本文报告了我国463名10-19岁儿童青少年的最大有氧活动能力的发展特征。在青春早期,男女童的最大吸氧量绝对值均随年龄增长而增加,男童由1.75升/分增至3.10升/分,女童由1.44升/分增至2.07升/分,女童增长较少;以后女童即稳定于这一水平,男童仍略有增长。按身高及按最大心率计标的相对值与其有相似的特征。按体重和瘦体重计算的相对值,在男女童都未见随年龄增长的规律。男童VO2max绝对值及各  相似文献

摘要 目的:调查社区2型糖尿病(T2DM)患者血糖自我管理水平及分析T2DM并发糖尿病周围神经病变(DPN)的影响因素。方法:于2016年6月~2017年6月期间采用整群随机抽样法随机抽取苏州市6个社区符合纳排标准的539例T2DM患者进行问卷调查,了解社区T2DM患者血糖自我管理水平情况,对患者进行体格检查并检测血生化指标,统计社区T2DM患者的DPN发生情况,采用多因素logistic回归分析T2DM患者并发DPN的影响因素。结果:本次研究共发放调查问卷539份,实际回收531份,其中T2DM并发DPN者86例,根据是否并发DPN将所有入选患者分为DPN组(n=86)和无DPN组(n=445)。社区T2DM患者的糖尿病自我管理行为量表(SDSCA)平均得分率为(49.38±5.23)%,DPN组和无DPN组在病程、体质量指数(BMI)、腰围、糖化血红蛋白(HbAlc)、空腹胰岛素(FINS)、低 密 度 脂 蛋 白(LDL-C)、血 尿 素 氮(BUN)、血肌酐(Cr)、合并外周动脉疾病(PAD)、合并糖尿病视网膜病变(DR)中比较差异有统计学意义(P<0.05)。多因素logistic回归分析结果显示:病程≥7年、HbAlc≥8 mmol/L、合并PAD、合并DR、BMI≥25 kg/m2是社区 T2DM患者并发DPN的危险因素(P<0.05)。结论:苏州市6个社区的T2DM患者血糖自我管理水平较低,且T2DM并发DPN的概率较高,病程≥7年、HbAlc≥8 mmol/L、合并PAD、合并DR、BMI≥25 kg/m2均是社区 T2DM患者并发DPN的危险因素,临床可对上述危险因素采取积极有效的措施,以有效降低T2DM并发DPN的发生率。  相似文献

A large number of post‐translational modifications (PTMs) in proteins are buried in the unassigned mass spectrometric (MS) spectra in shot‐gun proteomics datasets. Because the modified peptide fragments are low in abundance relative to the corresponding non‐modified versions, it is critical to develop tools that allow facile evaluation of assignment of PTMs based on the MS/MS spectra. Such tools will preferably have the ability to allow comparison of fragment ion spectra and retention time between the modified and unmodified peptide pairs or group. Herein, MMS2plot, an R package for visualizing peptide‐spectrum matches (PSMs) for multiple peptides, is described. MMS2plot features a batch mode and generates the output images in vector graphics file format that facilitate evaluation and publication of the PSM assignment. MMS2plot is expected to play an important role in PTM discovery from large‐scale proteomics datasets generated by liquid chromatography‐MS/MS. Exosomes are membrane‐bound extracellular vesicles that are produced in the endosomal compartment of most mammalian cell types and then released. Exosomes are effective carriers for the intercellular material transfer of material that can influence a series of physiological and pathological processes in recipient cells. Among loaded cargoes, non‐coding RNAs (ncRNAs) vary for the exosome‐producing cell and its homeostatic state, and characterization of the biogenesis and secretion of exosomal ncRNAs and the functions of these ncRNAs in skeletal muscle myogenesis remain preliminary. In this review, we will describe what is currently known of exosome biogenesis, release and uptake of exosomal ncRNAs, as well as the varied functions of exosomal miRNAs in skeletal muscle myogenesis.  相似文献

The arrest of meiotic prophase in mammalian oocytes within fully grown follicles is dependent on cyclic adenosine monophosphate (cAMP) regulation. A large part of cAMP is produced by the Gs-linked G-protein-coupled receptor (GPR) pathway. In the present study, we examined whether GPR3 is involved in the maintenance of meiotic arrest in porcine oocytes. Expression and distribution of GPR3 were examined by western blot and immunofluorescence microscopy, respectively. The results showed that GPR3 was expressed at various stages during porcine oocyte maturation. At the germinal vesicle (GV) stage, GPR3 displayed a maximal expression level, and its expression remained stable from pro-metaphase I (MI) to metaphase II (MII). Immunofluorescence staining showed that GPR3 was mainly distributed at the nuclear envelope during the GV stage and localized to the plasma membrane at pro-MI, MI and MII stages. RNA interference (RNAi) was used to knock down the GPR3 expression within oocytes. Injection of small interfering double-stranded RNA (siRNA) targeting GPR3 stimulated meiotic resumption of oocytes. On the other hand, overexpression of GPR3 inhibited meiotic maturation of porcine oocytes, which was caused by increase of cGMP and cAMP levels and inhibition of cyclin B accumulation. Furthermore, incubation of porcine oocytes with the GPR3 ligand sphingosylphosphorylcholine (SPC) inhibited oocyte maturation. We propose that GPR3 is required for maintenance of meiotic arrest in porcine oocytes through pathways involved in the regulation of cAMP and cGMP.  相似文献

Purpose

The rodent retina does not exhibit a positive OFF-response in the electroretinogram (ERG), which makes it difficult to evaluate its OFF-pathway functions in vivo. We studied the rod-driven OFF pathway responses by using a dark-adapted 10-Hz flicker ERG procedure in mouse.

