Background and Aims: Leaves expand during a given period of time until they reachtheir final size and form, which is called determinate growth.Duration of leaf expansion is stable when expressed in thermal-timeand in the absence of stress, and consequently it is often proposedthat it is controlled by a robust programme at the plant scale.The usual hypothesis is that growth cessation occurs when cellexpansion becomes limited by an irreversible tightening of cellwall, and that leaf size is fixed once cell expansion ceases.The objective of this paper was to test whether leaf expansioncould be restored by rewatering plants after a long soil water-deficitperiod. Methods: Four experiments were performed on two different species (Arabidopsisthaliana and Helianthus annuus) in which the area of leavesthat had apparently reached their final size was measured uponreversal of water stresses of different intensities and durations. Key Results: Re-growth of leaves that had apparently reached their finalsize occurred in both species, and its magnitude depended onlyon the time elapsed from growth cessation to rewatering. Leafarea increased up to 186% in A. thaliana and up to 88% in H.annuus after rewatering, with respect to the leaves of plantsthat remained under water deficit. Re-growth was accounted forby cell expansion. Increase in leaf area represented actualgrowth and not only a reversible change due to increased turgor. Conclusions: After the leaf has ceased to grow, leaf cells retain their abilityto expand for several days before leaf size becomes fixed. Aresponse window was identified in both species, during whichthe extent of leaf area recovery decreased with time after the‘initial’ leaf growth cessation. These results suggestthat re-growth after rewatering of leaves having apparentlyattained their final size could be a generalized phenomenon,at least in dicotyledonous plants.  相似文献

Recent findings demonstrate that alkaline/neutral invertases (A/N-Invs), enzymes that catalyze the breakdown of sucrose into glucose and fructose, are essential proteins in plant life. The fact that different isoforms are present in multiple locations makes them candidates for the coordination of metabolic processes. In the present study, we functionally characterized the encoding gene of a novel A/N-Inv (named A/N-InvC) from Arabidopsis, which localizes in mitochondria. A/N-InvC is expressed in roots, in aerial parts (shoots and leaves) and flowers. A detailed phenotypic analysis of knockout mutant plants (invc) reveals an impaired growth phenotype. Shoot growth was severely reduced, but root development was not affected as reported for A/N-InvA mutant (inva) plants. Remarkably, germination and flowering, two energy demanding processes, were the most affected stages. The effect of exogenous growth regulators led us to suggest that A/N-InvC may be modulating hormone balance in relation to the radicle emergence. We also show that oxygen consumption is reduced in inva and invc in comparison with wild-type plants, indicating that both organelle isoenzymes may play a fundamental role in mitochondrion functionality. Taken together, our results emphasize the involvement of mitochondrial A/N-Invs in developmental processes and uncover the possibility of playing different roles for the two isoforms located in the organelle.  相似文献

Data from Bering-Chukchi-Beaufort Seas bowhead whales (Balaena mysticetus), harvested during 1973–2021 by aboriginal subsistence hunters, were used to estimate reproductive parameters: length at sexual maturity (LSM), age at sexual maturity (ASM), pregnancy rate (PR), and calving interval. Sexual maturity (N = 187 females) was determined from the presence/absence of corpora in the ovaries, or a fetus. Using sampling bias-corrected logistic regression, LSM was estimated at 13.5 m, 95% CI [13.0, 13.8]. There was a downward trend in LSM over time, statistically significant with one method but marginal with another. A growth model translated this estimate to an ASM estimate of 23.5 years, 95% CI [20.4, 26.7]. Pregnancy rate was determined from mature females (N = 125), and from a subset limited to certain autumn-caught whales (n = 37) to reduce bias. The PR was estimated at 0.46 globally, 95% CI [0.36, 0.55] and 0.38 for the autumn sample, 95% CI [0.20, 0.51]. Both estimated PRs are consistent with a 3-year calving interval, because the larger estimate includes two cohorts of pregnant whales harvested in spring, and bowhead whale gestation is longer than 12 months. These analyses represent the most conclusive empirical estimates of ASM, LSM, and PR for this bowhead whale stock from the largest available data sets to date.  相似文献

Here, we investigated which stress responses were influenced by the MpkC and SakA mitogen‐activated protein kinases of the high‐osmolarity glycerol (HOG) pathway in the fungal pathogen Aspergillus fumigatus. The ΔsakA and the double ΔmpkC ΔsakA mutants were more sensitive to osmotic and oxidative stresses, and to cell wall damaging agents. Both MpkC::GFP and SakA::GFP translocated to the nucleus upon osmotic stress and cell wall damage, with SakA::GFP showing a quicker response. The phosphorylation state of MpkA was determined post exposure to high concentrations of congo red and Sorbitol. In the wild‐type strain, MpkA phosphorylation levels progressively increased in both treatments. In contrast, the ΔsakA mutant had reduced MpkA phosphorylation, and surprisingly, the double ΔmpkC ΔsakA had no detectable MpkA phosphorylation. A. fumigatus ΔsakA and ΔmpkC were virulent in mouse survival experiments, but they had a 40% reduction in fungal burden. In contrast, the ΔmpkC ΔsakA double mutant showed highly attenuated virulence, with approximately 50% mice surviving and a 75% reduction in fungal burden. We propose that both cell wall integrity (CWI) and HOG pathways collaborate, and that MpkC could act by modulating SakA activity upon exposure to several types of stresses and during CW biosynthesis.  相似文献

