues to snake, T191S and P194L, reduced alpha-bungarotoxin binding to undetectable levels. The P194L mutation alone abolished toxin binding. Mutation of three Torpedo alpha 1 residues to neuronal alpha 3-subunit residues, W187E, Y189K, and T191N, also abolished detectable alpha-bungarotoxin binding. Conversion of Try-189 to Asn which is present in the snake sequence (Y189N) abolished toxin binding. It is concluded that in the sequence of the alpha subunit of Torpedo encompassing Cys-192 and Cys-193, Try-189 and Pro-194 are important determinants of alpha-bungarotoxin binding. Tyr-189 may interact directly with cationic groups or participate in aromatic-aromatic interactions while Pro-194 may be necessary to maintain a conformation conductive to neurotoxin binding.  相似文献

Different isolates of influenza virus show a high degree of amino acid sequence variation in their surface glycoproteins. Conserved residues located in the substrate-binding pocket of the influenza virus neuraminidase are therefore likely to be involved in substrate binding or enzyme catalysis. In order to study the structure and function of the active site of this protein, a full-length cDNA clone of the neuraminidase gene from influenza A/Tokyo/3/67 was subcloned into aN M13 vector and amino acid substitutions were made in selected residues by using the oligonucleotide mismatch technique. The mutant neuraminidase genes were expressed from a recombinant SV40 vector, and the proteins were analyzed for synthesis, transport to the cell surface, and proper three-dimensional folding by internal and surface immunofluorescence. The mutant neuraminidase proteins were then assayed to determine the effect of the amino acid substitution on enzyme activity. Twelve of the 14 mutant proteins were correctly folded and were transported to the cell surface in a manner identical with that of the wild type. Two of these have full enzyme activity, but seven mutants, despite correct three-dimensional structure, have completely lost neuraminidase activity. Two mutants were active at low pH. The properties of the mutant enzymes suggest a possible mechanism of neuraminidase action.  相似文献

Diet-induced hyperhomocysteinemia produces endothelial and cardiac dysfunction and promotes thrombosis through a mechanism proposed to involve oxidative stress. Inducible nitric oxide synthase (iNOS) is upregulated in hyperhomocysteinemia and can generate superoxide. We therefore tested the hypothesis that iNOS mediates the adverse oxidative, vascular, thrombotic, and cardiac effects of hyperhomocysteinemia. Mice deficient in iNOS (Nos2−/−) and their wild-type (Nos2+/+) littermates were fed a high methionine/low folate (HM/LF) diet to induce mild hyperhomocysteinemia, with a 2-fold increase in plasma total homocysteine (P<0.001 vs. control diet). Hyperhomocysteinemic Nos2+/+ mice exhibited endothelial dysfunction in cerebral arterioles, with impaired dilatation to acetylcholine but not nitroprusside, and enhanced susceptibility to carotid artery thrombosis, with shortened times to occlusion following photochemical injury (P<0.05 vs. control diet). Nos2−/− mice had decreased rather than increased dilatation responses to acetylcholine (P<0.05 vs. Nos2+/+ mice). Nos2−/− mice fed control diet also exhibited shortened times to thrombotic occlusion (P<0.05 vs. Nos2+/+ mice), and iNOS deficiency failed to protect from endothelial dysfunction or accelerated thrombosis in mice with hyperhomocysteinemia. Deficiency of iNOS did not alter myocardial infarct size in mice fed the control diet but significantly increased infarct size and cardiac superoxide production in mice fed the HM/LF diet (P<0.05 vs. Nos2+/+ mice). These findings suggest that endogenous iNOS protects from, rather than exacerbates, endothelial dysfunction, thrombosis, and hyperhomocysteinemia-associated myocardial ischemia-reperfusion injury. In the setting of mild hyperhomocysteinemia, iNOS functions to blunt cardiac oxidative stress rather than functioning as a source of superoxide.  相似文献

The independent and interactive effects of light and simulated herbivory on growth and biomass allocation in the endangered emergent sedge, Scirpus ancistrochaetus Schuyler, were examined in a 15-week common garden experiment. Light level was manipulated through the use of shade cloth to attain shading levels of 0%, 30%, 63% and 90%, while herbivory was simulated by reducing plant height by 50% six weeks into the experiment. Multivariate Analysis of Variance (MANOVA) of final total (shoot + root) plant mass, leaf width and root to shoot ratio (R:S) indicated overall significant effects of both light and the interaction of light and clipping. Means of these variables decreased with decreasing light level. Furthermore, means of these variables were generally higher for clipped plants than unclipped plants in 0% shade, while they tended to be lower in clipped plants in all other levels of shade. Maximum leaf height was also significantly affected by light. Leaf height was similar in all treatments for the first 11 weeks. For the last four weeks of the experiment, however, leaf height was lowest in plants grown in 0% shade, highest in plants grown in 63 and 90% shade, and intermediate in plants grown in 30% shade. Clipped plants tended to be taller than unclipped plants at lower levels of shade, while the opposite was true at higher levels of shade, although effects were marginally significant. Both light and the interactive effect of light and clipping were important determinants of growth of S. ancistrochaetus in this experiment and therefore may be important influences on its distribution within and among pond habitats.  相似文献
