A phylogenetic analysis of the Juncaceae was conducted to assess relationships among the genera Juncus, Luzula and five other small South American genera (Distichia, Marsippospermum, Oxychloë, Patosia and Rostkovia). We examined parallel datasets from organelles (mtDNA: atp1 gene, cpDNA: trnL intron, trnL-F intergenic spacer, rbcL gene) with respect to qualities relevant to the phylogenetic analysis of the Juncaceae. The main aim of our work was to produce a robust phylogeny of the Juncaceae validated by data from both organelles. Our data confirm the monophyly of the genus Luzula, but do not provide support for monophyly of the genus Juncus. The majority of taxa clustered within two subgenera, Agathryon and Juncus, morphologically supported by the presence or absence of bracteoles and cymose or racemose inflorescences, respectively. The subgenus Juncus is divided into two separate clades, the first closely related to the subgenus Agathryon and the second in the most basal part of the tree. Moreover, small South American genera clustered together with Juncus sect. Graminifolii and also with Juncus sect. Juncus. In fact, comparison of results from separate analyses of mitochondrial and plastome genes demonstrates that the general resolution of main topology of the atp1 tree is similar to the separate rbcL tree; the genus Juncus is better resolved, but the genus Luzula remains mainly polytomic.  相似文献

Phenol biodegradation was carried out in a batch system by the bacterial strain Cupriavidus metallidurans in the presence of potassium humate that was prepared by alkaline extraction from oxyhumolite. The experiments were focused on the assessment of the humate effect on biodegradation activity of the tested bacterial strain. The achieved results demonstrated that the humate has a positive influence on the biodegradation of phenol and reduces the incubation time necessary for phenol removal. Higher biodegradation rate and more intensive growth were observed during the cultivation in presence of humate in comparison to the cultivation without its addition. Adsorption of the humate on bacterial biomass was observed as well. Subsequently, a phenol biodegradation testing in a continuous-flow system using a biofilm reactor was also carried out. Although the reactor was inoculated by C. metallidurans only, the microbial composition under an aerobic non-aseptic condition during this long-term cultivation changed. The phenol removal efficiency obtained in the biofilm reactor was higher than 92% when phenol concentration in a treated medium was 1200 mg l−1.  相似文献

Conformational changes of the Na+/K+-ATPase isolated large cytoplasmic segment connecting transmembrane helices M4 and M5 (C45) induced by the interaction with enzyme ligands (i.e. Mg2+ and/or ATP) were investigated by means of the intrinsic tryptophan fluorescence measurement and molecular dynamic simulations. Our data revealed that this model system consisting of only two domains retained the ability to adopt open or closed conformation, i.e. behavior, which is expected from the crystal structures of relative Ca2+-ATPase from sarco(endo)plasmic reticulum for the corresponding part of the entire enzyme. Our data revealed that the C45 is found in the closed conformation in the absence of any ligand, in the presence of Mg2+ only, or in the simultaneous presence of Mg2+ and ATP. Binding of the ATP alone (i.e. in the absence of Mg2+) induced open conformation of the C45. The fact that the transmembrane part of the enzyme was absent in our experiments suggested that the observed conformational changes are consequences only of the interaction with ATP or Mg2+ and may not be related to the transported cations binding/release, as generally believed. Our data are consistent with the model, where ATP binding to the low-affinity site induces conformational change of the cytoplasmic part of the enzyme, traditionally attributed to E2 → E1 transition, and subsequent Mg2+ binding to the enzyme-ATP complex induces in turn conformational change traditionally attributed to E1 → E2 transition.  相似文献

