Many Gram-negative pathogens translocate virulence proteins directly into host cells using a type III secretion system. This complex secretion machinery is composed of approximately 25 different proteins that assemble to span both bacterial membranes, and contact the host cell to form a direct channel between the bacterial and host cell cytoplasms. Assembly of the system and efficient secretion of virulence proteins through this apparatus require specific chaperones. Although the machinery is morphologically conserved among all bacteria, the secreted proteins vary widely and are responsible for the range of diseases caused by bacterial pathogens. Recent structures have given insights into important chaperone and effector proteins, as well as revealing the first atomic structures of portions of the secretion machinery itself.  相似文献

The assumption that ozone is not only a strong oxidant, but also an important inflammatory mediator, is heavily supported by the ample literature on the pulmonary toxicity and biological effects of environmental ozone and by the recent discovery that antibodies, human neutrophils, and inflammatory lesions catalyze the formation of ozone in vivo. We hypothesized that the pulmonary inflammation in asthma involves a vicious circle of ozone production and recruitment of white blood cells, which produce more ozone. Accordingly, we predicted that electron-rich olefins, which are known ozone scavengers, could be used for prophylactic treatment of asthma. In particular, volatile, unsaturated monoterpenes, could saturate the pulmonary membranes and thereby equip the airways with local chemical protection against either exogenous or endogenous ozone. Here we present experimental evidence using a sensitized rat model to support this hypothesis. Examination of the pulmonary function of sensitized rats that inhaled either limonene (unsaturated, ozone scavenger) or eucalyptol (saturated, inert to ozone) showed that limonene inhalation significantly prevents bronchial obstruction while eucalyptol inhalation does not cause any effect. The anti-inflammatory effect of limonene was also evident from pathological parameters, such as diminished peribronchiolar and perivascular inflammatory infiltrates.  相似文献

Over the past 40 years, much research has been carried out directed toward the characterization of the cannabinergic system. With the identification of two G-protein coupled receptors and the endogenous ligand, anandamide, pharmacological targets have expanded to encompass hydrolase and transport proteins as well as novel classes of cannabinoid ligands. Those ligands that demonstrate high affinity for the receptors and good biological efficacy are tied together through lipophilic regions repeatedly demonstrated necessary for activity. This review presents recent developments in the structure-activity relationships of several classes of cannabinoid ligands.  相似文献

The complement regulatory protein decay accelerating factor (DAF; CD55), inhibits the alternative complement pathway by accelerating decay of the convertase enzymes formed by C3b and factor B. We show, using surface plasmon resonance, that in the absence of Mg(2+), DAF binds C3b, factor B, and the Bb subunit with low affinity (K(D), 14 +/- 0.1, 44 +/- 10, and 20 +/- 7 microm, respectively). In the presence of Mg(2+), DAF bound Bb or the von Willebrand factor type A subunit of Bb with higher affinities (K(D), 1.3 +/- 0.5 and 2.2 +/- 0.1 microm, respectively). Interaction with the proenzyme C3bB was investigated by flowing factor B across a C3b-coated surface in the absence of factor D. The dissociation rate was dependent on the time of incubation, suggesting that a time-dependent conformational transition stabilized the C3b-factor B interaction. Activation by factor D (forming C3bBb) increased the complex half-life; however, the enzyme became susceptible to rapid decay by DAF, unlike the proenzyme, which was unaffected. A convertase assembled with cobra venom factor and Bb was decayed by DAF, albeit far less efficiently than C3bBb. DAF did not bind cobra venom factor, implying that Bb decay is accelerated, at least in part, through DAF binding of this subunit. It is likely that DAF binds the complex with higher affinity/avidity, promoting a conformational change in either or both subunits accelerating decay. Such analysis of component and regulator interactions will inform our understanding of inhibitory mechanisms and the ways in which regulatory proteins cooperate to control the complement cascade.  相似文献

Adult mass changes, egg morphometrics, chick growth rates, fledging masses, reproductive success and reasons for reproductive failure were examined in rockhopper penguins at Macquarie Island from 1993/1994 to 1995/1996. Mean arrival masses, growth rates of chicks and fledging masses exhibited inter-annual variability, while egg morphometrics, hatching success (68.0±6.0%) and reproductive success (47.3±8.3%) were constant between years. Reproductive failures occurred primarily during incubation, with the majority of eggs lost to great skuas. Logistic regressions revealed that no variable significantly explained hatching success, and only in 1994/1995 was fledging success significantly correlated with the position of nest in the colony (those in the centre were more successful than those on the periphery). Reproductive success during this study was relatively high, and therefore an assessment during poor years would be instructive, particularly in relation to aspects of the penguins’ foraging ecology.  相似文献

