Aim

The abyssal Clarion-Clipperton Zone (CCZ), Pacific Ocean, is an area of commercial importance owing to the growing interest in mining high-grade polymetallic nodules at the seafloor for battery metals. Research into the spatial patterns of faunal diversity, composition, and population connectivity is needed to better understand the ecological impacts of potential resource extraction. Here, a DNA taxonomy approach is used to investigate regional-scale patterns of taxonomic and phylogenetic alpha and beta diversity, and genetic connectivity, of the dominant macrofaunal group (annelids) across a 6 million km2 region of the abyssal seafloor.

Location

The abyssal seafloor (3932–5055 m depth) of the Clarion-Clipperton Zone, equatorial Pacific Ocean.

Methods

We used a combination of new and published barcode data to study 1866 polychaete specimens using molecular species delimitation. Both phylogenetic and taxonomic alpha and beta diversity metrics were used to analyse spatial patterns of biodiversity. Connectivity analyses were based on haplotype distributions for a subset of the studied taxa.

Results

DNA taxonomy identified 291–314 polychaete species from the COI and 16S datasets respectively. Taxonomic and phylogenetic beta diversity between sites were relatively high and mostly explained by lineage turnover. Over half of pairwise comparisons were more phylogenetically distinct than expected based on their taxonomic diversity. Connectivity analyses in abundant, broadly distributed taxa suggest an absence of genetic structuring driven by geographical location.

Main Conclusions

Species diversity in abyssal Pacific polychaetes is high relative to other deep-sea regions. Results suggest that environmental filtering, where the environment selects against certain species, may play a significant role in regulating spatial patterns of biodiversity in the CCZ. A core group of widespread species have diverse haplotypes but are well connected over broad distances. Our data suggest that the high environmental and faunal heterogeneity of the CCZ should be considered in future policy decisions.  相似文献

Little is known about the effects of either long-term cutting with herbage removal or no management on the nutritional status of Agrostis capillaris and Festuca rubra meadows. We asked how management affected soil and herbage chemical properties, herbage yield and sward height. The experiment with cutting (C) and unmanaged (U) treatments was undertaken from 1999 to 2009 in the Jizera Mts., Czech Republic. Soil and herbage samples were taken from 2003 to 2009.Potassium was the only nutrient for which the soil concentration decreased under the C treatment. Soil K concentration was 175 and 210 mg kg?1 in the C and U treatments, respectively, in the last year of the experiment. Herbage yield as well as sward height were significantly higher in the unmanaged than in the cutting treatment in all years. Concentrations of P and K in the herbage biomass were lower in the cutting than in the unmanaged treatment, but concentrations of N were lower only in some years. Concentrations of Ca and Mg were not affected by the different treatments. A strong positive relationship between concentration in soil and herbage was recorded only for K.In contrast to K, ten years of contrasting management was too short a period to affect plant-available (Mehlich III) P, Ca and Mg concentrations, but it was long enough to affect herbage chemical properties and yield.  相似文献

Clean-up targets for toxic metals require that the site be “fit for purpose”. This means that targets are set with respect to defined receptors that reflect intended land-use. In this study, the likely threat of human exposure to toxic metals has been evaluated by simulating the human digestion process in vitro. The effects of key attributes (i.e. sample fraction size, pH, Kd and total metal concentrations) on the bioavailability of Cu and Ni were also investigated. Total metal concentration was the key explanatory factor for Cu and Ni bioavailability. A comparative ranking of metal concentrations in the context of tolerable daily intakes for Cu and Ni confirmed that the pH has the greatest impact on metals bioavailability. Rapid screening of key attributes and total toxic metal doses can reveal the relative hazard imposed on human, and this approach should be considered when defining threshold values for human protection.  相似文献

Aim

Typha latifolia causes serious problems in wet meadows by overgrowing and suppressing other native plants. To determine suitable management for T. latifolia control, we addressed the following question: What are the effects of long-term cutting at different frequencies (once or twice per year and no management) and biomass removal on cover and other characteristics of T. latifolia, and on sward productivity and plant species composition?

Location

Malá Strana nature reserve, Jizerské hory Mountains, Czechia.

Methods

A long-term experiment arranged in a randomised block design with three blocks was established in 2005. Data were collected from five treatments: unmanaged control; cutting once a year in June without biomass removal and with biomass removal; cutting twice per year in June and August without biomass removal and with biomass removal. Percentage cover of T. latifolia and other vascular plant species was visually estimated and T. latifolia characteristics (tiller density, height, dry-matter biomass [DMB] yield and litter), sward height and DMB yield were measured during 2005–2018 at the end of June.

Results and Discussion

Regular cutting once or twice per year regardless of cut biomass removal led to reductions in tiller density, height, litter and DMB yield of T. latifolia. Biomass removal had only a slight tendency to affect T. latifolia characteristics. The higher frequency of cutting significantly decreased the mean T. latifolia cover, litter and DMB yield. Cutting once or twice per year regardless of biomass removal led to successive changes in plant species composition but had no effect on the species richness and evenness.

Conclusions

Cutting at least once per year without biomass removal seems to be sufficient to achieve a decrease in DMB yield and litter of T. latifolia plants, and thereby maintain the wet-meadow vegetation without loss of species richness and also preventing the overgrowth of shrubs and trees.  相似文献

The pentameric WASH complex facilitates endosomal protein sorting by activating Arp2/3, which in turn leads to the formation of F-actin patches specifically on the endosomal surface. It is generally accepted that WASH complex attaches to the endosomal membrane via the interaction of its subunit FAM21 with the retromer subunit VPS35. However, we observe the WASH complex and F-actin present on endosomes even in the absence of VPS35. We show that the WASH complex binds to the endosomal surface in both a retromer-dependent and a retromer-independent manner. The retromer-independent membrane anchor is directly mediated by the subunit SWIP. Furthermore, SWIP can interact with a number of phosphoinositide species. Of those, our data suggest that the interaction with phosphatidylinositol-3,5-bisphosphate (PI(3,5)P2) is crucial to the endosomal binding of SWIP. Overall, this study reveals a new role of the WASH complex subunit SWIP and highlights the WASH complex as an independent, self-sufficient trafficking regulator.  相似文献
