In pemphigoid gestationis (PG), autoantibodies target collagen XVII, a hemidesmosomal transmembrane protein, which is an important element in cutaneous epithelial adhesion and signalling. We report that collagen XVII is expressed in the first trimester and term syncytial and cytotrophoblastic cells of normal placenta and in epithelial cells of amniotic membrane. Immunoelectron microscopy confirmed the localization of collagen XVII to the hemidesmosomes of amniotic epithelium. Examination of three PG placentas showed mild villitis, but there were no differences between collagen XVII expression levels or immunostaining signals as compared to normal placenta. Collagen XVII expression was also detected in cultured extravillous trophoblast HTR-8/SVneo cells, where collagen XVII expression was upregulated by PMA and TGF-beta1. Interestingly, the presence of Col15, the cell migration domain of collagen XVII, induced the migration of HTR-8/SVneo cells in transmigration assay. Analysis of amniotic fluid samples at different gestational weeks revealed that a large quantity of collagen XVII ectodomain was shed into amniotic fluid throughout pregnancy. Biochemical and immunoblotting analysis indicated that the ectodomain in amniotic fluid is structurally very similar to the ectodomain produced by cultured keratinocytes. Cultured cells from amniotic fluid samples also expressed collagen XVII. Our results suggest that collagen XVII may contribute to the invasion of extravillous trophoblasts during placental development and is also required for the integrity of amniotic basement membrane. Although the exact pathomechanism of PG is still largely unknown, the clinical symptoms of PG are initiated after the expression of collagen XVII in placenta during the first trimester of pregnancy.  相似文献

Numerous species of microscopic fungi inhabit mosses and hepatics. They are severely overlooked and their identity and nutritional strategies are mostly unknown. Most of these bryosymbiotic fungi belong to the Ascomycota. Their fruit‐bodies are extremely small, often reduced and simply structured, which is why they cannot be reliably identified and classified by their morphological and anatomical characters. A phylogenetic hypothesis of bryosymbiotic ascomycetes is presented. New sequences of 78 samples, including 61 bryosymbionts, were produced, the total amount of terminals being 206. Of these, 202 are Ascomycetes. Sequences from the following five gene loci were used: rDNA SSU, rDNA LSU, RPB2, mitochondrial rDNA SSU, and rDNA 5.8S. The program TNT was used for tree search and support value estimation. We show that bryosymbiotic fungi occur in numerous lineages, one of which represents a newly discovered lineage among the Ascomycota and exhibits a tripartite association with cyanobacteria and sphagna. A new genus Trizodia is proposed for this basal clade. Our results demonstrate that even highly specialized life strategies can be adopted multiple times during evolution, and that in many cases bryosymbionts appear to have evolved from saprobic ancestors. © The Willi Hennig Society 2009.  相似文献

Circadian rhythms play a very important role on metabolic process and have considerable effects on growth, especially in ectotherms. Like variation in hormone levels, the sensitivity of target cells may show diurnal or seasonal fluctuations. The aim of this study was to compare the effects of morning versus evening injections of growth hormone and prolactin on malic enzyme, glucose-6-phosphate dehydrogenase, isocitrate dehydrogenase, aspartate aminotransferase, alanine aminotransferase and Na+,K+-ATPase in a teleost Anabas testudineus. Activities of malic enzyme, glucose-6-phosphate dehydrogenase and isocitrate dehydrogenase of the two control groups themselves differ significantly at morning and evening. Early morning administration of growth hormone increases malic enzyme, glucose-6-phosphate dehydrogenase and isocitrate dehydrogenase activities while evening administration of growth hormone does not effect these enzymes. Transaminase activities were stimulated by morning and evening administration of GH and PRL. Na+,K+-ATPase activity was stimulated by morning administration and inhibited by evening treatment of both hormones. The results reveal that a given hormone may provide a different message to the target tissues at different periods of the day.  相似文献

We monitored the behaviour of farmed silver foxes housed in family units in enlarged cage systems from weaning until late October. Activity of family members, use of space available, and aggressive acts were recorded. Aggressiveness between family members increased from July until October, leading to a more scattered use of the available space. Furthermore, the mean activity level of family members increased, and the synchrony of activity decreased. We conclude that social tension in the fox families increased gradually during the autumn, leading to dispersion of the family members.
