Fibre bundles of glycerinated rabbit psoas muscle about 0.5-1.0 mm thick were incubated in 5 mM tris-(hydroxymethyl)-aminomethane (Tris), pH 8.0 for 5-20 hours at 4 degrees C. This treatment leads to selective removal of some proteins in the M-bands and H-zones of sarcomeres. Effects of extraction were analyzed on the basis of electron micrographs of longitudinal sections of muscle specimens, gel electrophoresis patterns of myofibrils and of the extracts, and measurements of the creatine kinase activity of myofibrils. In the X-ray diffraction patterns of the fibre bundles subjected to prolonged extraction a drastic decrease in the intensity of "442 A" and "223 A" meridional reflections and a considerably smaller decrease in the intensity of "212 A" meridional reflections were observed. The "147 A" meridional reflection remains practically unchanged. It was concluded that: (1) The reflections "442 A" and "223 A" were contributed mainly by diffraction on the minor proteins located in the central part of the thick filaments in between the C-zones. This is contrary to the widely accepted viewpoint according to which the appearance of "442 A" reflection is caused only by the C-protein component of the thick filaments. (2) The "147 A" meridional reflection is contributed mainly by C-protein and light meromyosin of the thick filaments. 相似文献

X-ray patterns from frog skeletal muscles at rest show a series of relatively weak meridional reflections which may be indexed as the 5, 7, 9, 11 and 13 orders of the repeat period of about 2 x 385 A. According to the model of the thin filaments structure, suggested by V. V. Lednev and G. M. Frank (1977), this period is specific for the activated or "switched on" state of the actin--containing filaments. At the same time, according to the generally accepted model (suggested in 1972), the axial repeat period of the thin filament structure is approximately equal to 385 A and does not depend on the functional state of the muscle. The existence of the repeat period of about 2 x 385 A in the thin filaments of a resting muscle suggests that even at rest the thin filaments of vertebrate skeletal muscle are not completely inhibited. It may be suggested that partial activation of the thin filaments in a resting muscle is the result of formation of long life rigorlike crossbridges, the existence of which was postulated by D. K. Hill in 1968 on the basis of his studies on resting tension in the frog skeletal muscle. 相似文献

Amyloid fibrils are β-sheet-rich protein aggregates that are strongly associated with a variety of neurodegenerative maladies, such as Alzheimer’s and Parkinson’s diseases. Even if the secondary structure of such fibrils is well characterized, a thorough understanding of their surface organization still remains elusive. Tip-enhanced Raman spectroscopy (TERS) is one of a few techniques that allow the direct characterization of the amino acid composition and the protein secondary structure of the amyloid fibril surface. Herein, we investigated the surfaces of two insulin fibril polymorphs with flat (flat) and left-twisted (twisted) morphology. It was found that the two differ substantially in both amino acid composition and protein secondary structure. For example, the amounts of Tyr, Pro, and His differ, as does the number of carboxyl groups on the respective surfaces, whereas the amounts of Phe and of positively charged amino and imino groups remain similar. In addition, the surface of protofilaments, the precursors of the mature flat and twisted fibrils, was investigated using TERS. The results show substantial differences with respect to the mature fibrils. A correlation of amino acid frequencies and protein secondary structures on the surface of protofilaments and on flat and twisted fibrils allowed us to propose a hypothetical mechanism for the propagation to specific fibril polymorphs. This knowledge can shed a light on the toxicity of amyloids and define the key factors responsible for fibril polymorphism. Raman spectroscopy was employed as a nondestructive technique for the analysis of dry traces of oral fluid to differentiate between smoker and nonsmoker donors with the aid of advanced statistical tools. A genetic algorithm (GA) was used to determine eight spectral regions that contribute the most to the differentiation of smokers and nonsmokers.
