本文用凝集素为探针,探索糖复合物在胚泡着床中的作用,报道了与甘露糖苷有专一结合的伴刀豆凝集素(ConA)有明显的抗小鼠胚泡着床作用。妊娠4d的小鼠,每只子宫角中注入Con A 25μg,22只子宫角中只有4只子宫角有胚泡着床,着床率为18.2%,与生理盐水对照组的着床率87.5%相比有明显差异。将相同剂量的Con A先与0.4mol/L α-甲基-D-甘露糖苷温育1—2h后再注入子宫,20只子宫角中有15只子宫角有胚泡着床,着床率提高到75%。用辣根过氧化物酶直接标记法证明,着床前子宫内膜细胞表面有Con A受体存在,并随着妊娠天数而增加,尤其是间质细胞,发情期时时为阴性反应,到着床期蜕膜细胞膜表面呈现出大量Con A受体。提示精复合物在着床中的重要作用。与甘露糖苷同样专一结合的,但寡糖结构专一性与Con A不同的豌豆凝集素注入子宫则无抗着床效应,着床率为85.7%。由此可以推测,N-连接的包含二个未被取代的或只在C-2位被取代的α-甘露糖苷的寡糖参于胚泡与子宫内膜相互作用的着床过程。  相似文献

Anthropogenic ocean acidification is likely to have negative effects on marine calcifying organisms, such as shelled pteropods, by promoting dissolution of aragonite shells. Study of shell dissolution requires an accurate and sensitive method for assessing shell damage. Shell dissolution was induced through incubations in CO2‐enriched seawater for 4 and 14 days. We describe a procedure that allows the level of dissolution to be assessed and classified into three main types: Type I with partial dissolution of the prismatic layer; Type II with exposure of underlying crossed‐lamellar layer, and Type III, where crossed‐lamellar layer shows signs of dissolution. Levels of dissolution showed a good correspondence to the incubation conditions, with the most severe damage found in specimens held for 14 days in undersaturated condition (Ω ~ 0.8). This methodology enables the response of small pelagic calcifiers to acidified conditions to be detected at an early stage, thus making pteropods a valuable bioindicator of future ocean acidification.  相似文献

The objective of this study was to quantify the incidence of underlying cervical intraepithelial neoplasia (CIN) among women referred for colposcopy with three consecutive inadequate smears. The design was a retrospective cohort study analysing data from a regional colposcopy database at Cervical Screening Wales. Women who were referred to all the colposcopy clinics in Wales with three consecutive inadequate smears, the third inadequate smear being taken between 1 April 2001 and 31 March 2002 constituted the study population. The results of the fourth smear taken at the colposcopy clinic after three consecutive inadequate smears, the worst biopsy results from any of the subsequent colposcopies and the relationship between the result of the fourth smear taken at colposcopy clinic and any histology result were the main outcome measures. The accuracy of the colposcopic opinion was also examined. Of the 433 women identified as having been referred because of three consecutive inadequate smears, 302 were linked to either a subsequent smear and/or a biopsy result. An adequate smear result was available for 85% of these women when the smear was taken in the colposcopy clinic; 77% were reported as negative and 8% were abnormal. Of the 347 women seen in the colposcopy clinic, high-grade CIN was seen in 3% and low-grade lesion in 8%. The sensitivity and specificity of the fourth inadequate smear test in predicting underlying CIN were 15% and 84% respectively, with a positive predictive value of 8%. The sensitivity and specificity of colposcopy in predicting histological CIN among patients with three inadequate smears was 70% and 49%, respectively, and the positive predictive value was 44%. This study raises the question as to whether three consecutive inadequate smears should be considered as an indication for colposcopy, or merely for a further smear to be taken in circumstances where there is a greater likelihood getting an adequate result.  相似文献

蓖麻蚕Philosamia cynthia ricni血淋巴含两种凝集素,一种凝集兔新鲜红血球,凝血活力被L-鼠李糖和D-半乳糖抑制;另一种凝集戊二醛固定的人和鸡的红血球,凝血活力被岩藻糖抑制.它们在蚕的不同生长阶段及在蚕体各组织中的分布和凝血活力显著不同.血淋巴中这两种凝集素的凝血活力明显比其他组织中高.卵中测不到这两种凝集素活力.本文对这两种凝集素在蚕体中可能的生理功能进行了讨论.  相似文献

2-Carboxy indolines were synthesised as potential antagonists at the glycine site of the NMDA receptor, based on a pharmacophore developed from other series. None of the indolines had significant potency, possibly due to the lack of coplanarity of the carboxyl with the aromatic ring. The corresponding indoles are potent antagonists, as exemplified by 19, IC50 27 nM.

Molecular modelling suggests that 2-carboxyindolines may not bind to the Glycine/NMDA receptor due to the lack of coplanarity between the aromatic ring and the carboxylate.  相似文献

The use of in situ air sparging (IAS) has increased rapidly since the early 1990s, and it is now likely to be the most practiced engineered in situ remediation option when targeting the treatment of hydrocarbon-impacted aquifers. To date, IAS system design has remained largely empirical, with significant variability in approaches and results. Here, the valuable knowledge gained from IAS studies and applications over the past decade has been integrated into a new paradigm for feasibility assessment, pilot testing, design, and operation. The basis for this Design Paradigm, the initial feasibility assessment, monitoring, and the overall design approach are discussed in detail here; other referenced documents contain the details of specific recommended activities. The proposed design approach is unique in that it contains two design routes; the first is a non-site-specific approach requiring minimal site characterization and testing (Standard Design Approach), while the second is a more site-specific approach (Site-Specific Design Approach).  相似文献

In situ air sparging (IAS) is a technology commonly used for treatment of submerged source zones and dissolved groundwater plumes. The acceptance of IAS by regulatory agencies, environmental consultants, and industry is remarkable considering the degree of skepticism initially surrounding the technology in the early 1990s. Much has been learned and reported in the literature since that time, but it appears that practice has changed little. In particular, conventional pilot testing, design, and operation practices reflect a lack of appreciation of the complex phenomena governing IAS performance and the unforgiving nature of this technology. Many systems are poorly monitored and likely to be inefficient or ineffective. Key lessons-learned since the early 1990s are reviewed and their implications for practice are discussed here. Of particular importance are issues related to: (a) the understanding of air flow distributions and the effects of geology and injection flowrate, (b) the need to characterize air flow distributions at the pilot- and field-scale, (c) how changes in operating conditions (e.g., pulsing) can affect performance improvements and reduce equipment costs, and (d) how conventional monitoring approaches are incapable of assessing if systems are performing as designed.  相似文献
