Alginates being depolymerized during their alkaline extraction, reducing extraction time could help producing higher rheological quality alginates. The purpose of the present work is to study fresh Laminaria digitata destructuration during alkaline extraction and its link to extraction kinetics. Both alginate extraction yield and mean diameter of algae particles were followed for different values of agitation level and initial size of algae pieces. Results highlighted the existence of a link between extraction yield and algal destructuration. Those elements and the specificity of L.digitata structure have been taken into account to propose a kinetics model based on a fluid-particle reaction with decreasing size particles. The model parameters have been adjusted thanks to acquisition data and its predictive capacity was assessed by validation data. Provided predictions appeared to be relevant and the model structure suitability was confirmed, as extraction yield kinetics specific shape was quite reliably described. 相似文献

Sequential detections of different proteins on Western blot save time and precious samples. The main problem concerning reprobing is that stripping buffers can unbind both the antibody and the tested antigen. An original reprobing method has been set up based on horseradish peroxidase (HRP) inhibition after enhanced chemiluminescence detection. Instead of removing previously fixed antibodies as common stripping buffers do, the HRP activity linked to the secondary antibody is irreversibly inhibited by excess of hydrogen peroxide. A 15-min incubation allows one to perform at least five different sequential detections without losing significant amounts of blotted proteins. 相似文献

Chenopodium album L. (fat-hen) with a Ser264-Gly mutation is resistant to photosystem II-inhibiting herbicides like the triazinone metamitron, a key herbicide in sugar beet. In recent years, this resistant biotype may cause unsatisfactory weed control in Belgian sugar beet. However, the dimension of the problem was yet unknown. Therefore, a survey was conducted in 2008 covering the whole Belgian sugar beet area. In randomly selected fields, C. album plants surviving weed control were counted and sampled. First, the number of surviving plants was used to estimate the prevalence of fields with unsatisfactory control and to classify the surveyed fields. Then, the share of the resistant biotype in each field was determined with cleaved amplified polymorphic sequence-analysis (CAPS-analysis) on sampled leaves. Finally, all results were visualised on the map of Belgium. Twenty percent of the fields had more than 500 surviving plants per hectare and were thus classified as fields with unsatisfactory C. album control. The resistant biotype was present in 95% of these fields and even in 74% of the sampled fields with good weed control. No pattern was found during mapping. These results indicate that the metamitron-resistant biotype has spread over the whole sugar beet area but that it is not (yet) causing severe problems in every field. To get a more accurate estimation of the portion of resistant plants in the field and the effect of herbicide treatment on this biotype, an elaborate survey will be conducted in 2010 on fields that have both untreated and treated plots installed. 相似文献

The molecular basis of Plasmodium vivax chloroquine (CQ) resistance
is still unknown. Elucidating the molecular background of parasites that are
sensitive or resistant to CQ will help to identify and monitor the spread of
resistance. By genotyping a panel of molecular markers, we demonstrate a similar
genetic variability between in vitro CQ-resistant and sensitive phenotypes of
P. vivax parasites. However, our studies identified two
loci (MS8 and MSP1-B10) that could be used to discriminate
between both CQ-susceptible phenotypes among P. vivax isolates in
vitro. These preliminary data suggest that microsatellites may be used to identify
and to monitor the spread of P. vivax-resistance around the

Alkylglycerols (alkyl-Gro) are ether lipids abundant in shark liver oil (SLO), and oral SLO or alkyl-Gro mix from this source have several in vivo biological activities including stimulation of haematopoiesis an immunological defences, or anti-tumour and anti-metastasis activities in vivo. Composition of natural alkyl-Gro mix contains several alkyl-Gro varying by chain length and unsaturation, and individual anti-tumour activity of each molecule present in natural mix remained unknown. We synthesized six prominent constituents of natural alkyl-Gro mix, namely 12:0, 14:0 16:0, 18:0, 16:1 n-7, and 18:1 n-9 alkyl-Gro. Using an in vivo model of grafted tumour in mice (3LL cells), we studied and compared the oral anti-tumour and anti-metastasis activities of each of these 6 alkyl-Gro. 16:1 and 18:1 alkyl-Gro showed strong activity in reducing lung metastasis number, while saturated alkyl-Gro had weaker (16:0) or no (12:0, 14:0, 18:0) effect. Spleen weights at day 20 after graft were also measured and showed tremendous variations depending on the treatment. Tumour graft resulted in a raise in spleen weight in control group, this raise was nearly abolished in 16:1 and 18:1 alkyl-Gro-treated mice, and was reduced in 14:0 and 16:0 alkyl-Gro-treated mice. Conversely, 18:0 alkyl-Gro-treated mice showed spleen weigh raise as compared with untreated grafted mice. Fatty acid acylation is a functionally important modification of proteins. In the liver, however, acylated proteins remain largely unknown. This work was aimed at investigating fatty acid acylation of proteins in cultured rat hepatocytes. Incubation of these cells with [9,10-3H] myristic acid followed by two-dimensional electrophoresis separation of the delipidated cellular proteins and autoradiography evidenced the reproducible and selective incorporation of radioactivity from the precursor into 18 well-resolved proteins in the 10--120 kDa range and the 4--7 pH range. Radiolabeling of these proteins resulted from covalent linkage to the precursor [9,10-3H] myristic acid or to its elongation product, palmitic acid. The majority of the covalent linkages between the proteins and the fatty acids were broken by base hydrolysis, which indicated that the linkage was of thioester or ester-type. Only one of the studied proteins was attached to myristic acid via an amide linkage which resisted the basic treatment but was broken by acid hydrolysis. After incubation with [9,10-3H] palmitic acid, only two proteins previously detected with myristic acid were radiolabeled. Finally, the identified acylated proteins may be grouped into two classes: proteins involved in signal transduction (the alpha subunit of a heterotrimeric G protein and several small G proteins) and cytoskeletal proteins (cytokeratins, actin). 相似文献

Palynomorphs are reported for the first time from the Nishihiro Formation (Wakayama Prefecture, Outer Zone of southwest Japan). The Nishihiro Formation consists of brackish to shallow marine deposits, dated as late Barremian to Aptian from geological correlations. Spores prevail in the assemblage, representing Filicopsida (mainly Cyatheaceae and Anemiaceae), Marchantiopsida and Lycopsida. The pollen assemblage is dominated by Coniferales, whereas Gnetales and Bennettitales/Cycadales are only rarely observed. Moreover, we report angiosperm pollen grains of the genus Retimonocolpites for the first time in the Early Cretaceous sediments of Japan. Pollen grains of the Retimonocolpites Group are typical of early angiosperms and commonly found in assemblages from the early to mid-Cretaceous of all paleofloristic provinces. Until this paper, the oldest angiosperm fossils in Japan were represented by a single seed and a wood reported from the Albian of Hokkaido. The oldest reliable angiosperm pollen grains were reported in Hokkaido from the Cenomanian, and in Honshu from the Coniacian. Thus, Retimonocolpites pollen grains reported in the present study represent the oldest record of angiosperms in Japan. They indicate an appearance of the angiosperms in Japan older than thought until now, which is consistent with that proposed elsewhere in eastern Asia. 相似文献