Interleukin-22 (IL-22) is a member of the interleukin-10 cytokine family, which is involved in anti-microbial defenses, tissue damage protection and repair, and acute phase responses. Its signaling mechanism involves the sequential binding of IL-22 to interleukin-22 receptor 1 (IL-22R1), and of this dimer to interleukin-10 receptor 2 (IL-10R2) extracellular domain. We report a 1.9A crystal structure of the IL-22/IL-22R1 complex, revealing crucial interacting residues at the IL-22/IL-22R1 interface. Functional importance of key residues was confirmed by site-directed mutagenesis and functional studies. Based on the X-ray structure of the binary complex, we discuss a molecular basis of the IL-22/IL-22R1 recognition by IL-10R2. STRUCTURED SUMMARY:  相似文献

Cytogenetic studies involving the family Prochilodontidae have shown that these fish can be characterized by a constant diploid number and a conserved karyotypic macrostructure. This study focused on comparative physical chromosomal mapping using 18S and 5S rDNA to compare the species Semaprochilodus insignis and S. taeniurus. Our results indicated the conservation of large number of conventional chromosomal markers. The molecular cytogenetic analyses of the location of the 18S rDNA indicated the maintenance of a chromosome pair bearing these sites in both species analyzed, and it appears to be a conserved character among the majority of the species of this family. The stability of the number of 5S ribosomal DNA sites and their chromosomal localization as has been reported for the Prochilodontidae was not, however, confirmed for S. insignis and S. taeniurus, as these species showed multiple specific rDNA 5S sites. As such, and in spite of the fact that a number of studies indicate that the family Prochilodontidae has a conserved karyotypic structure, the utilization of molecular tools that use chromosomal segments as markers revealed that this presumed stability cannot be extended to the genome level for the species S. insignis and S. taeniurus.  相似文献

Myotoxin II, a Lys49 catalytically inactive phospholipase A(2) homologue from Atropoides nummifer venom, was purified, characterized and crystallized. The crystals belongs to the tetragonal system, space group P4(3)2(1)2, with unit cell parameters (a=b=68.66 and c=63.87 angstroms). Diffraction data were collected to a resolution of 2.32 angstroms. Biodiversity is declining globally, primarily due to anthropogenic threats. Therefore, effective conservation efforts must integrate human and environmental components. Social-ecological systems research is increasingly being adopted as a means of studying complex relationships between people and the environment. I assess how researchers are employing social-ecological systems approaches or frameworks to the study of tropical ecosystems. I reviewed articles published in Biotropica from 2010 through 2022 searching for research on social-ecological systems. A broad keyword search revealed only 2 articles using a variation of social-ecological systems, human-environment systems, or coupled human and natural systems. This contrasts with a growing number of articles published with these search terms in other conservation-related journals, primarily led by environmental scientists. After reviewing titles for all 1298 research articles published during this period, I selected 12 articles for inclusion in the virtual special issue “Social-Ecological Systems Research in Topical Ecosystems”. These articles cover a broad range of geographical locations, ecosystem types, species, and conservation themes. Social-ecological systems frameworks offer an integrated way to study complex relationships between humans and nature, yet this type of research appears under-utilized by authors in Biotropica. I offer seven guidelines for authors interested in pursuing this research such as developing collaborations between social and environmental scientists.  相似文献

Hereditary hemochromatosis (HH) is the most common genetic disease among individuals of European descent. Two mutations (845G-->A, C282Y and 187C-->G, H63D) in the hemochromatosis gene (HFE gene) are associated with HH. About 85-90% of patients of northern European descent with HH are C282Y homozygous. The prevalence of HH in the Brazilian population, which has a very high level of racial admixture, is unknown. The aims of the present study were to identify individuals with diagnostic criteria for HH among patients with a body iron overload attended at the university hospital of the Faculty of Medicine of Ribeirao Preto from 1990 to 2000, and to evaluate the prevalence of HFE mutations. We screened first-degree relatives for HFE mutations. Four of 72 patients (three men and one woman, mean age 47 years) fulfilled the criteria for HH. HFE mutations were studied in three patients [two C282Y homozygotes (patients 1 and 2) and one H63D heterozygote]. Patient 1 had four children (all C282Y heterozygotes with no iron overload) and seven brothers and sisters: two sisters (66 and 76 years old) were C282Y homozygotes and both had an iron overload (a liver biopsy in one showed severe iron deposits), one sister (79 years old) was a compound heterozygote with no iron overload, one brother (78 years old) was a C282Y heterozygote with no iron overload, two individuals were H63D heterozygotes (one brother, 49 years old, obese, with a body iron overload and abnormal liver enzymes - a biopsy showed non-alcoholic steatohepatitis, and one 70-year-old sister with no iron overload). Patient 2 had two children (22 and 24 years old who were C282Y heterozygotes with no iron overload) but no brothers or sisters. These results showed that HH was uncommon among individuals attended at our hospital, although HFE mutations were found in all patients. Familial screening is valuable for the early diagnosis of individuals at risk since it allows treatment to be initiated before the onset of the clinical manifestations of organ damage associated with HH.  相似文献
