Apolipoprotein AI (apoAI), the major protein component of HDL, is one of the best predictors of coronary artery disease (CAD), with high apoAI and HDL levels being correlated with low occurrences of CAD. The primary function of apoAI is to recruit phospholipid and cholesterol for assembly of HDL particles. Like other exchangeable apolipoproteins, lipid-free apoAI forms a mixture of different oligomers even at 1.0 mg/mL. This self-association property of the exchangeable apolipoproteins is closely associated with the lipoprotein-binding activity of this protein family. It is unclear if the self-association property of apolipoprotein is required for its lipoprotein-binding activity. We developed a novel method for engineering an oligomeric protein to a monomeric, biologically active protein. Using this method, we generated a monomeric mouse apoAI mutant that is active. This mutant contains the first 216 residues of mouse apoAI and replaces six hydrophobic residues with either polar or smaller hydrophobic residues at the defined positions (V118A/A119S/L121Q/T191S/T195S/T199S). Cross-linking results show that this mutant is greater than 90% monomeric at 8 mg/mL. CD, DSC, and NMR results indicate that the mutant maintains an identical secondary, tertiary structure and stability as those of the wild-type mouse apoAI. Lipid-binding assays suggest that the mutant shares an equal lipoprotein-binding activity as that of the wild-type apoAI. In addition, both the monomeric mutant and the wild-type protein make nearly identical rHDL particles. With this monomeric mouse apoAI, high-quality NMR data has been collected, allowing for the NMR structural determination of lipid-free apoAI. On the basis of these results, we conclude that this apoAI mutant is a monomeric, active apoAI useful for structural determination.  相似文献

目的:以甘油为底物构建高效的3-羟基丙酸生产菌株。方法:以自身携带乙醛脱氢酶的E.coli BL21(DE3)plysS作为宿主,异源表达源自Klebsiella pneumoniae的甘油脱水酶基因dhaB。结果:重组菌E.coli HP获得的甘油脱水酶比活力在1.0mmol/L IPTG的诱导下达到了77.2 U/mg,摇瓶条件下,3-HP的最大产量为5.44 g/L,摩尔转化率为53%,该产量比目前报道的最高水平(4.4 g/L)提高了23.6%。结论:重组菌株E.coli HP实现了甘油向3-羟基丙酸(3-HP)的高效生物转化。  相似文献

应用地学统计学的原理和方法方法研究了不同时期桃-点叶蝉及草间小黑蛛种群的空间结构和空间相关性。结果表明,不同时期桃-点叶蝉种群的半变异函数曲线皆为球型,其空间格局为聚集型,变程在25.71-37.14m之间,草间小黑蛛种群的拟合并变异函数也表现为球型,呈聚集空间格局,空间变程在25.12-44.06m之间,并与桃-点叶蝉种群在数量和空间上有较强的追随关系,说明草间小黑蛛种群是桃一点叶蝉种群的优势种天敌。  相似文献

Bacterial peptidoglycan is the cell wall component responsible for various biological activities. Its cytoplasmic precursor UDP-N-acetylmuramyl pentapeptide is biosynthesized by the first six enzymes of peptidoglycan synthetic pathways (Mur enzymes), which are all proved to be important targets for antibiotic screening. In our present work, the genes encoding Mur enzymes from Escherichia coli were co-expressed in the cell-free protein synthesis (CFPS) system, and the activities of Mur enzymes derived from CFPS system were validated by the synthesis of the final product UDP-N-acetylmuramyl pentapeptide. Then this in vitro reconstituted Mur biosynthetic pathway was used to screen a panel of specific antisense oligonucleotides for MurA and MurB. The selected oligonucleotides were proved to eliminate the expression of Mur enzymes, and thus inhibit the Mur biosynthetic pathway. The present work not only developed a rapid method to reconstruct and regulate a biosynthetic pathway in vitro, but also may provide insight into the development of novel antibiotics targeting on peptidoglycan biosynthetic pathway.  相似文献

