A broad spectrum of events that come under the category of green tide are recognized world-wide as a response to elevated
levels of seawater nutrients in coastal areas. Green tides involve a wide diversity of sites, macroalgal species, consequences,
and possible causes. Here we review the effect of natural and man-induced environmental fluctuations on the frequency and
apparent spread of green tides. This article highlights the need for interdisciplinary research aimed at shedding light on
the basic mechanisms governing the occurrence and succession of green algae in coastal seas. This will result in more effective
management and mitigation of the effects of green tides, thus safeguarding the intrinsic and commercial value of coastal marine

Sunitinib, a small-molecule multi-targeted tyrosine kinase inhibitor, has been applied in phase II clinical trial as second-line treatment for advanced gastric cancer. In this study, we determined the effect of Sunitinib on the multidrug resistance in gastric cancer cells selected by vincristine. Our results showed that Sunitinib significantly enhanced the cytotoxicity of adriamycin, vincristine, etoposide, 5-Fluorouracil, and cisplatin in multidrug-resistant gastric cancer cells (SGC7901/VCR). Sunitinib significantly increased the intracellular accumulation and retention of rhodamine 123 in the SGC7901/VCR cells. However, Sunitinib, at a concentration that reverses MDR, had no significant effect on P-gp protein or mRNA expression levels. In addition, the present study revealed that Sunitinib inhibited Stat3 and down-regulated Bcl-2 in SGC7901/VCR cells, which might also contribute to the reversal of MDR. In conclusion, Sunitinib reverses multidrug resistance in gastric cancer cells by inhibiting P-gp transporter function and modulating Stat3 and Bcl-2. Further study with Sunitinib may be helpful for developing combination therapeutic strategy or circumventing gastric cancer MDR to other conventional anti-cancer drugs. 相似文献

Engineering of Plasmin-Resistant Forms of Streptokinase and Their Production in Bacillus subtilis: Streptokinase with Longer Functional Half-Life 下载免费PDF全文
The short in vivo half-life of streptokinase limits its efficacy as an efficient blood clot-dissolving agent. During the clot-dissolving process, streptokinase is processed to smaller intermediates by plasmin. Two of the major processing sites are Lys59 and Lys386. We engineered two versions of streptokinase with either one of the lysine residues changed to glutamine and a third version with both mutations. These mutant streptokinase proteins (muteins) were produced by secretion with the protease-deficient Bacillus subtilis WB600 as the host. The purified muteins retained comparable kinetics parameters in plasminogen activation and showed different degrees of resistance to plasmin depending on the nature of the mutation. Muteins with double mutations had half-lives that were extended 21-fold when assayed in a 1:1 molar ratio with plasminogen in vitro and showed better plasminogen activation activity with time in the radial caseinolysis assay. This study indicates that plasmin-mediated processing leads to the inactivation of streptokinase and is not required to convert streptokinase to its active form. Plasmin-resistant forms of streptokinase can be engineered without affecting their activity, and blockage of the N-terminal cleavage site is essential to generate engineered streptokinase with a longer in vitro functional half-life. 相似文献

研究了黄瓜品种津研4号(感枯萎病)、津春4号(抗枯萎病)和云南黑籽南瓜根系分泌物对津研4号黄瓜枯萎病发生的影响及其原因.结果表明:感病品种根系分泌物处理的黄瓜枯萎病发病早,接种后第15天病株率显著高于对照,至第20天时病株率与对照相近;而抗病品种根系分泌物处理的病株率一直显著小于对照.感病品种根系分泌物浇灌的植株株高、鲜质量降低,根系活力下降、电导度(伤害度)增加,而抗病品种和云南黑籽南瓜根系分泌物处理对植株影响较小.感病品种根系分泌物促进了黄瓜枯萎病菌的生长,而抗病品种和云南黑籽南瓜根系分泌物则抑制了病菌生长. 相似文献

We previously showed that Wnt3a could stimulate human embryonic stem (hES) cell proliferation and affect cell fate determination. In the absence of feeder cell--derived factors, hES cells cultured under a feeder-free condition survived and proliferated poorly. Adding recombinant Wnt3a in the absence of feeder cell derived-factors stimulated hES cell proliferation but also differentiation. In the present study, we further extended our analysis to other Wnt ligands such as Wntl and Wnt5a. While Wntl displayed a similar effect on hES cells as Wnt3a, Wnt5a had little effect in this system. Wnt3a and Wntl enhanced proliferation of undifferentiated hES cells when feeder-derived self-renewal factors and bFGF are also present. To explore the possibility to promote the proliferation of undifferentiated hES cells by activating the Wnt signaling, we overexpressed Wnt3a or Wntl gene in immortalized human adult fibroblast (HAFi) cells that are superior in supporting long-term growth of undifferentiated hES cells than primary mouse embryonic fibroblasts. HAFi cells with or without a Wnt tmnsgene can be propagated indefinitely. Over-expression of the Wnt3a gene significantly enhanced the ability of HAFi feeder cells to support the undifferentiated growth of 3 different hES cell lines we tested. Co-expression of three commonly-used drug selection genes in Wnt3a-overpressing HAFi cells further enabled us to select rare hES clones after stable transfection or transduction. These immortalized engineered feeder cells (W3R) that co-express growth-promoting genes such as Wnt3a and three drug selection genes should empower us to efficiently make genetic modified hES cell lines for basic and translational research. 相似文献

A prenylated flavonoid, moralbanone, along with seven known compounds kuwanon S, mulberroside C, cyclomorusin, eudraflavone B hydroperoxide, oxydihydromorusin, leachianone G and alpha-acetyl-amyrin were isolated from the root bark of Morus alba L. Leachianone G showed potent antiviral activity (IC(50) = 1.6 microg/ml), whereas mulberroside C showed weak activity (IC(50) = 75.4 microg/ml) against herpes simplex type 1 virus (HSV-1). 目的探讨氟康唑3日间隔疗法治疗念珠菌性外阴、阴道炎的临床疗效。方法110例患者随机分为两组.治疗组60例采用氟康唑3日间隔疗法(第1日服药150mg后.第4、7日各再服1次),对照组50例1次单剂量口服氟康唑150mg。结果治疗组和对照组有效率分别为86.67%、72.00%.差异有显著性(χ^2=9.46,P〈0.05).结论氟康唑3日间隔疗法治疗念珠菌性外阴、阴道炎疗效好。毒副作用小,安全性高。 相似文献

多角体计数、对流免疫电泳、单向免疫扩散及火箭免疫电泳测定的结果表明:26℃适于茶尺蠖核型多角体病毒(EoNPV)的增殖。多角体含量或其相对值随着时间的推移而增长,并渐趋于平稳,两者间呈Logistic曲线关系。单位体重或单头幼虫所含的多角体数量(y_1或y_2)、扩散环直径(y_3)和火箭峰值(y_4)与时间(t)的关系式分别为:y_1=(8.1481)/(1 EXP(9.4210-0.0608t))×10~9PIB/克;y_2=(6.1596)/(1 EXP(5.4809-0.0376t))×10~8PIB/头,y_3=(1.4)/(1 EXP(2.710-0.015t))cm;y_4=(3.52)/(1 EXP(4.580-0.040t))cm。但30℃下,EoNPV增殖严重受抑制,饲毒后24~168小时内难以测出多角体,其后多角体含量也极显著低于26℃,乃至难以被三种免疫测定法测出。 相似文献