Materials and Methods

Conventional ERGs and 10-Hz dark-adapted flicker ERGs were obtained in wild-type mice (C57BL/6), in mice with pure rod (cpfl1) or pure cone (rho−/−) function, and in nob1 mice which have a selective ON-pathway defect. To isolate the response from ON or OFF pathway, glutamate analogs 2-amino-4-phosphobutyric acid (APB, an ON pathway blocker) and cis-2, 3-piperidine-dicarboxylic acid (PDA, an OFF pathway blocker), were injected intravitreally.

Results

The amplitude-intensity profile of the dark-adapted 10-Hz flicker ERG in the wild-type mice exhibits two peaks at middle and high light intensities. The two peaks represent rod- and cone-driven responses respectively. In APB-treated C57BL/6 mice and in nob1 mice, the dark-adapted ERG b-waves were absent. However, both rod- and cone-driven OFF pathway responses were evident with flicker ERG recording. At middle light intensities that activate only rod system, the flicker ERG responses in saline-injected nob1 mice were similar to those in APB-injected cpfl1 mice and wild-type mice. These responses are sensitive to PDA. The amplitudes of these rod-driven OFF pathway responses were approximately 20% of the total rod-driven flicker ERG responses.

Conclusion

We demonstrate that the rod-OFF bipolar cell pathway is functional in the outer retina. The dark-adapted flicker ERG is practical for the evaluation of rod- and cone-driven responses, and the residual OFF pathway signals in subjects with ON pathway defects.  相似文献

In non-mammalian vertebrates, the pineal gland functions as the central pacemaker that regulates the circadian rhythms of animal behavior and physiology. We generated a transgenic zebrafish line [Tg(Gnat2:gal4-VP16/UAS:nfsB-mCherry)] in which the E. coli nitroreductase is expressed in pineal photoreceptor cells. In developing embryos and young adults, the transgene is expressed in both retinal and pineal photoreceptor cells. During aging, the expression of the transgene in retinal photoreceptor cells gradually diminishes. By 8 months of age, the Gnat2 promoter-driven nitroreductase is no longer expressed in retinal photoreceptor cells, but its expression in pineal photoreceptor cells persists. This provides a tool for selective ablation of pineal photoreceptor cells, i.e., by treatments with metronidazole. In the absence of pineal photoreceptor cells, the behavioral visual sensitivity of the fish remains unchanged; however, the circadian rhythms of rod and cone sensitivity are diminished. Brief light exposures restore the circadian rhythms of behavioral visual sensitivity. Together, the data suggest that retinal photoreceptor cells respond to environmental cues and are capable of entraining the circadian rhythms of visual sensitivity; however, they are insufficient for maintaining the rhythms. Cellular signals from the pineal photoreceptor cells may be required for maintaining the circadian rhythms of visual sensitivity.  相似文献

施用有机物是作物增产、增加土壤有机质和改善土壤生物学性状的有效措施。在大田滴灌条件下,采用土壤酶学和微生物平板培养方法,研究了常规施肥(CF)减量20%—40%,配施3000、6000 kg/hm2有机类肥料对棉花产量、土壤微生物数量及土壤酶活性的影响。结果表明:降低CF用量20%—40%情况下,滴灌棉田配施3000、6000 kg/hm2的有机类肥料可获得与CF处理相持平的产量(4945—4978 kg/hm2),有机肥和生物有机肥配施处理间的棉花产量差异不显著(P0.05)。细菌、放线菌和假单胞杆菌数量均随有机肥用量的增加而升高,不同类有机肥配施间表现为OF+BFBFOF;真菌数量则随有机肥施量升高而降低,其中OF+BF配施处理最为显著。有机无机肥配施显著提高了土壤酶活性,80%CF和60%CF与有机肥配施处理土壤碱性磷酸酶、荧光素二乙酸酯酶(FDA)、β-葡萄糖苷酶和脱氢酶的活性比CF处理分别升高了3.8%、17%、18%、55%和10.1%、19%、20%、60%,不同类型有机肥对土壤酶活性影响差异不显著。土壤细菌/真菌比、土壤放线菌/真菌和假单胞杆菌/真菌比均随有机肥施量的增加而升高,施用化肥或有机肥均显著降低了假单胞杆菌/细菌比。细菌、放线菌和假单胞杆菌数量与土壤脱氢酶、β-葡萄糖苷酶、碱性磷酸酯酶和荧光素二乙酸酯酶活性均呈显著或极显著正相关,真菌数量与4种酶活性呈负相关。CF减量20%—40%配施以3000、6000 kg/hm2有机肥不仅不会导致棉花减产,而且对提高土壤酶活性、调节土壤细菌、真菌、放线菌群落组成结构,改善北疆绿洲滴灌棉田土壤生物学性状有显著作用。  相似文献