Six non-native invasive fish species from the Slovak part of the Tisa River basin, namely Carassius gibelio, Pseudorasbora parva, Ameiurus melas, A. nebulosus, Lepomis gibbosus, Perccottus glenii, and three gobiid species from the Danube River, namely Neogobius melanostomus, N. fluviatilis and N. kessleri, were investigated for endohelminth parasites. The expanding Asian cestode Nippotaenia mogurndae (syn. Amurotaenia perccotti) (Nippotaeniidea) has been introduced to Europe with its invasive host P. glenii. Pumpkinseed, Lepomis gibbosus, is a new definitive host of Proteocephalus percae and it is reported as the second intermediate host of the bothriocephalidean cestode Triaenophorus nodulosus in Slovakia.  相似文献

Hemiparasitic plants gain virtually all mineral nutrients and water from their host plant whilst organic carbon is provided, at least in part, by their own photosynthetic activity, although their rates of assimilation are substantially lower than that found in non-parasitic plants. Hence, hemiparasites must gain at least some of their organic carbon heterotrophically from the host plant. Despite this, heterotrophic carbon gain by root hemiparasites has been investigated only for a few genera. We investigated heterotrophic carbon gain by two root hemiparasites, Rhinanthus minor L. and Euphrasia rostkoviana Hayne (Orobanchaceae), using natural abundance stable isotope (δ13C) profiles of both parasites attached to C3 (wheat) and C4 (maize) hosts coupled to a linear two-source isotope-mixing model to estimate the percentage of carbon in the parasite that was derived from the host. Both R. minor and E. rostkoviana attached to maize hosts were significantly more enriched in 13C than those attached to wheat hosts with R. minor becoming more enriched in 13C than E. rostkoviana. The natural abundance 13C profiles of both parasites were not significantly different from their wheat hosts, but were less enriched in 13C than maize hosts. Using a linear two-source isotope-mixing model, we estimated that R. minor and E. rostkoviana adult plants derive c. 50 and 25% of their carbon from their hosts, respectively. In light of these results, we hypothesise that repeatedly observed negative effect of competition for light on hemiparasites acts predominantly in early ontogenetic stages when parasites grow unattached or the abstraction of host nutrients is less effective.  相似文献

In the last 15 years substantial advances have been made to place isotope labels in native and glycosylated proteins for NMR studies and structure determination. Key developments include segmental isotope labeling using Native Chemical Ligation, Expressed Protein Ligation and Protein Trans-Splicing. These advances are pushing the size limit of NMR spectroscopy further making larger proteins accessible for this technique. It is just emerging that segmental isotope labeling can be used to define inter-domain interactions in NMR structure determination. Labeling of post-translational modified proteins like glycoproteins remains difficult but some promising developments were recently achieved. Key achievements are segmental and site-specific labeling schemes that improve resonance assignment and structure determination of the glycan moiety. We adjusted the focus of this perspective article to concentrate on the NMR applications based on recent developments rather than on labeling methods themselves to illustrate the considerable potential for biomolecular NMR.  相似文献

Some African grey parrots (Psittacus erithacus), the most famous being Pepperberg's parrot Alex, are able to imitate human speech and produce labels referentially. In this study, the aim was to teach ten African grey parrots from two laboratories to label items. Training three parrots from the first laboratory for several months with the Model/Rival method, developed by Pepperberg, in which two humans interact in front of the subject to demonstrate the use of a label, led to disappointing results. Similarly, seven parrots from the second laboratory, having been trained with several variants of Model/Rival attained little success. After the informal observation of the efficiency of other methods (i.e. learning to imitate labels either spontaneously or with specific learning methods and use of these labels referentially), four different teaching methods were tested with two birds: the Model/Rival; Repetition/Association which consisted of repeating a label and presenting the item only when the parrot produced the label; Intuitive in which the experimenter handled an item and repeated its name in front of the subject; Diffusion in which labels with either variable or flat intonation were played back daily to parrots. One bird learned three labels, one of which was used referentially, with the Repetition/Association method. He learned one label non-referentially with the Model/Rival but no labels were acquired using the other methods. The second bird did not learn any labels. This study demonstrates that different methods can be efficient to teach labels referentially and it suggests that rearing conditions and interindividual variability are important features when assessing learning ability of African grey parrots.  相似文献
