Prostate stromal cells may play binary roles in the process of prostate cancer development. As the first to be encountered
by infiltrating prostate cancer cells, prostate stromal cells form the first defense line against prostate cancer progression
and metastasis. However, interaction between prostate cancer and stromal cells may facilitate the formation of a tumor microenvironment
favoring cancer cell growth and survival. To establish an experimental system for studying the interaction between cancer
and stromal cells, we isolated three matched pairs of normal and cancer-associated human prostate stromal clones. In this
report, we describe the morphologic and behavioral characteristics of these cells and their effect on LNCaP prostate cancer
cells in co-culture. Unlike LNCaP prostate cancer cells, the isolated prostate stromal clones are large fibroblast-like cells
with a slow proliferation rate. Growth and survival of these clones are not affected by androgens. The stromal cells display
high resistance to serum starvation, while cancer-associated stromal clones have differentiated survival ability. In co-culture
experiments, the stromal cells protected some LNCaP prostate cancer cells from death by serum starvation, and cancer-associated
stromal clones showed more protection. This work thus established a panel of valuable human prostate stromal cell lines, which
could be used in co-culture to study the interaction between prostate cancer and prostate stromal cells. 相似文献

A number of ophiostomatoid fungi were isolated from the spruce-infesting bark beetle, Ips perturbatus and its galleries collected from felled spruce trees and logs in northern BC and the Yukon Territory. Isolates were identified to species using morphological characteristics, nuclear ribosomal DNA and partial β-tubulin gene sequences. Thirteen morphological and phylogenetic species were identified among the isolates. Leptographium fruticetum, Leptographium abietinum, Ophiostoma bicolor, Ophiostoma manitobense, O. piceaperdum, and eight undescribed species of the genus Ophiostoma and the anamorph genera Leptographium, Hyalorhinocladiella, Ambrosiella and Graphium. A number of these species, i.e. L. fruticetum, Hyalorhinocladiella sp. 2, O. bicolor and O. manitobense, were isolated repeatedly from I. perturbatus, while others, i.e. Graphium sp. 1 and O. piceaperdum, seemed to be␣sporadic associates. Among all the isolates, L. fruticetum had the highest relative dominance in this survey. A high frequency of occurrence of this species with the beetle may indicate a specific relationship between the two partners. 相似文献

Trisomy 13 in humans is the third most common autosomal abnormality at birth, after trisomy 21 and trisomy 18. It has a reported incidence of between 1:5,000 and 1:30,000 live births. It is associated with multiple abnormalities, many of which shorten lifespan. We describe here the first reported case of a baboon (Papio hamadryas) with trisomy of chromosome 17, which is homologous to human chromosome 13. The trisomic infant was born to a consanguineous pair of baboons and had morphological characteristics similar to those observed in human trisomy 13, including bilateral polydactyly in the upper limbs, a patent foramen ovale, and pyelectasis. Molecular DNA analysis using human chromosome 13 markers was consistent with the affected infant inheriting two copies of chromosome 17 derived from the same parental chromosome. This trisomy was, therefore, due to either an error in meiosis II or the result of postzygotic nondisjunction. The parental origin, however, could not be determined. 相似文献

Increasing evidence indicates that replicative senescence and premature endothelial senescence could contribute to endothelial dysfunction. This study aims at testing the hypothesis that a high-fat diet may lead to premature vascular endothelial senescence in a nonhuman primate model. We isolated endothelial cells from left and right femoral arteries in 10 baboons before and after a 7-wk high-fat dietary treatment. We compared the morphological alterations, replicative capacities, and senescence-associated beta-galactosidase activities (SA-beta-gal) at these two time points. We found that high-fat diet increased the prevalence of endothelial senescence. Endothelial replicative capacities declined dramatically, and SA-beta-gal activities increased significantly in postdietary challenge. There was no change in telomeric length using quantitative flow fluorescence in situ hybridization analysis, suggesting that some stressors lead to cell senescence independent of telomere dysfunction. Our findings that high-fat diet causes endothelial damage through the premature senescence suggest a novel mechanism for the diet-induced endothelial dysfunction. 相似文献

Adventitious root development was investigated in Typha glauca plants grown under experimental conditions with the previous year's dead, sterile stalk either emerged above or submerged below the surface of Hoagland's solution. Adventitious roots emerged from buds in which most primordia had been earlier formed. Most roots elongated to 14–19 cm in 3–4 weeks and produced abundant lateral roots to their tips. Root apical meristem organization was typically monocotyledonous with a single tier of ground meristem/protoderm over the procambium. The ground meristem had zones of periclinal divisions in its innermost and outermost layers; the innermost layer initiated the endodermis and midcortex, and the outermost layers initiated the hypodermis. Crystalliferous cells with raphides were produced in the midcortex, and aerenchyma resulted from the radial expansion of schizogenous air spaces and some lysigeny in the midcortex. The procambium produced a vascular cylinder with 10–13 phloem and xylem poles, 6–9 large metaxylem elements, and central sclerenchyma. As roots stopped elongating, they narrowed, the vascular cylinder diminished in size, typical aerenchyma was lost from the cortex, crystal production ceased, and the rootcap diminished in size with its storage starch used up. Growth was determinate in these adventitious roots. The results suggested that a periclinally derived outer ground meristem was a prerequisite for a hypodermis, which, in turn, was necessary as a structural framework for aerenchyma. Studies have shown that after controlling for the effects of body size on brain size, the brains of adult humans, rhesus monkeys, and chimpanzees differ in relative size, where males have a greater volume of cerebral tissue than females. We assess whether head circumference sexual dimorphism is present during early development by evaluating sex differences in relative head circumference in living fetuses and infants within the first year of life. Head circumference is used as a proxy for brain size in the fetus and infant. Femur length is used as a proxy for body length in the fetus. Ultrasonography was used to obtain fetal measures, and anthropometry was used to obtain postnatal measures in humans, rhesus monkeys, baboons, and common marmosets. We show that statistically significant but low levels of head circumference sexual dimorphism are present in humans, rhesus monkeys, and baboons in early life. On average, males have head circumferences about 2% larger than females of comparable femur/body length in humans, rhesus monkeys, and baboons. No evidence for head circumference sexual dimorphism in the common marmoset was found. Dimorphism was present across all body size ranges. We suggest that head circumference sexual dimorphism emerges largely postnatally and increases throughout maturation, particularly in humans who reach adult dimorphism values greater than the monkeys. We suggest that brain dimorphism is not likely to impose an additional energetic burden to the gestating or lactating mother. Finally, some of the problems with ascribing functional significance to brain size sexual dimorphism are discussed, and the energetic implications for brain size sexual dimorphism in infancy are assessed. 相似文献
