Abstract Cecropins and other natural antimicrobial peptides are widely distributed in animals from insects to mammals. These proteins have been shown to be major constituents of the innate immune systems of animals for nonspecific defense of the host against various bacteria and parasites. Therefore, exploitation of this natural innate defense system may lead to the development of effective methods for protecting fish from invasion by microbial pathogens. Recently, we have demonstrated that the introduction of cecropin transgenes into Japanese medaka (Oryzias latipes) conferred resistance to infection by fish bacterial pathogens. Aside from a few reports documenting the antiviral effect of antimicrobial peptides including cecropins against mammalian viruses, there is no evidence for the effect of these peptides against fish viruses. In this article we present results of in vitro characterization of native cecropin B and a synthetic analogue, CF17, against several important fish viral pathogens—namely, infectious hematopoietic necrosis virus (IHNV), viral hemorrhagic septicemia virus (VHSV), snakehead rhabdovirus (SHRV), and infectious pancreatic necrosis virus (IPNV). Upon coincubation of these peptides and viruses, the viral titers yielded in fish cells were reduced from several fold to 104-fold. Direct disruption of the viral envelope and disintegration of the viral capsids may be involved in the inhibition of viral replication by the peptides. Results of our studies demonstrate the potential of manipulating antimicrobial peptide genes by transgenesis to combat viral infection in fish.  相似文献

Several factors decrease plant survival throughout their lifecycles. Among them, seed dispersal limitation may play a major role by resulting in highly aggregated (contagious) seed and seedling distributions entailing increased mortality. The arrival of seeds, furthermore, may not match suitable environments for seed survival and, consequently, for seedling establishment. In this study, we investigated spatio-temporal patterns of seed and seedling distribution in contrasting microhabitats (bamboo and non-bamboo stands) from the Brazilian Atlantic Forest. Spatial distribution patterns, spatial concordance between seed rain and seedling recruitment between subsequent years in two fruiting seasons (2004–2005 and 2007–2009), and the relation between seeds and seedlings with environmental factors were examined within a spatially-explicit framework. Density and species richness of both seeds and seedlings were randomly distributed in non-bamboo stands, but showed significant clustering in bamboo stands. Seed and seedling distributions showed across-year inconsistency, suggesting a marked spatial decoupling of the seed and seedling stages. Generalized linear mixed effects models indicated that only seed density and seed species richness differed between stand types while accounting for variation in soil characteristics. Our analyses provide evidence of marked recruitment limitation as a result of the interplay between biotic and abiotic factors. Because bamboo stands promote heterogeneity in the forest, they are important components of the landscape. However, at high densities, bamboos may limit recruitment for the plant community by imposing marked discordances of seed arrival and early seedling recruitment.  相似文献

Presently there are no good assays for comparing somatic mutation frequencies and spectra between different vertebrate and invertebrate organisms. Here we describe a new lacZ mutation reporter system in D. melanogaster, which complements existing systems in the mouse. The results obtained with the new model indicate two-to threefold higher frequencies of spontaneous mutations than in the mouse, with most of the mutations characterized as large genome rearrangements.  相似文献

Abstract. Virgin mantis females, Hierodula patellifera (Serville) (Dictyoptera: Mantidae), exhibit a characteristic calling posture. When holding the body below a branch or leaf, the female curls the abdomen ventrally, flexing it away from the wings and exposing its dorsal surface. The curling is accompanied by pumping movements. The average age at which females start adopting this calling posture is 14 days after adult moult, and it is related to their nutritional stage. Once initiated, females exhibit the posture everyday until they mate. After mating, the behaviour is completely suppressed. Males are attracted by virgin females adopting the calling posture but are not attracted to mated females. The characteristics of the posture and the responsive behaviour of the males indicate that this female calling involves the release of sex pheromones.  相似文献

Habitat selection, which is mainly a consequence of competition and predation, allows species to coexist. The present study was conducted in two reserves in an Atlantic Forest area in Santa Catarina State, southern Brazil, and provided information on several large mammal species through photographic records. Records were related to certain environmental parameters, such as width of passages (trails and roads), vegetation density and proximity to water, in order to assess the relationship between each mammal species and its microhabitat. Thirty-two camera trap stations were placed during 17 months for 150.8 (±62.2) days on average. Terrestrial mammals tended to use different habitats: Puma concolor used mainly dirt roads and open areas; Leopardus pardalis, Cerdocyon thous and Nasua nasua used more large trails and intermediate-forested sites; and Cuniculus paca, Dasypus novemcinctus, Leopardus tigrinus, Eira barbara and Leopardus wiedii were recorded more often on narrow trails and in densely forested sites. Some of these forest species, such as D. novemcinctus, C. paca and L. pardalis, also showed relationships with watercourses. Information on habitat selection allows more effort to be addressed to the habitat associated with focal species, and indicates the significance of environmental heterogeneity, which makes it possible for species to coexist.  相似文献