The efficacy of a new mass-transfer method for estimating milk intake was examined in Antarctic fur seals (Arctocephalus gazella) at Iles Kerguelen. Our method differed from previous mass-transfer approaches in that we estimated milk-mass transfer as the maternal mass lost (MML; kg) during an attendance bout, less the mass lost to metabolic maintenance (MMLE) over that time. MML was significantly related to pup mass-gain (PMG) and attendance bout duration (d days) as follows: MML=1.106PMG+1.002d (r2=0.998). Based on this and previous studies, we estimated that the MMLE was 0.0285 kg kg–1 day–1 for lactating females; and we developed the following milk-mass transfer equation: MMLM=1.106PMG+1.002d–0.0285MMd (where MM is maternal mass). Milk-mass intake was also estimated in an additional 21 pups, using the isotopic dilution method. These values were then compared with estimates based on the milk mass-transfer equation for the same individual pups. A pair-wise comparison indicated that milk-mass transfer estimated using tritium dilution methods were significantly lower than those based on mass-transfer (MMLM). Furthermore, the absolute PMG exceeded tritium dilution estimates of milk-mass transfer in 35% of cases. In contrast, all milk-mass transfer estimates using the mass transfer method were greater than PMG. Overestimation of metabolic water production (MWP), leading to a smaller proportion of the total water intake being attributed to milk ingestion, is believed to be the most likely cause for significant underestimation of milk-mass transfer using the tritium dilution method. Consumption of exogenous water by pups is the most likely reason for the overestimation of MWP, although errors in estimated milk water content may have also contributed to underestimates. We conclude that, in our study, the mass-transfer method provided a more reliable estimate of milk-mass transfer than the isotopic dilution method; and we argue that, under certain conditions, it provides a practical alternative method where the assumptions of isotopic dilution methodology (e.g., all exogenous water from maternal milk) and quantitative parameters (e.g., maternal milk water content) may either be violated or impractical to measure.  相似文献

Vertebrates exhibit varied behavioural and physiological tactics to promote reproductive success. We examined mechanisms that could enable female loggerhead turtles to undertake nesting activities and maintain seasonal reproduction despite recent shark injuries of varying severity. We proposed that endocrinal mechanisms that regulate both a turtle's stress response and reproductive ability are modified to promote successful and continued reproduction. Irrespective of the degree of injury, females did not exhibit increased levels of the stress hormone corticosterone, nor decreased levels of the reproductive steroid testosterone; hormone responses consistent with stress. When exposed to a capture stressor, females with shark injury did not exhibit any greater corticosterone response than controls. In addition, breeding females showed a reduced corticosterone stress response compared to non-breeding females. Reduced endocrinal responses following shark injury, and during breeding in general may, in part, enable females to maintain behavioural and physiological commitment to reproduction.  相似文献

The first reaction of mitochondrial beta-oxidation, which is catalyzed by acyl-CoA dehydrogenases, was studied with unsaturated fatty acids that have a double bond either at the 4,5 or 5,6 position. The CoA thioesters of docosahexaenoic acid, arachidonic acid, 4,7,10-cis-hexadecatrienoic acid, 5-cis-tetradecenoic acid, and 4-cis-decenoic acid were effectively dehydrogenated by both rat and human long-chain acyl-CoA dehydrogenases (LCAD), whereas they were poor substrates of very long-chain acyl-CoA dehydrogenases (VLCAD). VLCAD, however, was active with CoA derivatives of long-chain saturated fatty acids or unsaturated fatty acids that have double bonds further removed from the thioester function. Although bovine LCAD effectively dehydrogenated 5-cis-tetradecenoyl-CoA (14:1) and 4,7,10-cis-hexadecatrienoyl-CoA, it was nearly inactive toward the other unsaturated substrates. The catalytic efficiency of rat VLCAD with 14:1 as substrate was only 4% of the efficiency determined with tetradecanoyl-CoA, whereas LCAD acted equally well on both substrates. The conclusion of this study is that LCAD serves an important, if not essential function in the beta-oxidation of unsaturated fatty acids.  相似文献

The composition and yield of oil in 615 trees representing the natural populations of Melaleuca alternifolia, or tea tree, was investigated. A sixth distinct oil chemotype was identified. Of the six chemotypes, one chemotype is dominated by terpinen-4-ol, one by 1,8-cineole, one by terpinolene and the remaining three chemotypes are all dominated by 1,8-cineole and differ in either terpinen-4-ol or terpinolene content. Whilst most chemotypes are present throughout the distribution range, a definite correspondence of oil types with geographic location was found. Terpinen-4-ol types predominate in and around the Bungawalbin basin in the Casino area of northern New South Wales (NSW), high 1,8-cineole types predominate toward the southern end of the distribution around Grafton and terpinolene types predominate in southern Queensland. Preliminary formulae have been developed to allow comparisons of oil data obtained by steam distillation with a static headspace gas chromatography method.  相似文献

This study examined the effect of melatonin (MLT) on in vitro phagocytosis of testicular macrophages taken from control and streptozotocin (STZ)-induced diabetic rats and the possible mechanism of its action. The phagocytic activity was measured as a number of latex beads ingested by 100 macrophages (PI, phagocytic index) in consecutive time points of the incubation. Changes in intracellular free calcium level [Ca2+]i in isolated macrophages in vitro were measured with the use of ratio-image fluorescence microscopy (fluorescent dye: Fura2/AM). Phagocytic index in macrophages isolated from healthy rats was 20% higher than in those from diabetic animals. Melatonin in physiological concentration (10−7 M) significantly (p < 0.05) increased the PI in testicular macrophages from control animals (PI = 68 ± 5 with MLT compared to PI = 46 ± 7 without MLT) while no such effect was observed in the cells from diabetic rats (PI = 36 ± 23 with MLT compared to PI = 31 ± 11 without MLT). Basal [Ca2+]i was significantly (p < 0.01) higher in macrophages from diabetic rats compared to control. Stimulation of both control and diabetic testicular macrophages with 10−7 M MLT resulted in a significant (p < 0.05) increase in [Ca2+]i in cells incubated in 2.5 mM calcium solution while no such response was observed in calcium-free Tyrode solution. However, MLT evoked [Ca2+]i response in macrophages isolated from diabetic animals was much lower than in macrophages isolated from age-matched controls and the time needed for maximal response was much longer. Lack of response in calcium-free solution suggests that extracellular calcium may be necessary to trigger MLT response and in its progression.  相似文献