探讨缺氧环境下,白细胞介素8(Interleukin-8,IL-8)对人骨髓间充质干细胞(Human bone marrow mesenchymal stem cells,hBMSC)增殖和自噬能力的影响以及机制。在缺氧模型下,未进行刺激的hBMSC为缺氧对照组;以100μmol/L人IL-8蛋白刺激的MSC为IL-8组;若先添加50μmol/L MK2206(Akt蛋白抑制剂)培养30 min,然后再添加100μmol/L IL-8则为Akt抑制剂组,在正常条件下培养的MSC为正常对照组。利用Ed U细胞增殖实验、TUNEL细胞凋亡实验、Western blotting或ELISA等实验分别检测各组MSC细胞Ed U标记阳性细胞的数目、细胞凋亡、自噬蛋白(LC-3)和Akt/STAT3等蛋白的表达。相对于缺氧对照组和Akt抑制剂组,IL-8明显提高hBMSC增殖和细胞自噬,并降低hBMSC的凋亡率,IL-8组hBMSC的Akt、STAT3和VEGF等蛋白表达增高。结果表明,缺氧环境下,IL-8通过Akt-STAT3通路发挥对MSC的保护作用,对保护MSC抗缺血缺氧性损伤,促进MSC在再生医学中应用具有重要意义。  相似文献

Bacterial wilt caused by Ralstonia solanacearum is a serious soil-borne disease of Solanaceae crops. In this study, the soil microbial effects of silicon-induced tomato resistance against R. solanacearum were investigated through pot experiment. The results showed that exogenous 2.0 mM Si treatment reduced the disease index of bacterial wilt by 19.18 % to 52.7 % compared with non-Si-treated plants. The uptake of Si was significantly increased in the Si-treated tomato plants, where the Si content was higher in the roots than that in the shoots. R. solanacearum inoculation resulted in a significant increase of soil urease activity and reduction of soil sucrase activity, but had no effects on soil acid phosphatase activity. Si supply significantly increased soil urease and soil acid phosphatase activity under pathogen-inoculated conditions. Compared with the non-inoculated treatment, R. solanacearum infection significantly reduced the amount of soil bacteria and actinomycetes by 52.5 % and 16.5 %, respectively, but increased the ratio of soil fungi/soil bacteria by 93.6 %. After R. solanacearum inoculation, Si amendments significantly increased the amount of soil bacteria and actinomycetes and reduced soil fungi/soil bacteria ratio by 53.6 %. The results suggested that Si amendment is an effective approach to control R. solanacearum. Moreover, Si-mediated resistance in tomato against R. solanacearum is associated with the changes of soil microorganism amount and soil enzyme activity.  相似文献

Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms underlying colonization of Acremonium spp. remain unclear.In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng,enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin biosynthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid(IAA) and jasmonic acid(JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. notoginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate(Me JA)(2–15 μmol/L) and 1-naphthalenacetic acid(NAA)(10–20 μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and mi R393 boverexpressing lines, and the colonization was rescued by Me JA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. D212 in host plants.  相似文献

诺加霉素是重要的蒽环类抗肿瘤抗生素,由黑胡桃链霉菌ATCC27451发酵产生。本研究从诺加霉素产生茵中克隆得到560bp的氨基甲基化酶(snogA)编码基因片段,并将其插入基因整合型质粒pKCll39的多克隆位点,构建得到基因中断质粒pLMX-3-58。通过接合转移和同源重组,构建得到氨基甲基化酶编码基因被中断的重组菌株删-3-59。基因重组突变株基因型验证结果表明,中断质粒以正确方式整合入基因组,将氨基甲基化酶编码基因中断。发酵验证结果表明,重组茵株发酵产物中不含有诺加霉素。本研究表明snogA基因在诺加霉素生物合成途径中是必需的。这为进一步阐明诺加霉素生物合成途径和组合生物合成改造诺加霉素提供了参考。  相似文献