Oxidative stress due to excessive reactive oxygen species (ROS) and depleted antioxidants such as glutathione (GSH) can give rise to apoptotic cell death in acutely diabetic hearts and lead to heart disease. At present, the source of these cardiac ROS or the subcellular site of cardiac GSH loss [i.e., cytosolic (cGSH) or mitochondrial (mGSH) GSH] has not been completely elucidated. With the use of rotenone (an inhibitor of the electron transport chain) to decrease the excessive ROS in acute streptozotocin (STZ)-induced diabetic rat heart, the mitochondrial origin of ROS was established. Furthermore, mitochondrial damage, as evidenced by loss of membrane potential, increases in oxidative stress, and reduction in mGSH was associated with increased apoptosis via increases in caspase-9 and -3 activities in acutely diabetic hearts. To validate the role of mGSH in regulating cardiac apoptosis, L-buthionine-sulfoximine (BSO; 10 mmol/kg ip), which blocks GSH synthesis, or diethyl maleate (DEM; 4 mmol/kg ip), which inactivates preformed GSH, was administered in diabetic rats for 4 days after STZ administration. Although both BSO and DEM lowered cGSH, they were ineffective in reducing mGSH or augmenting cardiomyocyte apoptosis. To circumvent the lack of mGSH depletion, BSO and DEM were coadministered in diabetic rats. In this setting, mGSH was undetectable and cardiac apoptosis was further aggravated compared with the untreated diabetic group. In a separate group, GSH supplementation induced a robust amplification of mGSH in diabetic rat hearts and prevented apoptosis. Our data suggest for the first time that mGSH is crucial for modulating the cell suicide program in short-term diabetic rat hearts.  相似文献

The way we currently understand vertebrate development is undoubtedly associated with the research undertaken at the "Institut d'Embryologie Cellulaire et Moleculaire" at Nogent-sur-Marne during the last decades. Working in this Institute has been a privilege for many junior and senior researchers. Eight years ago, in this stimulating environment, an exciting observation followed by a series of revealing experiments gave rise to a novel field of research. This study provided evidence for the existence of a molecular clock underlying chick somite formation. In this review, we focus on the cascade of studies that have followed this discovery. Thus far, it has been demonstrated that the molecular clock is operating in several vertebrate models namely chick, mouse, zebrafish, frog and medaka, probably functioning to provide cells with multidimensional positional information. Loss and gain of function experiments and detailed gene promoter analyses have proved very useful in understanding how the clock machinery works. Recent data has also led to the fascinating hypothesis that the clock might not be an exclusive property of somitic cells, but rather a mechanism used by a wide range of embryonic tissues. Meanwhile, the clock "keeps ticking" and many questions are still waiting for an answer.  相似文献

The identification of mycobacterial species in clinical isolates is essential for making patient care decisions. Polymerase chain reaction (PCR) restriction enzyme analysis (PRA) is a simple and rapid identification method, based on amplification of 441 bp of the hsp65 gene and restriction with BstEII and HaeIII. As a contribution to the validation of PRA, a multicenter study was performed in eight laboratories located in Argentina, Brazil, Colombia, Chile, and Guadeloupe. Each laboratory received 18 coded isolates from the collection of the Institute of Tropical Medicine (Antwerp, Belgium), representing duplicates of nine laboratory strains: Mycobacterium terrae CIPT 140320001, Mycobacterium scrofulaceum CIPT 140220031, Mycobacterium flavescens ATCC 14474, Mycobacterium triviale ATCC 23292, Mycobacterium nonchromogenicum ATCC 19530, Mycobacterium chitae ATCC 19627, Mycobacterium abscessus ATCC 19977, Mycobacterium kansasii ATCC 12478, and Mycobacterium peregrinum ATCC 14467. A detailed protocol including amplification, enzymatic digestion, and gel preparation was provided to each laboratory. Two laboratories identified correctly all 18 (100%) isolates, one identified correctly 17 (94.5%), two identified 14 (77.7%), one identified 11 (61%), and two identified 8 (44.4%) isolates. Errors detected in laboratories with more than 77% accuracy were associated with electrophoresis running conditions and an unspecific amplicon produced by a single strain. Lower accuracy was mainly related to inappropriate use of DNA markers and insufficient training in interpretation of patterns. In conclusion, the PRA method was readily implemented in some Latin American and Caribbean laboratories of mycobacteria, but improvements in critical points, as gel running conditions and training in interpretiation of patterns, are needed in order to improve accuracy. In others, improvement in critical points is still necessary.  相似文献
