Summary A test system was set up where the build-up of a biofilm on a defined surface could be studied in a carbon source limited chemostat.The attachment of P. putida ATCC 11172 to glass when growing on L-asparagine was studied at different dilution rates (specific growth rates) from 0.1 to 1.5 h–1 The number of attached colony forming units (cfu) increased with dilution rate from 1×106 cfu/cm2 at 0.1 h–1 to 4×107 cfu/cm2 at 1.0 h–1 and then the attachment decreased to about 6×106 cfu/cm2 at higher dilution rates (1.1–1.5 h–1). The number of attached cfu was measured after 24 h exposure. The value of the maximum specific growth rate in batch culture was 0.6 h–1.The total amount of attached cell-mass followed roughly the same pattern as the viable count.The viable count of the cells suspended in the growth medium showed its lowest value at the same dilution rate as resulted in maximum adhesion.It was shown that the effect of growth rate on the biofilm build-up of P. putida is significant, and ought to be borne in mind when continuous culture systems are set up and results evaluated. 相似文献

The activity of the lysosomal dipeptidyl aminopeptidase II (DAP II) was measured by quantitative histochemical methods in the S1/S2 segments of the proximal tubule using freeze dried and celloidin mounted cryostat sections (FDC sections) of rat kidney. The methodological studies show that there is a linear relationship between the amount of reaction product and reaction time for the first 5 min, as well as section thickness between 4 and 10 microns. Maximal DAP II activities were demonstrated at pH 5.5. The Km of DAP II was about 2.3 mM. In addition to the methodological studies, DAP II activity was also measured in the proximal tubule (S1/S2 segments) of experimental animals (sham-operated and castrated male and female rats). Sham-operated females showed significantly higher DAP II activities than males. DAP II activity increased significantly in castrated males so that there were no significant differences between castrated males, sham-operated and castrated females. The quantitative histochemical results are largely in agreement with biochemical data published earlier. 相似文献

Biochemical fluorometric methods were used to investigate aminopeptidase A (APA; E.C.3.4.11.7) in the rat kidney homogenate and glomeruli and to compare it with aminopeptidase M (APM; E.C.3.4.11.2). It is shown that APA is a calcium-ion-dependent enzyme, while APM is not. To clarify the functional importance of APA and APM in the kidney, their activities were measured under the influence of angiotensins. Fluorimetric measurements in renal homogenate (with 2-naphthylamide derivatives as substrates), which represents mixed-enzyme tissue preparations containing a variety of peptidases besides APA and APM, showed a Km of 0.13 mM for APA and competitive inhibition of ANG II (K1 = 0.015 mM), and a Km of 0.24 for APM and competitive inhibition by ANG III (K1 = 0.003 mM). The remaining two angiotensins showed non-competitive inhibition of APA (ANG I, III) and APM (ANG I, II) in this preparation. For comparison purposes, fluorometric measurements were performed in microdissected glomeruli which contain only APA. A Km of 0.23 mM for the APA and a competitive inhibition of APA by ANG I and II were determined. Thus it was possible to show biochemically that APA is equivalent to angiotensinase A and that both APA and APM participate in angiotensin degradation in the kidney. This study describes the ultrastructural characteristics of the middle cerebral artery and its related neural elements in the squirrel monkey and baboon. The cytoarchitecture of the M-1 segment as well as that of the smaller extracerebral and intracerebral vessels is comparable in both animals.Smooth muscle elements are occasionally found within the intimai lining. The nerve bundles associated with vessels contain fewer myelinated fibers as the vessel diameter decreases. Comparative results are presented on the effectiveness of rat-liver S9 or microsomal mix (M mix) in activating cyclophosphamide (CP) and its ability to induce a clastogenic effect in human lymphocytes in vitro. Structural chromosome changes were analysed exclusively in 1st division (M1) metaphases post-exposure. A high genotoxic response was observed for both metabolizing systems used. With an exposure of 2 h to different concentrations of S9 or M mix, the highest aberration yields were always found for the highest protein content. For CP treatment times of 1, 2 or 4 h together with S9 mix (protein content 10 mg/ml) or M mix (4 mg/ml), the latter was more efficient. With both systems, a lower clastogenic effect of CP was found at 4 h exposure than at 1 h or 2 h. Only a weak cytotoxic effect, reflected mainly by the reduction in the percentage of 3rd cycle cells (M3), and measured in terms of the proportion of M1, M2 and M3 cells, was induced by both systems. 相似文献
